  • The development of a marine natural product-based antifouling paint

    • Problems with tin and copper antifouling compounds have highlighted the need to develop new environmentally friendly antifouling coatings. Bacteria isolated from living surfaces in the marine environment are a promising source of natural antifouling compounds. Four isolates were used to produce extracts that were formulated into ten water-based paints. All but one of the paints showed activity against a test panel of fouling bacteria. Five of the paints were further tested for their ability to inhibit the settlement of barnacle larvae, Balanus amphitrite, and algal spores of Ulva lactuca, and for their ability to inhibit the growth of U. lactuca. Two paints caused a significant decrease in the number of settled barnacles. One paint containing extract of Pseudomonas sp. strain NUDMB50-11, showed excellent activity in all assays. The antifouling chemicals responsible for the activity of the extract were isolated, using bioassay guided fractionation, and their chemical structures determined.

  • Kinase-dependent and kinase-independent functions of EphA4 receptors in major axon tract formation in vivo

    • The EphA4 receptor tyrosine kinase regulates the formation of the corticospinal tract (CST), a pathway controlling voluntary movements, and of the anterior commissure (AC), connecting the neocortical temporal lobes. To study EphA4 kinase signaling in these processes, we generated mice expressing mutant EphA4 receptors either lacking kinase activity or with severely downregulated kinase activity. We demonstrate that EphA4 is required for CST formation as a receptor for which it requires an active kinase domain. In contrast, the formation of the AC is rescued by kinase-dead EphA4, suggesting that in this structure EphA4 acts as a ligand for which its kinase activity is not required. Unexpectedly, the cytoplasmic sterile-alpha motif (SAM) domain is not required for EphA4 functions. Our findings establish both kinase-dependent and kinase-independent functions of EphA4 in the formation of major axon tracts.

    • The Emu-Myc transgenic mouse appears to be an accurate model of human Burkitt's lymphoma that bears MYC/Immunoglobulin gene translocations. Id2, a negative regulator of basic helix-loop-helix transcription factors, has also been proposed as a Myc target gene that drives the proliferative response of Myc by binding to and overriding the checkpoint functions of the retinoblastoma tumor suppressor protein. Targeted deletion of Id2 in mice results in defects in B-cell development and prevents the development of peripheral lymphoid nodes. In precancerous B cells and lymphomas that arise in Emu-Myc transgenic mice and in Burkitt's lymphomas, Id2 is overexpressed, suggesting that it plays a regulatory role in lymphoma development. Surprisingly, despite these connections, Emu-Myc mice lacking Id2 succumb to lethal B-cell lymphoma at rates comparable with wild-type Emu-Myc transgenics. Furthermore, precancerous splenic B cells lacking Id2 do not exhibit any significant defects in Myc-induced target gene transactivation and proliferation. However, due to their lack of secondary lymph nodes, Emu-Myc mice lacking Id2 rather succumb to disseminated lymphoma with an associated leukemia, with pronounced infiltrates of the bone marrow and other major organs. Collectively these findings argue that targeting Id2 functions may be ineffective in preventing Myc-associated malignancies.

    • The dielectric relaxation was determined in a time-domain spectrometer between 100 and 295 K and from 0.1 to 1000 Hz for series of dendritic liquid crystalline polymers with scaffolds based on 2,2-bis(hydroxymethyl)propionic acid (bis-MPA) of generations 0-3 with 4-((R)-1-methylheptyloxy)phenyl 4-{4'-[10-(hydroxy-carbonyl)decyloxy]-phenyl} benzoate as mesogen and for the second generation of dendrimers with 4-[10-(hydroxycarbonyl)decyloxy]-biphenyl 4-[4'-(2-(R)-octyloxy)-3-nitrophenyl] benzoate as mesogen. Dielectric data are also reported for a side-chain liquid crystalline polymer based on {4''-[(R)-(-)-2-octyloxyl-3-nitro-phenyl 4-(4'-(11-acryloyl-oxyundecyloxy)phenyl}benzoate. The low temperature subglass process (gamma) assigned to the spacer group was essentially insensitive to molecular architecture and to the presence of a pendant nitro group in the mesogen. The high temperature sub-glass process (beta) assigned to a 180 degrees flip of part of mesogen was only moderately sensitive to the molecular architecture. The introduction of a pendant nitro group in the mesogen led to a substantial increase in both activation energy and relaxation strength. The results suggest that the phenylene ring with the pendant nitro group participates in the beta process.
