  • "Vi klimatforskare stödjer Greta och skolungdomarna"
  • 2019
  • Ingår i: Dagens nyheter (DN debatt). - 1101-2447.
  • Tidskriftsartikel (populärvet., debatt m.m.)abstract
    • DN DEBATT 15/3.

  • Phase 1 dose de-escalation trial of the endogenous folate [6R]-5,10-methylene tetrahydrofolate in combination with fixed-dose pemetrexed as neoadjuvant therapy in patients with resectable rectal cancer.
  • 2015
  • Ingår i: Investigational new drugs. - : Springer Science and Business Media LLC. - 1573-0646 .- 0167-6997. ; 33:5, s. 1078-1085
  • Tidskriftsartikel (refereegranskat)abstract
    • Background Modufolin® ([6R]-5,10-methylene tetrahydrofolate; [6R]-MTHF) is an endogenous biomodulator that is being developed as an alternative to leucovorin, a folate prodrug used in the treatment of colorectal cancer. The objective of this phase 1 dose de-escalation trial was to estimate the minimum tolerated dose of [6R]-MTHF to be used in combination with pemetrexed 500mg/m(2) in the neoadjuvant treatment of patients with rectal cancer. Methods Adult patients (≥18years) with resectable rectal adenocarcinoma were allocated to [6R]-MTHF doses of 500, 100, 50, and 10mg/m(2) in combination with pemetrexed 500mg/m(2). [6R]-MTHF was administered as an intravenous (i.v.) bolus injection 1week prior to the first dose of pemetrexed and then once weekly for 9weeks; pemetrexed was administered by i.v. infusion once every 21days for three cycles. Results Twenty-four patients (mean [SD] age, 63.1 [12.9] years) were enrolled in the study. A total of 72 treatment-related adverse events (AEs) were reported, of which the most common were fatigue (n=17; 23.6%), nausea (n=10; 13.9%), and diarrhea (n=5; 6.9%). The incidence of treatment-related AEs by [6R]-MTHF dose level (500, 100, 50, 10mg/m(2)) was 11.1% (n=8), 13.9% (n=10), 45.8% (n=33), and 29.2% (n=21), respectively. There were no dose-limiting toxicities, and only two (2.8%) treatment-related AEs were grade 3 in severity. Of the 11 serious AEs reported, none were considered to be related to [6R]-MTHF treatment. Conclusions The results of this phase 1 study indicate that the estimated minimum tolerated dose of [6R]-MTHF was 100mg/m(2) once weekly in combination with pemetrexed 500mg/m(2). The low toxicity profile of [6R]-MTHF supports its further evaluation as a component of systemic chemotherapy in the management of colon and rectal cancer.

  • Effect of stabilization method and freeze/thaw-aided precipitation on structural and functional properties of proteins recovered from brown seaweed (Saccharina latissima)
  • 2019
  • Ingår i: Food Hydrocolloids. - : Elsevier BV. - 0268-005X. ; 96, s. 140-150
  • Tidskriftsartikel (refereegranskat)abstract
    • - Structural, functional and nutritional properties of protein recovered from brown seaweed, S. latissima with alkaline solubilization/isoelectric precipitation as a function of different post-harvest stabilization methods were studied. The latter included freezing at −20 °C/-80 °C, oven-drying, sun-drying, freeze-drying and ensilaging. Also, the efficacy of freeze/thaw-aided precipitation (F/T) in improving protein recovery of the process was evaluated. The freeze-dried, oven-dried, and −20 °C frozen seaweeds resulted in significantly higher protein yield than the −80°C-frozen, sun-dried and ensiled biomasses. F/T increased protein precipitation and doubled total protein yield. Sun-drying and −20°C-freezing caused extensive protein degradation as revealed by SDS-PAGE and HP-SEC, while oven-drying altered the seaweed protein structure with less α-helices. Functional properties of the seaweed proteins were remarkably affected by stabilization condition and F/T, but nutritional value of the proteins was only dependent on stabilization method. Thus, to efficiently recover seaweed proteins, its post-harvest stabilization condition must be carefully chosen based on the final application of the proteins. © 2019 The Authors

  • Three-dimensional modeling of the Ca II H and K lines in the solar atmosphere
  • 2018
  • Ingår i: Astronomy and Astrophysics. - : EDP Sciences. - 0004-6361 .- 1432-0746. ; 611
  • Tidskriftsartikel (refereegranskat)abstract
    • Context. CHROMIS, a new imaging spectrometer at the Swedish 1-m Solar Telescope (SST), can observe the chromosphere in the H and K lines of Ca II at high spatial and spectral resolution. Accurate modeling as well as an understanding of the formation of these lines are needed to interpret the SST/CHROMIS observations. Such modeling is computationally challenging because these lines are influenced by strong departures from local thermodynamic equilibrium, three-dimensional radiative transfer, and partially coherent resonance scattering of photons. Aims. We aim to model the Ca II H and K lines in 3D model atmospheres to understand their formation and to investigate their diagnostic potential for probing the chromosphere. Methods. We model the synthetic spectrum of Ca II using the radiative transfer code Multi3D in three different radiation-magnetohydrodynamic model atmospheres computed with the Bifrost code. We classify synthetic intensity profiles according to their shapes and study how their features are related to the physical properties in the model atmospheres. We investigate whether the synthetic data reproduce the observed spatially-averaged line shapes, center-to-limb variation and compare this data with SST/CHROMIS images. Results. The spatially-averaged synthetic line profiles show too low central emission peaks, and too small separation between the peaks. The trends of the observed center-to-limb variation of the profiles properties are reproduced by the models. The Ca II H and K line profiles provide a temperature diagnostic of the temperature minimum and the temperature at the formation height of the emission peaks. The Doppler shift of the central depression is an excellent probe of the velocity in the upper chromosphere.
