    • Stable junctions between a single carbon chain and two single-wall carbon nanotubes were produced via coalescence of functionalized fullerenes filled into a single-wall carbon nanotube and directly imaged by in situ transmission electron microscopy. First principles quantum chemical calculations support the observed stability of such molecular junctions. They also show that short carbon chains bound to other carbon structures are cumulenes and stable semiconductors due to Peierls-like distortion. Junctions like this can be regarded as archetypical building blocks for all-carbon molecular electronics.

    • This paper presents sedimentological, geochemical, and biological data from Lake Prespa (Albania/Former Yugoslav Republic of Macedonia/Greece). The 320 cm core sequence (Co1215) covers the last 17 ka calBP and reveals significant change in climate and environmental conditions on a local and regional scale. The sediment record suggests typical stadial conditions from 17.1 to 15.7 ka calBP, documented through low lake productivity, well-mixed conditions, and cold-resistant steppe catchment vegetation. Warming is indicated from 15.7 ka calBP with slightly increased in-lake productivity, gradual expansion of trees, and decreasing erosion through disappearance of local ice caps. Between 14.5 and 11.5 ka calBP relatively stable hydrological conditions are documented. The maximum in tree taxa percentages during the Bolling/Allerod interstadial (14.5-13.2 ka calBP) indicates increased temperatures and moisture availability, whereas the increase of cold-resistant open steppe vegetation taxa percentages during the Younger Dryas (13.2-11.5 ka calBP) is coupled with distinct colder and drier conditions. The Holocene sequence from 11.5 ka calBP indicates ice-free winters, stratification of the water column, a relatively high lake trophic level and dense vegetation cover over the catchment. A strong climate related impact on the limnology and physical parameters in Lake Prespa is documented around 8.2 ka through a significant decrease in productivity, enhanced mixing, strong decomposition and soil erosion, and a coeval expansion of herbs implying cool and dry climate conditions. Intensive human activity in the catchment is indicated from around 1.9 ka calBP. This multiproxy approach improves our understanding of short- and long-term climate fluctuations in this area and their impact on catchment dynamics, limnology, hydrology, and vegetation.   • A community-based geological reconstruction of Antarctic Ice Sheet deglaciation since the Last Glacial Maximum

    • A robust understanding of Antarctic Ice Sheet deglacial history since the Last Glacial Maximum is important in order to constrain ice sheet and glacial-isostatic adjustment models, and to explore the forcing mechanisms responsible for ice sheet retreat. Such understanding can be derived from a broad range of geological and glaciological datasets and recent decades have seen an upsurge in such data gathering around the continent and Sub-Antarctic islands. Here, we report a new synthesis of those datasets, based on an accompanying series of reviews of the geological data, organised by sector. We present a series of timeslice maps for 20 ka, 15 ka, 10 ka and 5 ka, including grounding line position and ice sheet thickness changes, along with a clear assessment of levels of confidence. The reconstruction shows that the Antarctic Ice sheet did not everywhere reach the continental shelf edge at its maximum, that initial retreat was asynchronous, and that the spatial pattern of deglaciation was highly variable, particularly on the inner shelf. The deglacial reconstruction is consistent with a moderate overall excess ice volume and with a relatively small Antarctic contribution to meltwater pulse la. We discuss key areas of uncertainty both around the continent and by time interval, and we highlight potential priorities for future work. The synthesis is intended to be a resource for the modelling and glacial geological community.

  • Earth system governance: a research framework

    • The Earth System Science Partnership, which unites all major global change research programmes, declared in 2001 an urgent need to develop "strategies for Earth System management". Yet what such strategies might be, how they could be developed, and how effective, efficient and equitable such strategies would be, remains unspecified. It is apparent that the institutions, organizations and mechanisms by which humans currently govern their relationship with the natural environment and global biochemical systems are not only insufficient-they are also poorly understood. This article presents the science programme of the Earth System Governance Project, a new 10-year global research effort endorsed by the International Human Dimensions Programme on Global Environmental Change (IHDP). It outlines the concept of earth system governance as a challenge for the social sciences, and it elaborates on the interlinked analytical problems and research questions of earth system governance as an object of study. These analytical problems concern the overall architecture of earth system governance, agency beyond the state and of the state, the adaptiveness of governance mechanisms and processes as well as their accountability and legitimacy, and modes of allocation and access in earth system governance. The article also outlines four crosscutting research themes that are crucial for the study of each analytical problem as well as for the integrated understanding of earth system governance: the role of power, knowledge, norms and scale.

  • A Comprehensive Investigation on Common Polymorphisms in the MDR1/ABCB1 Transporter Gene and Susceptibility to Colorectal Cancer

    • ATP Binding Cassette B1 (ABCB1) is a transporter with a broad substrate specificity involved in the elimination of several carcinogens from the gut. Several polymorphic variants within the ABCB1 gene have been reported as modulators of ABCB1-mediated transport. We investigated the impact of ABCB1 genetic variants on colorectal cancer (CRC) risk. A hybrid tagging/functional approach was performed to select 28 single nucleotide polymorphisms (SNPs) that were genotyped in 1,321 Czech subjects, 699 CRC cases and 622 controls. In addition, six potentially functional SNPs were genotyped in 3,662 German subjects, 1,809 cases and 1,853 controls from the DACHS study. We found that three functional SNPs (rs1202168, rs1045642 and rs868755) were associated with CRC risk in the German population. Carriers of the rs1202168_T and rs868755_T alleles had an increased risk for CRC (P-trend = 0.016 and 0.029, respectively), while individuals bearing the rs1045642_C allele showed a decreased risk of CRC (P-trend = 0.022). We sought to replicate the most significant results in an independent case-control study of 3,803 subjects, 2,169 cases and 1,634 controls carried out in the North of Germany. None of the SNPs tested were significantly associated with CRC risk in the replication study. In conclusion, in this study of about 8,800 individuals we show that ABCB1 gene polymorphisms play at best a minor role in the susceptibility to CRC.

  • Structure and Dynamics of Soft Repulsive Colloidal Suspensions in the Vicinity of the Glass Transition

    • We use a combination of different scattering techniques and rheology to highlight the link between structure and dynamics of dense aqueous suspensions of soft repulsive colloids in the vicinity of a glass transition. Three different latex formulations with an increasing amount of the hydrophilic component resulting in either purely electrostatically or electrosterically stabilized suspensions are investigated. From the analysis of the static structure factor measured by small-angle X-ray scattering, we derive an effective volume fraction that includes contributions from interparticle interactions. We further investigate the dynamics of the suspensions using 3D cross-correlation dynamic light scattering (3DDLS) and rheology. We analyze the data using an effective hard sphere model and in particular compare the linear viscoelasticity and flow behavior to the predictions of mode coupling theory, which accounts for a purely kinetic glass transition determined by the equilibrium structure factor. We demonstrate that seemingly very different colloidal systems exhibit the same generic behavior when the effects from interparticle interactions are incorporated using an effective volume fraction description.

    • Prostate cancer is a common malignancy in men and the worldwide burden of this disease is rising. Lifestyle modifications such as smoking cessation, exercise, and weight control offer opportunities to reduce the risk of developing prostate cancer. Early detection of prostate cancer by prostate-specific antigen (PSA) screening is controversial, but changes in the PSA threshold, frequency of screening, and the use of other biomarkers have the potential to minimise the overdiagnosis associated with PSA screening. Several new biomarkers for individuals with raised PSA concentrations or those diagnosed with prostate cancer are likely to identify individuals who can be spared aggressive treatment. Several pharmacological agents such as 5α-reductase inhibitors and aspirin could prevent development of prostate cancer. In this Review, we discuss the present evidence and research questions regarding prevention, early detection of prostate cancer, and management of men either at high risk of prostate cancer or diagnosed with low-grade prostate cancer.

    • A whole-sediment toxicity test with Myriophyllum aquaticum has been developed by the German Federal Institute of Hydrology and standardized within the International Organization for Standardization (ISO; ISO 16191). An international ring-test was performed to evaluate the precision of the test method. Four sediments (artificial, natural) were tested. Test duration was 10 d, and test endpoint was inhibition of growth rate (r) based on fresh weight data. Eighteen of 21 laboratories met the validity criterion of r >= 0.09 d(-1) in the control. Results from 4 tests that did not conform to test-performance criteria were excluded from statistical evaluation. The inter-laboratory variability of growth rates (20.6%-25.0%) and inhibition (26.6%-39.9%) was comparable with the variability of other standardized bioassays. The mean test-internal variability of the controls was low (7% [control], 9.7% [solvent control]), yielding a high discriminatory power of the given test design (median minimum detectable differences [MDD] 13% to 15%). To ensure these MDDs, an additional validity criterion of CV <= 15% of the growth rate in the controls was recommended. As a positive control, 90 mg 3,5-dichlorophenol/kg sediment dry mass was tested. The range of the expected growth inhibition was proposed to be 35 +/- 15%. The ring test results demonstrated the reliability of the ISO 16191 toxicity test and its suitability as a tool to assess the toxicity of sediment and dredged material.     • Maar lake Laguna Potrok Aike is located north of the Strait of Magellan (south-eastern Patagonia). Seismic reflection profiles revealed a highly dynamic palaeoclimate history. Dunes were identified in the eastern part of the lake at approximately 30 to 80 m below the lake floor, overlying older lacustrine strata, and suggest that the region experienced dry conditions probably combined with strong westerly winds. It is quite likely that this can be linked to a major dust event recorded in the Antarctic ice cores during Marine Isotope Stage 4. The dunes are overlain by a series of palaeo-shorelines indicating a stepwise water-level evolution of a new lake established after this dry period, and thus a change towards wetter conditions. After the initial, rapid and stepwise lake-level rise, the basin became deeper and wider, and sediments deposited on the lake shoulder at approximately 33 m below present-day lake level point towards a long period of lake-level highstand between roughly 53.5 ka cal. bp and 30 ka cal. bp with a maximum lake level some 200 m higher than the desiccation horizon. This highstand was then followed by a regressional phase of uncertain age, although it must have happened some time between approximately 30 ka cal. bp and 6750 yrs cal. bp. Dryer conditions during the Mid-Holocene are evidenced by a dropping lake level, resulting in a basin-wide erosional unconformity on the lake shoulder. A second stepwise transgression between ca 5.8 to 5.4 ka cal. bp and ca 4.7 to 4 ka cal. bp with palaeo-shorelines deposited on the lake shoulder unconformity again indicates a change towards wetter conditions.

  • Design of a general-purpose European compound screening library for EU-OPENSCREEN

    • This work describes a collaborative effort to define and apply a protocol for the rational selection of a general-purpose screening library, to be used by the screening platforms affiliated with the EU-OPENSCREEN initiative. It is designed as a standard source of compounds for primary screening against novel biological targets, at the request of research partners. Given the general nature of the potential applications of this compound collection, the focus of the selection strategy lies on ensuring chemical stability, absence of reactive compounds, screening-compliant physicochemical properties, loose compliance to drug-likeness criteria (as drug design is a major, but not exclusive application), and maximal diversity/coverage of chemical space, aimed at providing hits for a wide spectrum of drugable targets. Finally, practical availability/cost issues cannot be avoided. The main goal of this publication is to inform potential future users of this library about its conception, sources, and characteristics. The outline of the selection procedure, notably of the filtering rules designed by a large committee of European medicinal chemists and chemoinformaticians, may be of general methodological interest for the screening/medicinal chemistry community. The selection task of 200K molecules out of a pre-filtered set of 1.4M candidates was shared by five independent European research groups, each picking a subset of 40K compounds according to their own in-house methodology and expertise. An in-depth analysis of chemical space coverage of the library serves not only to characterize the collection, but also to compare the various chemoinformatics-driven selection procedures of maximal diversity sets. Compound selections contributed by various participating groups were mapped onto general-purpose self-organizing maps (SOMs) built on the basis of marketed drugs and bioactive reference molecules. In this way, the occupancy of chemical space by the EU-OPENSCREEN library could be directly compared with distributions of known bioactives of various classes. This mapping highlights the relevance of the selection and shows how the consensus reached by merging the five different 40K selections contributes to achieve this relevance. The approach also allows one to readily identify subsets of target-or target-class-oriented compounds from the EU-OPENSCREEN library to suit the needs of the diverse range of potential users. The final EU-OPENSCREEN library, assembled by merging five independent selections of 40K compounds from various expert groups, represents an excellent example of a Europe-wide collaborative effort toward the common objective of building best-in-class European open screening platforms.
