    • Theelectronic structure in the new transition-metal carbide Ti4SiC3 has beeninvestigated by bulk-sensitive soft x-ray emission spectroscopy and compared tothe well-studied Ti3SiC2 and TiC systems. The measured high-resolution TiL, C K, and Si L x-ray emission spectra arediscussed with ab initio calculations based on density-functional theory includingcore-to-valence dipole matrix elements. The detailed investigations of the Ti-Cand Ti-Si chemical bonds provide increased understanding of the physicalproperties of these nanolaminates. A strongly modified spectral shape isdetected for the intercalated Si monolayers due to Si 3phybridization with the Ti 3d orbitals. As a result ofrelaxation of the crystal structure and the charge-transfer from Ti(and Si) to C, the strength of the Ti-C covalentbond is increased. The differences between the electronic and crystalstructures of Ti4SiC3 and Ti3SiC2 are discussed in relation tothe number of Si layers per Ti layer in thetwo systems and the corresponding change of materials properties.

  • Electronic structure investigation of Ti3AlC2 , Ti3SiC2 , and Ti3GeC2 by soft x-ray emission spectroscopy

    • The electronic structures of epitaxially grown films of Ti3AlC2 , Ti3SiC2 , and Ti3GeC2 have been investigated by bulk-sensitive soft x-ray emission spectroscopy. The measured high-resolution Ti L , C K , Al L , Si L , and Ge M emission spectra are compared with ab initio density-functional theory including core-to-valence dipole matrix elements. A qualitative agreement between experiment and theory is obtained. A weak covalent Ti-Al bond is manifested by a pronounced shoulder in the Ti L emission of Ti3AlC2 . As Al is replaced with Si or Ge, the shoulder disappears. For the buried Al and Si layers, strongly hybridized spectral shapes are detected in Ti3AlC2 and Ti3SiC2 , respectively. As a result of relaxation of the crystal structure and the increased charge-transfer from Ti to C, the Ti-C bonding is strengthened. The differences between the electronic structures are discussed in relation to the bonding in the nanolaminates and the corresponding change of materials properties.

  • Targeted Resequencing and Analysis of the Diamond-Blackfan Anemia Disease Locus RPS19

    • BACKGROUND: The Ribosomal protein S19 gene locus (RPS19) has been linked to two kinds of red cell aplasia, Diamond-Blackfan Anemia (DBA) and Transient Erythroblastopenia in Childhood (TEC). Mutations in RPS19 coding sequences have been found in 25% of DBA patients, but not in TEC patients. It has been suggested that non-coding RPS19 sequence variants contribute to the considerable clinical variability in red cell aplasia. We therefore aimed at identifying non-coding variations associated with DBA or TEC phenotypes. METHODOLOGY/PRINCIPAL FINDINGS: We targeted a region of 19'980 bp encompassing the RPS19 gene in a cohort of 89 DBA and TEC patients for resequencing. We provide here a catalog of the considerable, previously unrecognized degree of variation in this region. We identified 73 variations (65 SNPs, 8 indels) that all are located outside of the RPS19 open reading frame, and of which 67.1% are classified as novel. We hypothesize that specific alleles in non-coding regions of RPS19 could alter the binding of regulatory proteins or transcription factors. Therefore, we carried out an extensive analysis to identify transcription factor binding sites (TFBS). A series of putative interaction sites coincide with detected variants. Sixteen of the corresponding transcription factors are of particular interest, as they are housekeeping genes or show a direct link to hematopoiesis, tumorigenesis or leukemia (e.g. GATA-1/2, PU.1, MZF-1). CONCLUSIONS: Specific alleles at predicted TFBSs may alter the expression of RPS19, modify an important interaction between transcription factors with overlapping TFBS or remove an important stimulus for hematopoiesis. We suggest that the detected interactions are of importance for hematopoiesis and could provide new insights into individual response to treatment.

  • Seeing through the blind! : ability of hospital staff to differentiate morphine from placebo, in neonates at a placebo controlled trial

    • AIM: To investigate whether professional training and/or clinical experience affect the ability of caregiver to assess clinical signs of pre-emptive morphine analgesia. METHODS: In the Neurological Outcomes & Pre-emptive Analgesia In Neonates trial preterm infants undergoing mechanical ventilation were randomized to receive continuous infusion, either of morphine or placebo blinded. Staff from centres in Sweden (Stockholm and Orebro) completed an assessment form. RESULTS: A total of 360 assessment forms were collected from 52 neonates. In 59% of the cases, caregivers correctly identified patients group. Comparable proportion of answers were correct between physicians, nurses and assistant nurses (63, 60 and 54%, respectively, p = 0.60). Staff with Neonatal intensive care unit experience <1 year identified 63%, as compared to 65% for working 1-5 year, and 55% that has been working >5 years (p = 0.28). Staff's ability to correctly identify group assignment was reduced by amount of additional morphine (p < 0.01) and severity of illness (p = 0.01). CONCLUSIONS: Clinical medical staffs, including neonatologists, have great difficulties in assessing the presence and severity of pain. Further studies should focus on the methods for assessment of prolonged pain in preterm neonates, define the effects of adequate analgesia, and investigate the clinical factors that may alter neonatal responses to acute and prolonged pain.

  • Single crystal CrN/ScN superlattice soft X-ray mirrors : epitaxial growth, structure, and properties

    • Single crystal CrN/ScN superlattice films with modulation periods of 1.64 nm were grown on MgO(001) substrates. By utilizing a magnetically enhanced plasma in the vicinity of the substrate and a negative substrate bias, ion/metal nitride flux ratios of 45 and 144 were achieved during deposition of CrN and ScN, respectively. The effects of ion energies in the range [16–58 eV] and substrate temperatures in the range [535–853 °C] on the composition, interface width, crystal quality, and microstructure evolution were investigated using elastic recoil detection analysis, hard X-ray reflectivity, X-ray diffraction, and transmission electron microscopy (TEM). Minimal interface widths of 0.2 nm = 1/2 nitride unit cell were achieved at a growth temperature of 735 °C and ion energies of 24 and 28 eV for CrN and ScN, respectively. Under these conditions, also an optimum in the crystal quality was observed for near stoichiometric composition of CrN and ScN. TEM confirmed a cube-on-cube epitaxial relationship for the system with CrN(001)ScN(001)MgO(001) and CrN[100]ScN[100]MgO[100]. Also, the layers were coherently strained to each other with no misfit dislocations, threading dislocations, surface cusps, voids or gas bubbles present. Higher ion energies or lower deposition temperatures gave over-stoichiometric films with poor superlattice modulation while higher growth temperatures yielded a decreased crystal quality, due to loss of N. As-deposited superlattices with only 61 periods exhibited an absolute soft X-ray reflectance of 6.95% at an energy of 398.8 eV (Sc 2p-absorption edge) which is comparable to the performance of Cr/Sc. The compositional modulation and phase structure was stable during extended annealing at 850 °C, which is the highest thermal stability for an X-ray multilayer mirror. It is concluded that the ScN layers serve as effective diffusion barriers to hinder decomposition of the CrN layers and stabilize the pseudomorphic superlattice structure. Nanoindentation experiments showed that the hardness of the CrN/ScN superlattice films was 19 GPa.

  • Wave energy converter with enhanced amplitude response at frequencies coinciding with Swedish west coast sea states by use of a supplementary submerged body

    • The full-scale direct-driven wave energy converter developed at Uppsala University has been in offshore operation at the Swedish west coast since 2006. Earlier simulations have now been validated by full-scale experiment with good agreement. Based on that, a theoretical model for a passive system having optimum amplitude response at frequencies coinciding with Swedish west coast conditions has been developed. The amplitude response is increased by adding supplementary inertia by use of the additional mass from a submerged body. A sphere with neutral buoyancy is chosen as the submerged body and modeled as being below the motion of the waves. The model is based on potential linear wave theory and the power capture ratio is studied for real ocean wave data collected at the research test site. It is found that the power capture ratio for the two body system can be increased from 30% to 60% compared to a single body system. Increased velocity in the system also decreases the value for optimal load damping from the generator, opening up the possibility to design smaller units.
