  • A saposin-lipoprotein nanoparticle system for membrane proteins

    • A limiting factor in membrane protein research is the ability to solubilize and stabilize such proteins. Detergents are used most often for solubilizing membrane proteins, but they are associated with protein instability and poor compatibility with structural and biophysical studies. Here we present a saposin-lipoprotein nanoparticle system, Salipro, which allows for the reconstitution of membrane proteins in a lipid environment that is stabilized by a scaffold of saposin proteins. We demonstrate the applicability of the method on two purified membrane protein complexes as well as by the direct solubilization and nanoparticle incorporation of a viral membrane protein complex from the virus membrane. Our approach facilitated high-resolution structural studies of the bacterial peptide transporter PeptT(S02) by single-particle cryo-electron microscopy (cryo-EM) and allowed us to stabilize the HIV envelope glycoprotein in a functional state.

    • Creative AI (notably GANs and VAEs) can generate convincing fakes of video footage, pictures, graphics, etc. In order to conceptualize the societal role of creative AI a new conceptual toolbox is needed. The paper provides metaphors and concepts for understanding the functioning of creative AI. It shows how the role of creative AI in relation to FAT ideals can be enriched by a dynamic and constructivist understanding of creative AI. The paper proposes to use Greimas’ actantial model as a heuristic in the operationalization of this type of understanding of creative AI.

  • Ethical struggles: Living stories from the frontline of retail banking

    • In the aftermath of the global financial crisis, the financial sector globally has become much more reg-ulated to ensure ’ethical’ conduct, yet we have still to understand how the banks has worked to ensure a more cultural shift towards ethical conduct. The purpose of our paper is to examine the cultural changes in three different retail banks, focusing specifically on ethical sales and advising among front-line personnel. Our research question is: how is the appropriate advisor discursively constructed by management and the advisors. Inspired by Bakhtin and Aristotle’s theoretical lenses on practical eth-ics, we bring forward a vocabulary for discussing the ’ethics’ of retail banking in the front line today as well as empirical evidence for the emergence of new plots, identities and cultures of ’good bank-ing’. We argue that our empirical material derived from the cases studies of three different Scandinavi-an banks contributes to an understanding of how ethics may become a governing principle of collec-tive organizational practices and culture in the post-crisis financial sector. Our main academic contri-bution concerns the identification of three key ethical resources and forces that are entangled into each other in an unfinalized and incomplete wholeness, mutually struggling with each other in bringing forward an ethical cultural conduct yet-to-be-achieved. The practical contribution consists in the em-pirical findings of different managerial approaches to growing ethical cultures among frontline per-sonnel. The managerial challenge remains within the ethical struggles involved in the becoming of ethical organizational cultures. As the world is indeterminate and incomplete, ethics remains an ongo-ing work and task compelling practitioners at all levels of the organization to take it on.

  • The 2.5D MST for sound propagation through arrays of cylinders parallel to the ground

    • In this work sound propagation through arrays of cylinders oriented parallel to the ground is of interest. The structures are placed in a three-dimensional domain and are insonified by a monopole or incoherent line source. Assuming a cross-sectionally invariant structure one can efficiently obtain the 3D pressure field for such arrangements by post-processing a series of 2D solutions - a technique usually referred to as a 2.5D transform. Since the initiation of the 2.5D transform for outdoor sound propagation it has been successfully applied together with frequency domain methods such as the Boundary Element Method and the Equivalent Sources Method. However, to predict for sound propagation through sonic crystal noise barriers the 2D Multiple Scattering Theory (2D MST) is often used, and has proven to be very efficient. We therefore introduce the 2.5D MST to solve for 3D scattering by clusters of acoustically rigid cylinders. It will be shown that only a few simple substitutions applied to the 2D MST kernel allows us to solve for imaginary wave numbers, which are needed in the 2.5D transform. The proposed method is numerically validated for two basic cases: (i) a point source above rigid ground, and (ii) off-axis scattering by a cylinder in free-field. Both are shown to be in excellent agreement with the respective reference calculations. We further demonstrate some calculation results for sound propagation through graded index sonic crystals, and find that off-axis insonification of these structures shifts the characteristic frequency response upwards, as could be expected. Finally, we also present calculation results for infinite and finite incoherent line sources and display the existence of a spectral smearing effect for both source types.

  • The fW-mean filter framework for topology optimization and analysis of Friedrichs systems

    • Part I. Topology optimization is the most general form of design optimization in which the optimal layout of material within a given region of space is to be determined. Filters are essential components of many successful density based topology optimization approaches. The generalized fW-mean filter framework developed in this thesis provides a unified platform for construction, analysis, and implementation of filters. Extending existing algorithms, we demonstrate that under special albeit relevant conditions, the computational complexity of evaluating generalized fW-mean filters and their derivatives is linear in the number of design degrees of freedom. We prove that generalized fW-mean filters guarantee existence of solutions to the penalized minimum compliance problem, the archetypical problem in density based topology optimization. In this problem, the layout of linearly elastic material that minimizes the compliance given static supports and loads is to be determined. We formalize the connection between mathematical morphology and the notion of minimum length scale of a layout of material and thereby provide a theoretical foundation for imposing and assessing minimum length scales in density based topology optimization. Elaborating on the fact that some sequences of generalized fW-mean filters provide differentiable approximations of morphological operators, we devise a method capable of imposing different minimum length scales on the two material phases in minimum compliance problems. Part II. The notion of Friedrichs systems, also known as symmetric positive systems, encompasses many linear models of physical phenomena. The prototype model is Maxwell's equations, which describe the evolution of the electromagnetic field in the presence of electrical charges and currents. In this thesis, we develop well-posed variational formulations of boundary and initial–boundary value problems of Friedrichs systems on bounded domains. In particular, we consider an inhomogeneous initial–boundary value problem that models lossless propagation of acoustic disturbances in a stagnant fluid. Galbrun's equation is a linear second order vector differential equation in the so-called Lagrangian displacement, which was derived to model lossless propagation of acoustic disturbances in the presence of a background flow. Our analysis of Galbrun's equation is centered on the observation that solutions to Galbrun's equation may be formally constructed from solutions to linearized Euler's equations. More precisely, the Lagrangian displacement is constructed as the solution to a transport-type equation driven by the Eulerian velocity perturbation. We present partial results on the well-posedness of Galbrun's equation in the particular case that the background flow is everywhere tangential to the domain boundary by demonstrating mild well-posedness of an initial–boundary value problem for linearized Euler's equations and that our construction of the Lagrangian displacement is well-defined. Moreover, we demonstrate that sufficiently regular solutions to Galbrun's equation satisfy an energy estimate.

Undersøkelsen danner et grunnlag for senere faser i prosjektet der vi skal utføre en postal spørreundersøkelse, analysere lokalavisers dekning av vindkraftutbygging, foreta dybdeintervju av nøkkelinformanter samt systematisere kunnskap om barrierer mot vindkraft.
