    • Plants mediated synthesis of gold nanoparticles (AuNPs) containing desired characteristics for their suitable potential applications has been a challenging task, which is causing a major hindrance towards its commercialization. Therefore, herein phytosynthesis of AuNPs with required size and morphology has been achieved through manipulating the reaction conditions including reaction temperature and volume of Elaeis guineensis leaves (EGL) extract. Furthermore, photocatalytic potential of EGL mediated AuNPs having different size and shape has also been explored for the removal of methylene blue (MB) under visible light irradiation. The reaction temperature and volume of EGL strongly influenced the size and morphology of AuNPs, which are directly associated with the photocatalytic activities. The experimental results revealed that predominantly spherical and ultra-smaller size AuNPs with particle size of 16.26 ± 5.84 nm, formed at 70 °C showed the highest removal efficiency up to 92.55 % in 60 min. This highest photocatalytic activity of AuNPs could be attributed to the availability of higher number of low-coordinated gold (Au) atoms in the MB aqueous solution, which might have boosted the adsorption of the MB on the surface of particles and accelerated the degradation phenomenon. The proposed photocatalytic degradation mechanism of AuNPs for MB was also explained. The highly photoactive EGL mediated AuNPs with controllable morphology and size could be an advance step in future in chemical and biomedical applications.

  • Machine Learning Methods Improve Prognostication, Identify Clinically Distinct Phenotypes, and Detect Heterogeneity in Response to Therapy in a Large Cohort of Heart Failure Patients

    • Background-Whereas heart failure (HF) is a complex clinical syndrome, conventional approaches to its management have treated it as a singular disease, leading to inadequate patient care and inefficient clinical trials. We hypothesized that applying advanced analytics to a large cohort of HF patients would improve prognostication of outcomes, identify distinct patient phenotypes, and detect heterogeneity in treatment response. Methods and Results-The Swedish Heart Failure Registry is a nationwide registry collecting detailed demographic, clinical, laboratory, and medication data and linked to databases with outcome information. We applied random forest modeling to identify predictors of 1-year survival. Cluster analysis was performed and validated using serial bootstrapping. Association between clusters and survival was assessed with Cox proportional hazards modeling and interaction testing was performed to assess for heterogeneity in response to HF pharmacotherapy across propensity-matched clusters. Our study included 44 886 HF patients enrolled in the Swedish Heart Failure Registry between 2000 and 2012. Random forest modeling demonstrated excellent calibration and discrimination for survival (C-statistic=0.83) whereas left ventricular ejection fraction did not (C-statistic=0.52): there were no meaningful differences per strata of left ventricular ejection fraction (1-year survival: 80%, 81%, 83%, and 84%). Cluster analysis using the 8 highest predictive variables identified 4 clinically relevant subgroups of HF with marked differences in 1-year survival. There were significant interactions between propensity-matched clusters (across age, sex, and left ventricular ejection fraction and the following medications: diuretics, angiotensin-converting enzyme inhibitors, )i-blockers, and nitrates, Pamp;lt;0.001, all). Conclusions-Machine learning algorithms accurately predicted outcomes in a large data set of HF patients. Cluster analysis identified 4 distinct phenotypes that differed significantly in outcomes and in response to therapeutics. Use of these novel analytic approaches has the potential to enhance effectiveness of current therapies and transform future HF clinical trials.

    • Baseline data from adult speakers are presented for time-domain parameters of EGG waveforms. 26 vocally healthy adults (13 males and 13 females) were recruited for the study. Four dependent variables were computed: mean contact quotient, mean peak-rate-of change in the contact area, index of contacting, and the audio crest factor. Small regions around the speech range distribution modes on the fo/SPL plane were used to define means and gradients. Males and females differed considerably in the audio crest factor of their speaking voice, and somewhat in their EGG contact quotient when measured at the mode point of the individual speech range profile. In males, contacting tended to increase somewhat with fo and SPL around the mode point, while in females it tended to decrease.

  • Community assessment to advance computational prediction of cancer drug combinations in a pharmacogenomic screen

    • The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca’s large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate computational strategies for predicting synergistic drug pairs and biomarkers. 160 teams participated to provide a comprehensive methodological development and benchmarking. Winning methods incorporate prior knowledge of drug-target interactions. Synergy is predicted with an accuracy matching biological replicates for >60% of combinations. However, 20% of drug combinations are poorly predicted by all methods. Genomic rationale for synergy predictions are identified, including ADAM17 inhibitor antagonism when combined with PIK3CB/D inhibition contrasting to synergy when combined with other PI3K-pathway inhibitors in PIK3CA mutant cells.

  • Diversity of Dust Properties in External Galaxies Confirmed by Polarization Signals from Type II Supernovae

    • Investigating interstellar (IS) dust properties in external galaxies is important not only to infer the intrinsic properties of astronomical objects but also to understand the star/planet formation in the galaxies. From the non–Milky Way–like extinction and interstellar polarization (ISP) observed in reddened Type Ia supernovae (SNe), it has been suggested that their host galaxies contain dust grains whose properties are substantially different from the Milky Way (MW) dust. It is important to investigate the universality of such non-MW-like dust in the universe. Here we report spectropolarimetry of two highly extinguished Type II SNe (SN 2022aau and SN 2022ame). SN 2022aau shows a polarization maximum at a shorter wavelength than MW stars, which is also observed in some Type Ia SNe. This is clear evidence for the existence of non-MW-like dust in its host galaxy (i.e., NGC 1672). This fact implies that such non-MW-like dust might be more common in some environments than expected, and thus it might affect the picture of the star/planet formation. On the other hand, SN 2022ame shows MW-like ISP, implying the presence of MW-like dust in its host galaxy (i.e., NGC 1255). Our findings confirm that dust properties of galaxies are diverse, either locally or globally. The present work demonstrates that further investigation of IS dust properties in external galaxies using polarimetry of highly reddened SNe is promising, providing a great opportunity to study the universality of such non-MW-like dust grains in the universe.

    • This paper outlines the development of a comparative research framework in subject didactics and applies this in the process of analysing the transformations from academic disciplines across different school subjects. The theoretical framework builds on the concepts of ‘powerful knowledge’ and ‘transformation’ and ‘epistemic quality’ within which transformation processes from the classroom to the societal level are considered as ‘trajectories of powerful knowledge and epistemic quality’. The framework is used to analyse the findings from recent empirical studies across school subjects that have been reported on in publications arising from the Knowledge and Quality across School Subjects and Teacher Education (KOSS) network. 1 The paper then focuses on analysing the transformations from disciplines across school subjects, given that the first boundary in defining powerful knowledge concerns knowledge that is specialized in both how it is produced and transmitted. To analyse this boundary, the findings from the empirical studies are grouped into broad subject categories. These are then compared with the corresponding disciplines by using the widely cited Biglan classification scheme of academic disciplines in higher education. Finally, we consider the implications for curriculum planning and teacher education policy and reflect on the concept of subject-specific educational content knowledge (SSECK).
