  • Performance Assessment of Wastewater Treatment Plants : Multi-Objective Analysis Using Plant-Wide Models

    • As the knowledge about anthropogenic impacts of climate change has grown, the awareness of the contributions from treatment of wastewater has widened the scope for wastewater treatment plants (WWTPs). Not only shall ever stricter effluent constraints be met, but also energy efficiency be increased, greenhouse gases mitigated and resources recovered. All under a constant pressure on costs. The main objective of this research has been to develop a plant-wide modelling tool to evaluate the performance of operational strategies for multiple objectives at the plant and for off-site environmental impact. The plant-wide model platform Benchmark Simulation Model no. 2 (BSM2) has been modified to improve the evaluation of energy efficiency and include greenhouse gas emissions. Furthermore, the plant-wide process model has been coupled to a life cycle analysis (LCA) model for evaluation of global environmental impact. For energy evaluation, a dynamic aeration system model has been adapted and implemented. The aeration model includes oxygen transfer efficiency, dynamic pressure in the distribution system and non-linear behaviour of blower performance. To allow for modelling of energy recovery via anaerobic co-digestion the digestion model of BSM2 was updated with a flexible co-digestion model allowing for dynamic co-substrate feeds. A feasible procedure for substrate characterisation was proposed. Emissions of the greenhouse gases CO2, CH4 and N2O were considered. The bioprocess model in BSM2 was updated with two-step nitrification, four-step denitrification and nitrifier denitrification to capture N2O production. Fugitive emissions of the three gases were included from digestion, cogeneration and sludge storage. The models were tested in case studies for the three areas of development: aeration, co-digestion and greenhouse gas production. They failed to reject the hypothesis that dynamic process models are required to assess the highly variable operations of wastewater treatment plants. All parts were combined in a case study of the Käppala WWTP in Lidingö, Sweden, for comparison of operational strategies and evaluation of stricter effluent constraints. The averaged model outputs were exported to an LCA model to include off-site production of input goods and impact of discharged residues and wastes. The results reveal trade-offs between water quality, energy efficiency, greenhouse gas emissions and abiotic depletion of elemental and fossil resources. The developed tool is generally applicable for WWTPs and the simulation results from this type of combined models create a good basis for decision support.

  • Differential conformational modulations of MreB folding upon interactions with GroEL/ES and TRiC chaperonin components

    • Here, we study and compare the mechanisms of action of the GroEL/GroES and the TRiC chaperonin systems on MreB client protein variants extracted from E. coli. MreB is a homologue to actin in prokaryotes. Single-molecule fluorescence correlation spectroscopy (FCS) and time-resolved fluorescence polarization anisotropy report the binding interaction of folding MreB with GroEL, GroES and TRiC. Fluorescence resonance energy transfer (FRET) measurements on MreB variants quantified molecular distance changes occurring during conformational rearrangements within folding MreB bound to chaperonins. We observed that the MreB structure is rearranged by a binding-induced expansion mechanism in TRiC, GroEL and GroES. These results are quantitatively comparable to the structural rearrangements found during the interaction of beta-actin with GroEL and TRiC, indicating that the mechanism of chaperonins is conserved during evolution. The chaperonin-bound MreB is also significantly compacted after addition of AMP-PNP for both the GroEL/ES and TRiC systems. Most importantly, our results showed that GroES may act as an unfoldase by inducing a dramatic initial expansion of MreB (even more than for GroEL) implicating a role for MreB folding, allowing us to suggest a delivery mechanism for GroES to GroEL in prokaryotes.

  • A Novel Security-Enhanced Agile Software Development Process Applied in an Industrial Setting

    • A security-enhanced agile software development process, SEAP, is introduced in the development of a mobile money transfer system at Ericsson Corp. A specific characteristic of SEAP is that it includes a security group consisting of four different competences, i.e., Security manager, security architect, security master and penetration tester. Another significant feature of SEAP is an integrated risk analysis process. In analyzing risks in the development of the mobile money transfer system, a general finding was that SEAP either solves risks that were previously postponed or solves a larger proportion of the risks in a timely manner. The previous software development process, i.e., The baseline process of the comparison outlined in this paper, required 2.7 employee hours spent for every risk identified in the analysis process compared to, on the average, 1.5 hours for the SEAP. The baseline development process left 50% of the risks unattended in the software version being developed, while SEAP reduced that figure to 22%. Furthermore, SEAP increased the proportion of risks that were corrected from 12.5% to 67.1%, i.e., More than a five times increment. This is important, since an early correction may avoid severe attacks in the future. The security competence in SEAP accounts for 5% of the personnel cost in the mobile money transfer system project. As a comparison, the corresponding figure, i.e., For security, was 1% in the previous development process.

  • Introducing a Novel Security-Enhanced Agile Software Development Process

    • In this paper, a novel security-enhanced agile software development process, SEAP, is introduced. It has been designed, tested, and implemented at Ericsson AB, specifically in the development of a mobile money transfer system. Two important features of SEAP are 1) that it includes additional security competences, and 2) that it includes the continuous conduction of an integrated risk analysis for identifying potential threats. As a general finding of implementing SEAP in software development, the developers solve a large proportion of the risks in a timely, yet cost-efficient manner. The default agile software development process at Ericsson AB, i.e. where SEAP was not included, required significantly more employee hours spent for every risk identified compared to when integrating SEAP. The default development process left 50.0% of the risks unattended in the software version that was released, while the application of SEAP reduced that figure to 22.5%. Furthermore, SEAP increased the proportion of risks that were corrected from 12.5% to 67.9%, a more than five times increment.

  • Simplified citrate anticoagulation for CRRT without calcium replacement.

    • Since 2012 citrate anticoagulation is the recommended anticoagulation strategy for CRRT. The main drawback using citrate compared with heparin as anticoagulant is the need for calcium replacement and the rigorous control of calcium levels. This study investigated the possibility to achieve anticoagulation while eliminating the need for calcium replacement. This was successfully achieved by including citrate and calcium in all CRRT solutions. Thereby the total calcium concentration was kept constant throughout the extracorporeal circuit while the ionized calcium was kept at levels low enough to avoid clotting. Being a completely new concept, only five patients with acute renal failure were included in a short, prospective, intensely supervised non-randomized pilot study.Systemic electrolyte levels and acid-base parameters were stable and remained within physiological levels. Ionized calcium levels declined slightly initially, but stabilized at 1.1 mmol/l. Plasma citrate concentrations stabilized at around 0.6 mmol/l. All post-filter ionized calcium levels were <0.5 mmol/l, i.e. an anticoagulation effect was reached. All filter pressures were normal indicating no clotting problems, and no visible clotting was observed. No calcium replacement was needed.This pilot study suggests that it is possible to perform regional citrate anticoagulation without the need for separate calcium infusion during CRRT.

  • Reported opening limitations as a TMD symptom: A clinical report on diagnoses and outcome

    • Objective: To study diagnoses of reported mouth opening limitations and the outcome after treatment, to better understand the symptom and the prognosis. Methods: New referrals to an Orofacial Pain & Temporomandibular Disorders (TMD) Clinic with the symptom of “jaw locking” in the referral were considered for this clinical prospective report. This referrals constituted 5.8 % of all the referrals during a ten month period, and 40 patients were included. Case history, clinical examination and diagnoses were made according to the RDC/TMD criteria. MRI and CT examinations of the temporomandibular joint were performed when indicated. Reported symptoms and clinical signs were compared for different diagnoses. The treatment was non-invasive and conservative, no patient underwent surgery. At end of treatment, the clinical examination was repeated, the primary and definite diagnoses were compared, and the improvement after treatment was evaluated. Results: The main diagnoses were disc displacement with reduction (DDwR), 23 %, disc displacement without reduction (DDwoR), 30 %, and myofascial pain with limited opening (wLO), 45 %. General hypermobility was significantly more frequent in the disc displacement diagnosis compared with myofascial pain (p < .05). After treatment, the mean opening capacity was good, regardless of the diagnosis. Conclusions: A report of jaw locking is not indicative of a single diagnosis of TMD. A diagnosis based on the history and a clinical examination is generally accurate. The symptom of opening limitations responds well to conservative treatment methods.

  • Analyses of activated sludge processes consisting of a plug-flow reactor and a non-ideal settler

    • An activated sludge process (ASP) consisting of a plug-flow reactor (PFR) and a non-ideal settler is modelled and analysed. One soluble substrate component and one particulate biomass are assumed. The biomass growth rate is described by a Monod function. The settler model includes hindered settling and compression. A model describing the steady-state behaviour of the ASP is derived which constrains the settler to work with a fixed sludge blanket height in the thickening zone. The model provides new understanding for these types of ASPs and may be used for novel design schemes. The numerical example suggests that the steady-state solutions of the ASP give a one-parameter family ofsolutions, where the parameter is the recycle ratio 푟.
