    • Since April 2004, preparatory work prior to CO2injection hasbeen conducted in the CO2SINK Project, the European Union’sfirst research and development activity on the in-situ testing ofgeological storage of CO2near the town of Ketzin, Germany.Carbon dioxide will be injected into a saline aquifer of the TriassicStuttgart Formation in an anticlinal structure of the northeastGerman Basin. The drilling of one injection and two observationwells will commence at the end of 2006. The predrilling phasefocuses on the baseline geological parameters of the anticline. TheStuttgart Formation is lithologically heterogeneous; it consists ofsandy channel-(string)-facies rocks, with good reservoir propertiesalternating with muddy flood-plain-facies rocks of poor reservoirquality. Playa-type rocks form the immediate cap rock above theCO2SINK reservoir. A geostatistical approach has been applied todescribe the reservoir architecture between and beyond well con-trol. This model forms the basis for the generation of reservoir-dynamic models of CO2injection that assist in the planning ofinjection operations and in the understanding of CO2plume evo-lution. A verification of the geometry of the reservoir and thestructural situation of its overburden is expected from a three-dimensional baseline seismic survey that was conducted in theautumn of 2005. Laboratory experiments under simulated in-situconditions were performed to evaluate the geophysical signatureof rocks saturated with CO2. The chemical composition of thegroundwater and the CO2flux in the soil were analyzed across theKetzin anticline, providing the baseline for a monitoring programduring and after injection of CO2, targeted at the detection ofpotential CO2leakage from the storage reservoir.

  • Peptidergic clock neurons in Drosophila : ion transport peptide and short neuropeptide F in subsets of dorsal and ventral lateral neurons

    • About 150 clock neurons are clustered in different groups in the brain of Drosophila. Among these clock neurons, some pigment-dispersing factor (PDF)-positive and PDF-negative lateral neurons (LNs) are principal oscillators responsible for bouts of activity in the morning and evening, respectively. The full complement of neurotransmitters in these morning and evening oscillators is not known. By using a screen for candidate neuromediators in clock neurons, we discovered ion transport peptide (ITP) and short neuropeptide F (sNPF) as novel neuropeptides in subpopulations of dorsal (LN(d)s) and ventral (s-LN(v)s) LNs. Among the six LN(d)s, ITP was found in one that coexpresses long neuropeptide F (NPF) and cryptochrome. We detected sNPF in two LN(d)s that also express cryptochrome; these cells are distinct from three LN(d)s expressing NPF. Thus, we have identified neuropeptides in five of the six LN(d)s. The three LN(d)s expressing cryptochrome, with either ITP or sNPF, are the only ones with additional projections to the accessory medulla. Among the five s-LN(v)s in the adult brain, ITP was detected in the fifth neuron that is devoid of PDF and sNPF in the four neurons that also express PDF. By using a choline acetyltransferase (Cha) Gal4, we detected Cha expression in the two sNPF producing LN(d)s and in the fifth s-LN(v). In the larval brain, two of the four PDF-producing s-LN(v)s coexpress sNPF. Our findings emphasize that the LN(d)s are heterogeneous both anatomically and with respect to content of neuropeptides, cryptochrome, and other markers and suggest diverse functions of these neurons.

  • A novel sporadic mutation G14739A of the mitochondrial tRNA(Glu) in a girl with exercise intolerance

    • We describe a 7-year-old girl who presented with loss of appetite, weakness and excercise intolerance. Enzyme investigation of the respiratory chain in muscle tissue revealed a combined complex I, III and IV deficiency. A novel heteroplasmic G-->A exchange at nucleotide position 14739 was found in the MTTE gene of the tRNA glutamic acid. The mutation load in muscle was 72%, urine sediment 38%, blood 31% and fibroblasts 29% and it correlated with COX-negative fibres. Our patient presented with a predominantly myopathic phenotype. The G14739A mutation is the third reported in the mitochondrial tRNA glutamic acid gene, and it occurred in a sporadic case.
