Robust estimation of bacterial cell count from optical density

- Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

Public toilets for women : how female municipal councillors expanded the right to the city in Sweden, c. 1910–1925

- This article shows that women’s formal political citizenship is intertwined with the right to the city. Using Carol Bacchi’s ‘What’s the Problem Represented to be’ approach, we study how female municipal councillors argued for more public toilets in Sweden's three largest cities. In their motions, the lack of public conveniences was mainly related to a binary discourse of gender, and arguments referring to biological needs of the female body and women as mothers proved more successful than the argument that uneven access to public toilets was an example of gender inequality. Class-based arguments against fees were largely ignored when the motions were transformed from potentially contentious political issues into technical issues of urban planning and municipal utilities. However, the material effects of the policies—public toilets placed mainly in working-class areas—led to an extension of possible radiuses of movement within the city for women, and perhaps especially working-class women.

- Påskhelgens upplopp har mötts med ett kompakt avståndstagande från det svenska politiska etablissemanget. Enligt justitieminister Morgan Johansson finns det bara ett sätt att möta upplopp, ”att sätta hårt mot hårt”. Denna tidnings ledarsida konstaterar närmast programmatiskt oförstående att “det finns inga förklaringar, inga undanflykter som håller” när den beskriver vad som skett. Ändå är det just förståelse av sociala fenomen som upplopp vi behöver om vi inte vill se liknande scener i framtiden.

- In comparison to disciplines within the humanities and social sciences working directly with living humans, as well as in comparison to oral history, historians primarily working with archival sources lack a living discussion on ethics in connection with our methods. At the same time, archival sources that contain sensitive personal information are central in many fields – such as queer history – and the Swedish Ethical Review Authority has recently started to examine academic compliance with the Law on Ethical Review.In relation to this and the need to find an ethical approach to my own sources, this essay is an attempt to stimulate a discussion on how historians can work with sensitive archival sources in an ethical manner. I do so by deliberating on three ethical positions – process ethics, ethics of care and duty-based ethics – and an attempt to reason regarding their respective consequences and benefits in terms of working with archival sources. The essay ends with an argument in favour of duty-based ethics and critique as a way to construct ourselves as ethical subjects in relation to our duty as historians.
