  • Analysis of Molecular Size Distributions of Cellulose Molecules during Hydrolysis of Cellulose by Recombinant Cellulomonas fimi -1,4-Glucanases

    • Four -1,4-glucanases (cellulases) of the cellulolytic bacterium Cellulomonas fimi were purified from Escherichia coli cells transformed with recombinant plasmids. Previous analyses using soluble substrates had suggested that CenA and CenC were endoglucanases while CbhA and CbhB resembled the exo-acting cellobiohydrolases produced by cellulolytic fungi. Analysis of molecular size distributions during cellulose hydrolysis by the individual enzymes confirmed these preliminary findings and provided further evidence that endoglucanase CenC has a more processive hydrolytic activity than CenA. The significant differences between the size distributions obtained during hydrolysis of bacterial microcrystalline cellulose and acid-swollen cellulose can be explained in terms of the accessibility of -1,4-glucan chains to enzyme attack. Endoglucanases and cellobiohydrolases were much more easily distinguished when the acid-swollen substrate was used.

    • The ionic conductivity of a polymeric fast-ion-conductor based on LiCF3SO3 salt and poly(acrylonitrile), [CH2CH(CN)]n), is enhanced by ∼5 orders of magnitude when the composition approaches the "polymer-in-salt" regime; i.e., when the salt content increases from N:Li=12:1 to 1.2:1 (or ∼70 wt % of salt). This is in contrast to common salt-in-polymer electrolytes where a conductivity maximum typically is encountered at intermediate compositions. We suggest that connectivity effects in a microscopically phase segregated material may influence the long-range migration of charge carriers. Conductivity data are augmented with Raman spectroscopic investigations, thus probing microscopic details regarding the state of the dissolved salt.

    • Future manufacturing systems will be required to be agile, flexible and fault-tolerant. The next-generation intelligent manufacturing systems will be multi-agent systems containing distributed control and application entities that dynamically collaborate to satisfy both local and global objectives. This paper focuses on the development of a generic control system design based on IEC-1499 function block standards, for real-time distributed manufacturing environments. The paper first describes the related research into the control system architecture using multi-agent cooperation, then reports on the intelligent controller design interface and automatic control code generation.
