  • New genetic loci link adipose and insulin biology to body fat distribution.

    • Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms.

  • In-Situ Probing of H2O Effects on a Ru-Complex Adsorbed on TiO2 Using Ambient Pressure Photoelectron Spectroscopy

    • Dye-sensitized interfaces in photocatalytic and solar cells systems are significantly affected by the choice of electrolyte solvent. In the present work, the interface between the hydrophobic Ru-complex Z907, a commonly used dye in molecular solar cells, and TiO2 was investigated with ambient pressure photoelectron spectroscopy (AP-PES) to study the effect of water atmosphere on the chemical and electronic structure of the dye/TiO2 interface. Both laboratory-based Al K alpha as well as synchrotron-based ambient pressure measurements using hard X-ray (AP-HAXPES) were used. AP-HAXPES data were collected at pressures of up to 25 mbar (i.e., the vapor pressure of water at room temperature) showing the presence of an adsorbed water overlayer on the sample surface. Adopting a quantitative AP-HAXPES analysis methodology indicates a stable stoichiometry in the presence of the water atmosphere. However, solvation effects due to the presence of water were observed both in the valence band region and for the S 1s core level and the results were compared with DFT calculations of the dye-water complex.

  • The lawn as a social and cultural phenomenon in Sweden

    • Lawns have a significant influence on the cityscape as one of the essential elements of green spaces and an important part of people's everyday lives. Most people in the Western world view lawns as a compulsory element of the urban landscape, almost an icon, without questioning their social, symbolic, ecological or aesthetic values. This research is a part of the conceptual framework and methodological approaches that are being used in an ongoing transdisciplinary collaboration project to study lawns in Sweden as a social and ecological phenomenon.The overall aim of this study was to investigate social and cultural perceptions of lawns, as well as motives behind decisions about the establishment and management of lawns in Sweden. Two multifamily housing typologies, the 'Million Programme' and People's Homes', were examined due to their dominance in Swedish cities. We also studied how an alternative vision of conventional lawns can be applied and accepted by urban residents. We estimated lawn cover in multi-family housing areas and links to people's perception and use of lawns. Questionnaires, semi-structured interviews and observational studies were used (N = 300). Our results showed that people like lawns even if they do not always directly use them. Lawns cover the most significant amount of outdoor spaces in all multi-family residential areas and accompany people everywhere from the house to the schoolyard or park. The total lawn cover in the study areas was 27.8%. Lawns were particularly valued as important places for different outdoor activities (playing, resting, picnicking, walking, socialising) and enjoying the green colour. However people do not want to use a vast monotonous lawn, but a variety of spaces that provide good conditions for different senses ( sound, smell, touch and sight) and activities. Alternative lawns were also appreciated by many citizens, politicians, planners and managers. The implementation of new types of lawns requires special planning and design solutions adjusted for each particular neighbourhood. (C) 2016 The Authors. Published by Elsevier GmbH.

  • Genome-wide meta-analysis uncovers novel loci influencing circulating leptin levels

    • Leptin is an adipocyte-secreted hormone, the circulating levels of which correlate closely with overall adiposity. Although rare mutations in the leptin (LEP) gene are well known to cause leptin deficiency and severe obesity, no common loci regulating circulating leptin levels have been uncovered. Therefore, we performed a genome-wide association study (GWAS) of circulating leptin levels from 32,161 individuals and followed up loci reaching P<10(-6) in 19,979 additional individuals. We identify five loci robustly associated (P<5 × 10(-8)) with leptin levels in/near LEP, SLC32A1, GCKR, CCNL1 and FTO. Although the association of the FTO obesity locus with leptin levels is abolished by adjustment for BMI, associations of the four other loci are independent of adiposity. The GCKR locus was found associated with multiple metabolic traits in previous GWAS and the CCNL1 locus with birth weight. Knockdown experiments in mouse adipose tissue explants show convincing evidence for adipogenin, a regulator of adipocyte differentiation, as the novel causal gene in the SLC32A1 locus influencing leptin levels. Our findings provide novel insights into the regulation of leptin production by adipose tissue and open new avenues for examining the influence of variation in leptin levels on adiposity and metabolic health.

  • Genetic studies of body mass index yield new insights for obesity biology.

    • Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

  • Different patterns of second-line treatment in type 2 diabetes after metformin monotherapy in Denmark, Finland, Norway and Sweden (D360 Nordic) : A multinational observational study.

    • Aims: The understanding of second-line use of glucose-lowering drugs (GLDs) in the general population with type 2 diabetes (T2D) treatment is important as recent results have shown cardiovascular benefits with sodium-glucose cotransporter-2 inhibitors (SGLT-2i) and glucagon-like peptide-1 receptor agonists (GLP-1RA). Our aim was to describe second-line GLD treatment patterns in four Nordic countries.Methods: All T2D patients treated with GLD between 2006 and 2015 were identified in prescribed drug registries in Denmark, Finland, Norway and Sweden, and linked with National Patient and Cause of Death Registries. Second-line treatment was defined as a prescription of a second GLD class following ≥6 months of metformin monotherapy. Index was the date of first dispense of the second-line drug.Results: A rapid uptake of newer GLDs (GLP-1RA, DPP-4i and SGLT-2i) over the 10-year observation period was seen in Denmark, Finland and Norway, while slower in Sweden. In 2015, 33,880 (3.1%) of 1,078,692 T2D patients initiated second-line treatment, and newer GLDs were more commonly used in Finland (92%), Norway (71%) and Denmark (70%) vs Sweden (44%). In 2015, the use of older GLDs (insulin and sulphonylureas) was 7-fold greater in Sweden compared to Finland (49% vs 7%), and 1.6-fold greater compared with Denmark and Norway (49% vs 30% and 29%, respectively).Conclusions: Despite comparable demography and healthcare systems in four neighbouring countries, surprisingly large differences in second-line use of newer GLDs were found. With recent evidence of potential cardiovascular benefits with newer GLDs, such differences may have an important impact on cardiovascular outcomes.

    • Large consortia have revealed hundreds of genetic loci associated with anthropometric traits, one trait at a time. We examined whether genetic variants affect body shape as a composite phenotype that is represented by a combination of anthropometric traits. We developed an approach that calculates averaged PCs (AvPCs) representing body shape derived from six anthropometric traits (body mass index, height, weight, waist and hip circumference, waist-to-hip ratio). The first four AvPCs explain >99% of the variability, are heritable, and associate with cardiometabolic outcomes. We performed genome-wide association analyses for each body shape composite phenotype across 65 studies and meta-analysed summary statistics. We identify six novel loci: LEMD2 and CD47 for AvPC1, RPS6KA5/C14orf159 and GANAB for AvPC3, and ARL15 and ANP32 for AvPC4. Our findings highlight the value of using multiple traits to define complex phenotypes for discovery, which are not captured by single-trait analyses, and may shed light onto new pathways.

    • In this paper we report on an attempt to let students, companies, and organisations themselves discover the kinds of technologies that could be useful when co-producing knowledge in a Master’s-level course in innovation and design. Traditionally, and for various good reasons such as security and stability, universities have had certain online tools and systems for collaboration, while companies and municipalities have had others. These systems support internal communication within organisations but do not necessarily enhance communication with external contacts. This use of different systems creates barriers to the iterative, recurring, convenient, non-hierarchical, and open online collaboration needed in an innovative design process involving multiple stakeholders. During a ten-week Master’s-level course in innovation and design in 2016 and 2017 the 38 students divided into five project groups established contact with five companies and organisations and could choose their own online tools in dialogue with them. This paper presents the students’ and organisations’ emerging practices during the process based on observations and reflective evaluations conducted during and after the course. The results are discussed in light of how socio-material assemblages formed in this special setting and how the results might be used to improve the teaching of online literacy in design collaboration. The result indicates that for co-production of knowledge in innovation and design projects, three new social media literacies would be useful: meta communication, peak performance, and design awareness.
