  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Effect of Enterohepatic Circulation on the Accumulation of Per- and Polyfluoroalkyl Substances : Evidence from Experimental and Computational Studies

    • The pharmacokinetic characteristics of per- and polyfluoroalkyl substances (PFAS) affect their distribution and bioaccumulation in biological systems. The enterohepatic circulation leads to reabsorption of certain chemicals from bile back into blood and the liver and thus influences their elimination, yet its influence on PFAS bioaccumulation remains unclear. We explored the role of enterohepatic circulation in PFAS bioaccumulation by examining tissue distribution of various PFAS in wild fish and a rat model. Computational models were used to determine the reabsorbed fractions of PFAS by calculating binding affinities of PFAS for key transporter proteins of enterohepatic circulation. The results indicated that higher concentrations were observed in blood, the liver, and bile compared to other tissues for some PFAS in fish. Furthermore, exposure to a PFAS mixture on the rat model showed that the reabsorption phenomenon appeared during 8-12 h for most long-chain PFAS. Molecular docking calculations suggest that PFAS can bind to key transporter proteins via electrostatic and hydrophobic interactions. Further regression analysis adds support to the hypothesis that binding affinity of the apical sodium-dependent bile acid transporter is the most important variable to predict the human half-lives of PFAS. This study demonstrated the critical role of enterohepatic circulation in reabsorption, distribution, and accumulation of PFAS.

  • From Handcrafted to Deep Features for Pedestrian Detection : A Survey

    • Pedestrian detection is an important but challenging problem in computer vision, especially in human-centric tasks. Over the past decade, significant improvement has been witnessed with the help of handcrafted features and deep features. Here we present a comprehensive survey on recent advances in pedestrian detection. First, we provide a detailed review of single-spectral pedestrian detection that includes handcrafted features based methods and deep features based approaches. For handcrafted features based methods, we present an extensive review of approaches and find that handcrafted features with large freedom degrees in shape and space have better performance. In the case of deep features based approaches, we split them into pure CNN based methods and those employing both handcrafted and CNN based features. We give the statistical analysis and tendency of these methods, where feature enhanced, part-aware, and post-processing methods have attracted main attention. In addition to single-spectral pedestrian detection, we also review multi-spectral pedestrian detection, which provides more robust features for illumination variance. Furthermore, we introduce some related datasets and evaluation metrics, and a deep experimental analysis. We conclude this survey by emphasizing open problems that need to be addressed and highlighting various future directions.   • SipMaskv2: Enhanced Fast Image and Video Instance Segmentation

    • We propose a fast single-stage method for both image and video instance segmentation, called SipMask, that preserves the instance spatial information by performing multiple sub-region mask predictions. The main module in our method is a light-weight spatial preservation (SP) module that generates a separate set of spatial coefficients for the sub-regions within a bounding-box, enabling a better delineation of spatially adjacent instances. To better correlate mask prediction with object detection, we further propose a mask alignment weighting loss and a feature alignment scheme. In addition, we identify two issues that impede the performance of single-stage instance segmentation and introduce two modules, including a sample selection scheme and an instance refinement module, to address these two issues. Experiments are performed on both image instance segmentation dataset MS COCO and video instance segmentation dataset YouTube-VIS. On MS COCO test-dev set, our method achieves a state-of-the-art performance. In terms of real-time capabilities, it outperforms YOLACT by a gain of 3.0% (mask AP) under the similar settings, while operating at a comparable speed. On YouTube-VIS validation set, our method also achieves promising results. The source code is available at https://github.com/JialeCao001/SipMask.

  • Vulnerability of blue foods to human-induced environmental change

    • Global aquatic foods are a key source of nutrition, but how their production is influenced by anthropogenic environmental changes is not well known. The vulnerability of global blue food systems to main environmental stressors and the related spatial impacts across blue food nations are now quantified. Global aquatic or 'blue' foods, essential to over 3.2 billion people, face challenges of maintaining supply in a changing environment while adhering to safety and sustainability standards. Despite the growing concerns over their environmental impacts, limited attention has been paid to how blue food production is influenced by anthropogenic environmental changes. Here we assess the vulnerability of global blue food systems to predominant environmental disturbances and predict the spatial impacts. Over 90% of global blue food production faces substantial risks from environmental change, with the major producers in Asia and the United States facing the greatest threats. Capture fisheries generally demonstrate higher vulnerability than aquaculture in marine environments, while the opposite is true in freshwater environments. While threats to production quantity are widespread across marine and inland systems, food safety risks are concentrated within a few countries. Identifying and supporting mitigation and adaptation measures in response to environmental stressors is particularly important in developing countries in Asia, Latin America and Africa where risks are high and national response capacities are low. These findings lay groundwork for future work to map environmental threats and opportunities, aiding strategic planning and policy development for resilient and sustainable blue food production under changing conditions.

  • China at a Crossroads : An Analysis of China's Changing Seafood Production and Consumption

    • China is a key player in global production, consumption, and trade of seafood. Given this dominance, Chinese choices regarding what seafood to eat, and how and where to source it, are increasingly important—for China, and for the rest of the world. This perspective explores this issue using a transdisciplinary approach and discusses plausible trajectories and implications for assumptions of future modeling efforts and global environmental sustainability and seafood supply. We outline China's 2030 projected domestic seafood production and consumption through an examination of available statistics, and qualitatively evaluate these in relation to key stated Chinese policy targets, consumer trends, and dominant political narratives. Our analysis shows that by 2030 China is likely to see seafood consumption outstrip domestic production. To meet the seafood gap China will likely attempt to increase domestic freshwater and offshore aquaculture, increase seafood imports, possibly expand the distant water fishing industry, and invest in seafood production abroad.

  • Four ways blue foods can help achieve food system ambitions across nations

    • Blue foods, sourced in aquatic environments, are important for the economies, livelihoods, nutritional security and cultures of people in many nations. They are often nutrient rich1, generate lower emissions and impacts on land and water than many terrestrial meats2, and contribute to the health3, wellbeing and livelihoods of many rural communities4. The Blue Food Assessment recently evaluated nutritional, environmental, economic and justice dimensions of blue foods globally. Here we integrate these findings and translate them into four policy objectives to help realize the contributions that blue foods can make to national food systems around the world: ensuring supplies of critical nutrients, providing healthy alternatives to terrestrial meat, reducing dietary environmental footprints and safeguarding blue food contributions to nutrition, just economies and livelihoods under a changing climate. To account for how context-specific environmental, socio-economic and cultural aspects affect this contribution, we assess the relevance of each policy objective for individual countries, and examine associated co-benefits and trade-offs at national and international scales. We find that in many African and South American nations, facilitating consumption of culturally relevant blue food, especially among nutritionally vulnerable population segments, could address vitamin B12 and omega-3 deficiencies. Meanwhile, in many global North nations, cardiovascular disease rates and large greenhouse gas footprints from ruminant meat intake could be lowered through moderate consumption of seafood with low environmental impact. The analytical framework we provide also identifies countries with high future risk, for whom climate adaptation of blue food systems will be particularly important. Overall the framework helps decision makers to assess the blue food policy objectives most relevant to their geographies, and to compare and contrast the benefits and trade-offs associated with pursuing these objectives.

  • Biomass valorization toward sustainable asphalt pavements : Progress and prospects

    • To cope with the global climate crisis and assist in achieving the carbon neutrality, the use of biomass materials to fully or partially replace petroleum-based products and unrenewable resources is expected to become a widespread solution. Based on the analysis of the existing literature, this paper firstly classified biomass materials with potential application prospects in pavement engineering according to their application and summarized their respective preparation methods and characteristics. The pavement performance of asphalt mixtures with biomass materials was analyzed and summarized, and the economic and environmental benefits of bio-asphalt binder were evaluated. The analysis shows that pavement biomass materials with potential for practical application can be divided into three categories: bio-oil, bio-fiber, and bio-filler. Adding bio-oil to modify or extend the virgin asphalt binder can mostly improve the low temperature performance of asphalt binder. Adding styrene-butadienestyrene (SBS) or other preferable bio-components for composite modification will have a further improved effect. Most of the asphalt mixtures prepared by using bio-oil modified asphalt binders have improved the low temperature crack resistance and fatigue resistance of asphalt mixtures, but the high temperature stability and moisture resistance may decrease. As a rejuvenator, most bio-oils can restore the high and low temperature performance of aged asphalt and recycled asphalt mixture, and improve fatigue resistance. Adding bio-fiber could significantly improve the high temperature stability, low temperature crack resistance and moisture resistance of asphalt mixtures. Biochar as a bio-filler can slow down the asphalt aging process and some other bio-fillers can improve the high temperature stability and fatigue resistance of asphalt binders. Through calculation, it is found that the cost performance of bio-asphalt has the ability to surpass conventional asphalt and has economic benefits. The use of biomass materials for pavements not only reduces pollutants, but also reduces the dependence on petroleum-based products. It has significant environmental benefits and development potential.
