    • The isotopic and chemical signatures for ice-age and Holocene ice from Summit, Greenland, and Penny Ice Cap, Baffin Island, Canada, are compared. The usual pattern of low δ180, high Ca2+ and high Cl- is presented in the Summit records, but Penny Ice Cap has lower than present Cl- in its ice-age ice. A simple extension of the Hansson model (Hansson, 1994) is developed and used to simulate these signatures. The low ice­ age Cl- from Penny Ice Cap is explained by having the ice-age ice originating many thou­sands of km inland near the centre of the Laurentide ice sheet and much further from the marine sources. Summit's flowlines all start close to the present site. The Penny Ice Cap early-Holocene δ180's had to be corrected to offset the Laurentide meltwater distortion.The analysis suggests that presently the Summit and Penny Ice Cap marine impurity ori­ ginates about 500 km away, and that presently Penny Ice Cap receives a significant amountoflocal continental impurity.

    • Disturbances in fatty acid metabolism are involved in the etiology of insulin resistance and the related dyslipidemia, hypertension, and procoagulant state. The fatty acid transport proteins (FATPs) are implicated in facilitated cellular uptake of nonesterified fatty acids (NEFAs), thus potentially regulating NEFA concentrations and metabolism. The aim of this study was to investigate polymorphic loci in the FATP4 gene with respect to associations with fasting and postprandial lipid and lipoprotein variables and markers of insulin resistance in 608 healthy, middle-aged Swedish men and to evaluate possible mechanisms behind any associations observed. Heterozygotes for a Gly209Ser polymorphism (Ser allele frequency 0.05) had significantly lower body mass index and, correcting for body mass index, significantly lower triglyceride concentrations, systolic blood pressure, insulin concentrations, and homeostasis model assessment index compared with common homozygotes. A three-dimensional model of the FATP4 protein based on structural and functional similarity with adenylate-forming enzymes revealed that the variable residue 209 is exposed in a region potentially involved in protein-protein interactions. Furthermore, the model indicated functional regions with respect to NEFA transport and acyl-coenzyme A synthase activity and membrane association. These findings propose FATP4 as a candidate gene for the insulin resistance syndrome and provide a structural basis for understanding FATP function in NEFA transport and metabolism.

    • An >11,550-yr-long record of atmospheric dust deposition was developed from an ice core (P95) drilled through the Penny ice cap, Baffin Island. The P95 record documents environmental changes that affected the production and transport of dust in the eastern Canadian Arctic during the late Pleistocene and Holocene. Dust deposition on the Penny ice cap was greatest in late-glacial time when the climate was dry and windy and comparatively low in the Holocene. Microparticles deposited during late-glacial time are finer than in Greenland cores, suggesting distinct dust sources and transport trajectories to each region. Dust deposition at the P95 site increased after ca. 7800 yr ago as the Penny ice cap receded and distance from local dust sources was reduced. Deflation of newly exposed marine sediments on southwestern Baffin Island may have contributed to this dust increase. The P95 and GISP2 (Greenland) dust records show diverging trends in the middle to late Holocene, reflecting the growing influence of regional environmental conditions (e.g., dust source area, snow cover extent) on atmospheric dust deposition. This study further demonstrates how valuable records of regional-scale paleoenvironmental changes can be developed from small circum-Arctic ice caps, even those affected by considerable melt.

A brief summary of related papers published in the literature within the areas of skin irritant and allergy patch testing, microdialysis and skin tumour circulation is presented, as well as early applications within other fields such as diabetology, wound healing and microvascular research.

  • Chromosome 3 linked frontotemporal dementia (FTD-3)

    • Background: The authors have identified and studied a large kindred in which frontotemporal dementia (FTD) is inherited as an autosomal dominant trait. The trait has been mapped to the pericentromeric region of chromosome 3. Methods: The authors report on the clinical, neuroimaging, neuropsychological, and pathologic features in this unique pedigree collected during 17 years of study. Results: Twenty-two individuals in three generations have been affected; the age at onset varies between 46 and 65 years. The disease presents with a predominantly frontal lobe syndrome but there is also evidence for temporal and dominant parietal lobe dysfunction. Late in the illness individuals develop a florid motor syndrome with pyramidal and extrapyramidal features. Structural imaging reveals generalized cerebral atrophy; H-2 O-15-PET scanning in two individuals relatively early and late in the disease shows a striking global reduction in cerebral blood flow affecting all lobes. On macroscopic pathologic examination, there is generalized cerebral atrophy affecting the frontal lobes preferentially. Microscopically, there is neuronal loss and gliosis without specific histopathologic features. Conclusions: FTD-3 shares clinical and pathologic features with other forms of FTD and fulfills international consensus criteria for FTD. There is involvement of the parietal lobes clinically, radiologically, and pathologically in FTD-3 in contrast to some forms of FTD. This more diffuse involvement of the cerebral cortex leads to a distinctive, global pattern of reduced blood flow on PET scanning.
