The Genome Sequence of Taurine Cattle : A Window to Ruminant Biology and Evolution

- To understand the biology and evolution of ruminants, the cattle genome was sequenced to about sevenfold coverage. The cattle genome contains a minimum of 22,000 genes, with a core set of 14,345 orthologs shared among seven mammalian species of which 1217 are absent or undetected in noneutherian (marsupial or monotreme) genomes. Cattle-specific evolutionary breakpoint regions in chromosomes have a higher density of segmental duplications, enrichment of repetitive elements, and species-specific variations in genes associated with lactation and immune responsiveness. Genes involved in metabolism are generally highly conserved, although five metabolic genes are deleted or extensively diverged from their human orthologs. The cattle genome sequence thus provides a resource for understanding mammalian evolution and accelerating livestock genetic improvement for milk and meat production.

- Decays of neutron-rich A similar to 190 nuclei have been studied following projectile fragmentation of a Pb-208 beam on a Be-9 target at the GSI Fragment Separator. Gamma-ray decays from previously reported isomeric states in Ta-188, W-190 and (192,193) Re were used as internal calibrations for the particle identification analysis, together with the identification of previously unreported isomeric decays in Ta-189 and W-191. The current work also identifies beta-delayed gamma rays following the decay of Ta-188 to W-188 for the first time.

- The low-lying structure of W-188,W-190,W-192 has been studied following beta decays of the neutron-rich mother nuclei Ta-188,Ta-190,Ta-192 produced following the projectile fragmentation of a 1-GeV-per-nucleon Pb-208 primary beam on a natural beryllium target at the GSI Fragment Separator. The beta-decay half-lives of Ta-188, Ta-190, and Ta-192 have been measured, with gamma-ray decays of low-lying states in their respective W daughter nuclei, using heavy-ion beta-gamma correlations and a position-sensitive silicon detector setup. The data provide information on the low-lying excited states in W-188, W-190, and W-192, which highlight a change in nuclear shape at W-190 compared with that of lighter W isotopes. This evolution of ground-state structure along the W isotopic chain is discussed as evidence for a possible proton subshell effect for the A similar to 190 region and is consistent with maximization of the gamma-softness of the nuclear potential around N similar to 116.

- Core-level photoelectron spectra of free neutral methane clusters have been recorded. These spectra exhibit well-resolved surface and bulk features as well as vibrational fine structure. The vibrational structure in the cluster signal is well reproduced by a theoretical model that assumes independent contributions from inter- and intramolecular modes. The intramolecular contribution to the vibrational line-shape is taken to be equal to that of the monomer in the gas phase, while the intermolecular part is simplified to line broadening. An estimate of the cluster size has been made on the basis of the observed surface-to-bulk intensity ratio.

- Fourier Transform mid-infrared (FT-IR), Fourier Transform Raman (FT-Raman) and photoluminescence spectroscopies and Two-Dimensional (2D) Correlation Spectroscopic Analysis were employed to examine the anionic and cationic local environments in mono-urethanesils doped with europium triflate (Eu(CF 3-SO3)3). The hybrid host framework of these materials is composed of a siliceous backbone bonded through urethane linkages to CH3-terminated polymer chains containing about 7 OCH 2CH2 units. Samples with ∞ ≥ n (composition) ≥5 (where n = OCH2CH2/Eu3+) were studied. In terms of ionic association, the level of complexity of these xerogels is very high. In all the compounds the triflate ions exist "free", weakly coordinated and forming cross-link separated ion pairs. At 20 ≥ n ≥ 5, in addition to all these species contact ion pairs occur. In agreement with these conclusions, photoluminescence establishes the presence of three distinct cation local sites (Eu3+/O=C(urethane cross-links), Eu3+/O-C- C(polyether chains) and weakly coordinated Eu3+/CF3SO 3- ionic pairs).

A novel pathogenic pathway of immune activation detectable before clinical onset in Huntington's disease.

- Huntington's disease (HD) is an inherited neurodegenerative disorder characterized by both neurological and systemic abnormalities. We examined the peripheral immune system and found widespread evidence of innate immune activation detectable in plasma throughout the course of HD. Interleukin 6 levels were increased in HD gene carriers with a mean of 16 years before the predicted onset of clinical symptoms. To our knowledge, this is the earliest plasma abnormality identified in HD. Monocytes from HD subjects expressed mutant huntingtin and were pathologically hyperactive in response to stimulation, suggesting that the mutant protein triggers a cell-autonomous immune activation. A similar pattern was seen in macrophages and microglia from HD mouse models, and the cerebrospinal fluid and striatum of HD patients exhibited abnormal immune activation, suggesting that immune dysfunction plays a role in brain pathology. Collectively, our data suggest parallel central nervous system and peripheral pathogenic pathways of immune activation in HD.

- A thermally activated photoluminescence memory effect, induced by a reversible order-disorder phase transition of the alkyl chains, is reported for highly organized bilayer alkyl/siloxane hybrids (see figure; left at room temperature, right at 120 degrees C). The emission energy is sensitive to the annihilation/formation of the hydrogen-bonded amide-amide array displaying a unique nanoscopic sensitivity (ca. 150 nm).
