  • Functional Notch signaling is required for BMP4-induced inhibition of myogenic differentiation

    • The bone morphogenetic protein (BMP) and Notch signaling pathways are crucial for cellular differentiation. In many cases, the two pathways act similarly; for example, to inhibit myogenic differentiation. It is not known whether this inhibition is caused by distinct mechanisms or by an interplay between Notch and BMP signaling. Here we demonstrate that functional Notch signaling is required for BMP4-mediated block of differentiation of muscle stem cells, i.e. satellite cells and the myogenic cell line C2C12. Addition of BMP4 during induction of differentiation dramatically reduced the number of differentiated satellite and C2C12 cells. Differentiation was substantially restored in BMP4-treated cultures by blocking Notch signaling using either theγ-secretase inhibitor L-685,458 or by introduction of a dominant-negative version of the Notch signal mediator CSL. BMP4 addition to C2C12 cells increased transcription of two immediate Notch responsive genes, Hes1 and Hey1, an effect that was abrogated by L-685,458. A 3 kb Hey1-promoter reporter construct was synergistically activated by the Notch 1 intracellular domain (Notch 1 ICD) and BMP4. The BMP4 mediator SMAD1 mimicked BMP activation of the Hey1 promoter. A synthetic Notch-responsive promoter containing no SMAD1 binding sites responded to SMAD1, indicating that DNA-binding activity of SMAD1 is not required for activation. Accordingly, Notch 1 ICD and SMAD1 interacted in binding experiments in vitro. Thus, the data presented here provide evidence for a direct interaction between the Notch and BMP signaling pathways, and indicate that Notch has a crucial role in the execution of certain aspects of BMP-mediated differentiation control.

  • Simulating thermal noise

    • Thermal noise measurements by space-borne antennas are commonly used to determine plasma parameters like the electron density and the plasma temperature from the noise spectra. It would be desirable to have a controlled experiment in which noise from a plasma with known properties is sampled in space and in time and which results can then be used to reproduce the satellite measurements. Here we examine the possibility to use particle-in-cell (PIC) simulations as such an experiment. In this work we present a statistically averaged noise spectrum computed with a PIC code for a simple single-Maxwellian and unmagnetized electron plasma and we compare it to both, the thermal noise spectrum for the corresponding real plasma and the noise spectrum we would anticipate from our numerical scheme. We find that we can produce noise fields with sufficiently low amplitudes to keep the plasma in a linear regime. We show that the simulation noise at low and at large wave numbers differs not only from thermal noise of a physical plasma but also from the numerical noise we would expect from our numerical scheme. We explain the drop of the noise power at low wave numbers by our initial conditions. We find experimentally the relation that connects the theoretical noise spectrum for our simulation code with that we actually measure, provided that the phase velocity of the noise is less than the maximum velocity of the computational particles.

    • The design and initial operation of a compact soft x-ray (SXR) multichord camera was studied. The line integrity emissivity was measured along up to 20 lines of sight using an array of miniaturized silicon photodiodes. The application of the diagnostic to a gas-fed magnetoplasma dynamic thruster (MPDT) with an external magnetic field was also discussed.

  • Features associated with epilepsy in the antiphospholipid syndrome

    • OBJECTIVE: To assess the frequency of epilepsy in primary and secondary antiphospholipid syndrome (APS); to analyze the clinical and laboratory features characterizing those with epilepsy in a cohort of 538 patients with APS; and to find associated features that would suggest risk factors for epilepsy in APS. METHODS: We analyzed the clinical features of patients with APS who had epilepsy and compared them to the clinical features of non-epileptic APS patients. RESULTS: Of 538 APS patients, 46 (8.6%) had epilepsy. Epilepsy was more prevalent among APS secondary to systemic lupus erythematosus (SLE) compared to primary APS (13.7% vs 6%; p < 0.05). The patients with epilepsy had a higher prevalence of central nervous system (CNS) manifestations including focal ischemic events (strokes or transient ischemic events, 54.3% vs 24.6%; p < 0.0001) and amaurosis fugax (15.2% vs 4.9%; p < 0.05). APS patients with epilepsy had a higher frequency of valvular pathology (30.4% vs 14.6%; p < 0.01), thrombocytopenia (43.5% vs 25%; p < 0.05), and livedo reticularis (26.1% vs 11.5%; p < 0.01). The multivariate logistic regression analysis found CNS thromboembolic events as the most significant factor associated with epilepsy, with an odds ratio (OR) of 4.05 (95% confidence interval, CI: 2.05-8), followed by SLE (OR 1.4, 95% CI 1.2-4.7), and valvular vegetations (OR 2.87, 95% CI 1-8.27). CONCLUSION: Epilepsy is common in APS and most of the risk seems to be linked to vascular disease as manifested by extensive CNS involvement, valvulopathy, and livedo reticularis and to the presence of SLE. These factors, however, explain only part of the increased occurrence of epilepsy in APS and other causes such as direct immune interaction in the brain should be investigated.

    • Regular and stochastic behavior in single particle orbits in static magnetic reversals have wide application in laboratory and physical plasmas. In a simple magnetic reversal, the system has three degrees of freedom but only two global (exact) constants of the motion, the system is nonintegrable and the particle motion can, under certain conditions, exhibit chaotic behavior. Here, we consider the dynamics when a constant shear field is added. In this case, the form of the potential changes from quadratic to velocity dependent. We use numerically integrated trajectories to show that the effect of the shear field is to break the symmetry of the system so that the topology of the invariant tori of regular orbits is changed. In this case, invariant tori take the form of nested Moebius strips in the presence of the shear field. The route to chaos is via bifurcation (period doubling) of the Moebius strip tori.
