    • The ATLAS Inner Detector consists of three sub-systems, the Pixel Detector at the innermost radius, the Semi-Conductor Tracker at intermediate radii, and the Transition Radiation Tracker (TRT) at the outermost radius in front of the electromagnetic calorimeter. The TRT provides a combination of continuous tracking with many projective measurements based on individual drift-tubes (or straws) and of electron identification based on radiator fibres or foils interleaved between the straws themselves. This paper describes the current status of design and construction of the various components of the TRT: the assembly of the barrel modules has recently been completed, that of the end-cap wheels is well underway, and the on-detector front-end electronics is in production. The detector modules and front-end electronics boards will be integrated together over the next year, the barrel and end-cap TRT parts will be assembled and tested with their SCT counterparts during 2005 and installation and commissioning in the ATLAS pit will take place at the end of 2005 and the beginning of 2006.     • A summary of the aging and material validation studies carried out for the ATLAS Transition Radiation Tracker (TRT) is presented. Particular emphasis is put on the different phenomena observed in straw tubes operating with the chosen Xe/CF4/CO2 mixture. The most serious effects observed are silicon deposition on the anode wire and damage of the anode wire gold plating. Etching phenomena and active radical effects are also discussed. With a careful choice of all materials and components, and with good control of the water contamination in the active gas, the ATLAS TRT will operate reliably for 10 years at the LHC design luminosity. To demonstrate this fully, more work is still needed on the gas system purification elements, in particular to understand their interplay with the active species containing fluorine created in the avalanche process under irradiation.

    • A prototype of the Transition Radiation Tracker (TRT) for the ATLAS experiment at the CERN LHC has been built and tested at the CERN SPS. Detailed studies of the drift-time measurements, alignment technique, hit registration efficiency, track and momentum accuracy were performed. A coordinate measurement accuracy of 150 Pin for a single TRT drift tube and momentum resolution of 0.8% for 20 GeV pions in a 1.56 T magnetic field were achieved. The results obtained are in agreement with the expected tracking performance of the ATLAS TRT. (C) 2001 Published by Elsevier Science B.V.

    • The objective was to determine the activation of white blood cells (WBCs) and endothelial cells in patients with healed venous ulcer and the influence of the standing position and of treatment with flavonoids. Ten patients with a healed venous ulcer were treated with flavonoid substance (90% diosmin), 1000 mg three times daily for 30 days. Blood samples were taken from arm and dorsal foot veins before and after standing for 30 minutes. Blood sampling was performed before treatment, after three days, one month and three months. The activation of WBCs was determined by measuring adhesion molecule CD11b and CD18 expression on the surface of granulocytes and monocytes. In addition, interleukin 6 (IL-6), IL-8, soluble E-selectin (sE-selectin), sL-selectin and sICAM-1 levels in serum were quantified. The results showed that standing did not influence any of the measured parameters significantly. Expression of CD11b adhesion molecules on granulocytes was significantly up-regulated (p=0.044) after treatment with flavonoids for one month, but this increase was not significant (p=0.056) two months after the treatment period compared with the baseline level. The expression of CD18 remained unchanged. Baseline expression of CD11b or CD18 on monocytes did not change significantly during the study period. Neither was any significant change observed in the levels of IL-6, IL-8 or the soluble adhesion molecules. It was concluded that flavonoid treatment for 30 days increased the expression of CD11b adhesion molecules on circulating granulocytes. No general effect on the inflammatory process could be observed as assessed by levels of cytokines and soluble adhesion molecules. Possible explanations for these findings could be that a decreased number of primed granulocytes leave the circulation due to a changed WBC/endothelial cell interaction or that flavonoids have a direct effect on granulocytes. Further studies are needed to clarify the mode of action of flavonoids in chronic venous disease.

    • BACKGROUND: To investigate the value of combined treatment with allopurinol and 5-aminosalicylic (5-ASA) based drugs as maintenance treatment for ulcerative colitis (UC). METHODS: 199 patients with UC in remission but with active disease during the preceding 3 years were included. Allopurinol 100 mg twice daily or placebo was added to the 5-ASA based maintenance treatment. Clinical and endoscopic follow up was performed after 1, 6 and 12 months. RESULTS: Intention-to-treat analysis after 6 and 12 months showed similar results in both groups. A log-rank test showed that 77% in the allopurinol compared to 59% in the placebo group were still in remission after 6 months (P=0.0083) and 62% and 53% after 12 months, respectively (P=0.0936). This was mainly due to a higher than expected number of relapses during the first 3 months in the placebo group. After the first 3 months, the rate of relapse in each group was similar. CONCLUSIONS: It appears possible that allopurinol in combination with 5-ASA is better than 5-ASA alone for a 6-month, but not a 12-month period. This has to be verified in further dose-ranging studies.

    • Ulcerative colitis (UC), a chronic inflammatory bowel disease, exhibits pronounced increase of T lymphocytes in the inflamed mucosa. To understand the role of intestinal T lymphocytes in the pathogenesis of UC their cytokine production in the mucosa was analysed. Intestinal T lymphocytes of UC, Crohn's disease and control patients were analysed for cytokine mRNA levels by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR) directly after isolation without in vitro stimulation. Frequencies of cytokine positive cells were determined in UC and control colon by immunomorphometry. T lymphocytes in normal colon expressed interleukin (IL)-2, interferon (IFN)-gamma, tumour necrosis factor (TNF)-alpha and transforming growth factor (TGF)-beta1, but not IL-4, IL-5 or IL-10. In UC, a highly significant increase in IL-10 mRNA levels in T lymphocytes and an increased frequency of IL-10 positive cells was seen in colon. IL-10 mRNA levels were also elevated in T lymphocytes of the non-inflamed ileum and correlated with disease activity at both locations. CD4+ T lymphocytes were the major source of IL-10 mRNA. IL-2, IFN-gamma and TNF-alpha mRNA levels were decreased in colonic T lymphocytes, and virtually no IL-2, IFN-gamma, TNF-alpha or TGF-beta positive cells were detected in basal lymphoid aggregates. However, scattered IL-10 positive cells were found here. Lamina propria outside the aggregates contained IL-10-, IFN-gamma, TNF-alpha and TGF-beta but not IL-2 positive cells. T cells of UC patients did not express IL-4 or IL-5. Taken, together the data suggest a generalized activation of IL-10 producing CD4+ T cells along the intestine of UC patients. The local environment seems to determine the biological consequences of elevated IL-10.
