  • Kostmann disease and other forms of severe congenital neutropenia
  • 2021
  • Ingår i: Acta Paediatrica. - : John Wiley & Sons. - 0803-5253 .- 1651-2227. ; 110:11, s. 2912-2920
  • Tidskriftsartikel (refereegranskat)abstract
    • Congenital neutropenia with autosomal recessive inheritance was first described by the Swedish paediatrician Rolf Kostmann who coined the term ‘infantile genetic agranulocytosis’. The condition is now commonly referred to as Kostmann disease. These patients display a maturation arrest of the myelopoiesis in the bone marrow and reduced neutrophil numbers and suffer from recurrent, often life-threatening infections. The molecular mechanism underlying congenital neutropenia has been intensively investigated, and mutations in genes that impinge on programmed cell death have been identified. The present review provides an overview of these studies.

  • Influenza-A mediated pre-existing immunity levels to SARS-CoV-2 could predict early COVID-19 outbreak dynamics
  • 2023
  • Ingår i: iScience. - : CELL PRESS. - 2589-0042. ; 26:12
  • Tidskriftsartikel (refereegranskat)abstract
    • Susceptibility to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections is highly variable and could be mediated by a cross-protective pre-immunity. We identified 14 cross-reactive peptides between SARS-CoV-2 and influenza A H1N1, H3N2, and human herpesvirus (HHV)-6A/B with potential relevance. The H1N1 peptide NGVEGF was identical to a peptide in the most critical receptor binding motif in SARS-CoV-2 spike protein that interacts with the angiotensin converting enzyme 2 receptor. About 62%-73% of COVID-19-negative blood donors in Stockholm had antibodies to this peptide in the early pre-vaccination phase of the pandemic. Seasonal flu vaccination enhanced neutralizing capacity to SARS-CoV-2 and T cell immunity to this peptide. Mathematical modeling taking the estimated pre -immunity levels to flu into account could fully predict pre-Omicron SARS-CoV-2 outbreaks in Stockholm and India. This cross-immunity provides mechanistic explanations to the epidemiological observation that influenza vaccination protected people against early SARS-CoV-2 infections and implies that flu-mediated cross-protective immunity significantly dampened the first SARS-CoV-2 outbreaks.

  • Combined L2-stable feedback and feedforward aeration control in a wastewater treatment plant
  • 2020
  • Ingår i: IEEE Transactions on Control Systems Technology. - : Institute of Electrical and Electronics Engineers (IEEE). - 1063-6536 .- 1558-0865. ; 28:3, s. 1017-1024
  • Tidskriftsartikel (refereegranskat)abstract
    • A nitrifying activated sludge process with ammonium feedback and inflow feedforward control is studied. The feedback and feedforward measurements are subject to time delay and the control loop includes a saturation. A Hammerstein-based model is therefore identified, including process delays and the nonlinearity. A Monod-type nonlinearity is used, as motivated by the oxygen injection efficiency reduction, with increasing dissolved oxygen concentration. The proposed feedback control strategy includes tuning of a linear lag compensator that has a limited low-frequency gain which allows global stability to be established with the Popov criterion. The feedforward controller attenuates the disturbance and provides an overall improvement of the control strategy. The performance of the combined feedback and feedforward aeration controller is evaluated with a benchmark model.

  • An Open-Source Finite Element Model of a Generic Car Seat: Development and Validation for Low-Severity Rear Impact Evaluations
  • 2022
  • Ingår i: Conference proceedings International Research Council on the Biomechanics of Injury, IRCOBI. - : IRCOBI. - 2235-3151. ; 2022-September, s. 229-242, s. 229-242
  • Konferensbidrag (refereegranskat)abstract
    • A Finite Element model of a generic Laboratory Seat was developed to replicate a physical counterpart used in rear-impact volunteer tests. The Laboratory Seat has a simplified design, developed to facilitate replication in computational models. The seat has a flat rigid base and the seatback consists of four horizontal panels attached to side posts by coil springs. The seat model was validated with results from component tests and sled tests, including the Anthropomorphic Test Device, BioRID II. An initial test series was carried out to generate data for component validation: the first set of tests to characterise the coil spring properties; and the second set comprising Impactor Tests on Head Restraint Foam to assess the head restraint material properties. For system level validation, sled tests were conducted both with the empty Laboratory Seat and with the BioRID II. The BioRID II tests were conducted in conjunction with an earlier volunteer test study. Both the component and the sled tests were reproduced in a virtual environment. Good agreement was achieved between the mechanical tests and the computational simulations. The seat model is freely available to use: https://openvt.eu/fem/open-access-laboratory-seat-model.

  • AKVANO (R) : A Novel Lipid Formulation System for Topical Drug Delivery-In Vitro Studies
  • 2022
  • Ingår i: Pharmaceutics. - : MDPI. - 1999-4923. ; 14:4
  • Tidskriftsartikel (refereegranskat)abstract
    • A novel formulation technology called AKVANO (R) has been developed with the aim to provide a tuneable and versatile drug delivery system for topical administration. The vehicle is based on a water-free lipid formulation where selected lipids, mainly phospholipids rich in phosphatidylcholine, are dissolved in a volatile solvent, such as ethanol. With the aim of describing the basic properties of the system, the following physicochemical methods were used: viscometry, dynamic light scattering, NMR diffusometry, and atomic force microscopy. AKVANO formulations are non-viscous, with virtually no or very minute aggregates formed, and when applied to the skin, e.g., by spraying, a thin film consisting of lipid bilayer structures is formed. Standardized in vitro microbiological and irritation tests show that AKVANO formulations meet criteria for antibacterial, antifungal, and antiviral activities and, at the same time, are being investigated as a non-irritant to the skin and eye. The ethanol content in AKVANO facilitates incorporation of many active pharmaceutical ingredients (>80 successfully tested) and the phospholipids seem to act as a solubilizer in the formulation. In vitro skin permeation experiments using Strat-M (R) membranes have shown that AKVANO formulations can be designed to alter the penetration of active ingredients by changing the lipid composition.
