    • Cytoskeletal organization was analysed in the vestibular ganglion cells and in the sensory epithelia of all five vestibular organs of the adult human temporal bone. A pancellular rigidity of the upper surfaces of the vestibular organs, evidenced by large quantities of intermediate filaments and actin, seems of importance for the mechanoelectrical transduction by opposing the motion of the cupulae and statoconial layers. Immunostaining for the calcium-binding protein synaptophysin in the sensory hair bundles and in the area close to the cuticular plates--the first of its kind to be demonstrated in human vestibular organs--indicates the presence of calcium-dependent ionic channels. The hypothesis is presented that the calyx might be involved in a short-loop feed back control of type I hair cells, i.e. of the mechanoelectrical transduction itself. Subpopulations of vestibular ganglion cells were identified by the staining pattern of cytoskeletal proteins, but not by ordinary ultrastructural analysis.

  • Validation Of Delayed Gadolinium-Enhanced Magnetic Resonance Imaging Of Cartilage And T2 Mapping For Quantifying Distal Metacarpus/Metatarsus Cartilage Thickness In Thoroughbred Racehorses

    • The purpose of this study was to determine whether delayed gadolinium-enhanced magnetic resonance imaging of cartilage (dGEMRIC) and T2 mapping are accurate techniques for measuring cartilage thickness in the metacarpus3/metatarsus3 (Mc3/Mt3) of Thoroughbred racehorses. Twenty-four Mc3/Mt3 cadaver specimens were acquired from six healthy racehorses. Cartilage thickness was measured from postintra-articular Gd-DTPA2 images acquired using short tau inversion recovery (STIR), and proton density weighted (PDw) sequences, and compared with cartilage thickness measured from corresponding histologic images. Two observers performed each histologic measurement twice at three different sites, with measurement times spaced at least 5 days apart. Histologic cartilage thickness was measured at each of the three sites from the articular surface to the bonecartilage interface, and from the articular surface to the mineralized cartilage interface (tidemark). Intra-observer repeatability was good to moderate for dGEMRIC where Mc3/Mt3 cartilage was not in contact with the proximal phalanx. Where the Mc3/Mt3 cartilage was in contact with the proximal phalanx cartilage, dGEMRIC STIR and T2 mapping PDw cartilage thicknesses of Mc3/Mt3 could not be measured reliably. When measured from the articular surface to the bonecartilage interface, histologic cartilage thickness did not differ from STIR or PDw cartilage thickness at the site where the Mc3/Mt3 cartilage surface was separated from the proximal phalanx cartilage (P > 0.05). Findings indicated that dGEMRIC STIR and T2 mapping PDw are accurate techniques for measuring Mc3/Mt3 cartilage thickness at locations where the cartilage is not in direct contact with the proximal phalanx cartilage.

    • Uropathogenic Escherichia coli elicit a host response that determines the severity of urinary tract infection (UTI). Specific adherence mechanisms allow the bacteria to initiate this process by targeting epithelial cells in the urinary tract mucosa. Epidemiological studies show a strong association of P-fimbriae with disease severity, suggesting that adherence mediated by these organelles has a direct effect on mucosal inflammation in vivo. The present study examined the ability of P-fimbriae to induce inflammation in the human urinary tract. Patients were subjected to intravesical inoculation with a non-fimbriated E. coli strain or transformants of this strain expressing P-fimbriae. The inflammatory response was analysed as a function of P-fimbrial expression. The P-fimbriated transformants invariably caused higher interleukin (IL)-8, IL-6 and neutrophil responses in the urinary tract than the ABU strain. Furthermore, loss of P-fimbrial expression in vivo was accompanied by a return to background levels of neutrophils, IL-6 and IL-8 in individual patients. The results demonstrate that the pap sequences confer on a non-fimbriated, avirulent strain the ability to induce a host response in the human urinary tract. P-fimbriae thus fulfil the 'molecular Koch-Henle postulates' linking a single virulence factor to host response induction.

  • Making room for sleep : A classroom based prevention program for adolescents

    • Sleep patterns go through important changes during adolescence and sleep deprivation is extremely common, with severe consequences for adolescents´ daily functioning (Owens et al., 2014).Previous school-based interventions attempting to prevent sleep problems and their consequences have shown an increase in adolescents´ sleep knowledge but no behavioral changes. These programs usually include sleep education, cognitive and behavioral strategies, and a motivational framework (Cassoff, Knäuper, Michaelsen, & Gruber, 2013). Although the importance of motivation and engagement is unquestionable, previous attempts at enhancing adolescents´ motivation to change sleep behaviors might have been too shallow. We developed a new program that specifically targets barriers adolescents may encounter when trying to change their sleep behaviors, such as use of information and communication technology (ICT) late in the evening, stress due to schoolwork, and extracurricular activities. So, the intervention did not only focus on sleep education but also on improving students’ time management skills and encouraging them to monitor their ICT use. We also developed an interactive app for the intervention to increase engagement and to take advantage of the opportunities of ICT.The aim of this study was to evaluate the effectiveness of this classroom delivered universal sleep intervention. More specifically, we wanted to investigate whether changes in technology usage and perceived stress preceded changes in adolescents´ sleep duration. The intervention consisted of 5x45 min sessions over 6-7 weeks. Students (N = 292) completed weekly questionnaires through the app including a sleep diary for school-week and weekend sleep (bedtime, wake time, sleep onset latency, and wake after sleep onset), mobile usage, and motivation to change. They also completed pre- and post-intervention assessments, including self-reported sleep hygiene, perceived stress, and mood. At the end of the intervention students provided a qualitative evaluation of the sleep program.The intervention is still ongoing and will be completed by the end of the school year (June 2016). So far, the intervention has shown good feasibility in the school context. Preliminary results will be presented on possible changes in sleep duration and sleep hygiene and the role of ICT use and stress.This study will give us important information on adolescents´ sleep and its barriers in our 24/7 society. The discussion will focus on the possibilities of using this kind of preventive school-based interventions, as well as how modern technology (i.e. an interactive app) may be used for promoting engagement in this age group.

  • Ethnic Harassment and Immigrant Youth's Engagement in Violent Behaviors : Understanding the Risk Factors

    • The present study aimed to examine whether ethnic harassment was related to violent behaviors among immigrant youth over time and to identify the risk factors. The sample comprised immigrant adolescents living in Sweden (N = 365; Mage  = 13.93, SD = 0.80). Results showed that the more youth were ethnically harassed, the more they engaged in violent acts over time. A separated identity significantly moderated the effect of ethnic harassment on youth's engagement in violent behaviors. Specifically, ethnic harassment positively predicted engagement in violent behaviors only at high levels of separated identity. Impulsivity and school ethnic composition did not act as moderators. The findings suggest that preventing violent behaviors among immigrant youth requires a focus on promoting positive interethnic relationships, and multicultural identity among immigrant youth.

  • lmprovement of 177Lu-octreotate treatment of small-intestine neuroendocrine tumors by hyperfractionation

    • Neuroendocrine tumor incidence is steadily rising, and the late diagnosis often results in metastatic disease and current treatments mostly prolong life and in-crease quality of life without increasing cure rate. 177Lu-octreotate was recently approved for treatment of neuroendocrine tumors, but the dosage scheme should be optimized. The aim of this thesis was to study the effects of fractionated admin-istration of 177Lu-octreotate in the human GOT1 tumor mouse model. GOT1 bearing mice were given 177Lu-octreotate as a single or fractionated admin-istration. The biodistribution and tumor volume response was followed with time. DNA and RNA were extracted from tumor tissue. DNA methylation was evaluated, and expression of genes involved in apoptosis determined. Hyperfractionated administration gave a more pronounced anti-tumoral effect and longer progression-free survival than single administration with the same total amount of 177Lu-octreotate. The methylation analysis of genes and promoters re-vealed 177Lu-octreotate treatment specific responses across the groups. Altered expression of apoptosis related genes in regrown tumors was modest, with vary-ing commonalities between the groups. Hyperfractionation generally resulted in a different apoptotic gene expression pattern compared with single-administration in regrown tumors. Hyperfractionation led to higher absorbed dose to tumor and lower to kidneys than single-administration. Expression of genes related to apop-tosis in tumors were similar between groups early after high dose level of 177Lu-octreotate, but sometimes with a trend towards higher gene regulation for the hy-perfractionated groups. The pro apoptotic genes BAX, FAS, GADD45A and TNFRSF10B were significantly regulated at several early time-points for both high dose groups. In conclusion, hyperfractionation of 177Lu-octreotate shows promise compared with single administration, and should be tested clinically.
