  • Diabetic osteopathy : a study in the rat

    • The present study on non-obese Goto-Kakizaki (GK) rats with type-2 diabetes and neuropathy was an attempt to describe and define pertinent features of diabetic osteopathy. Altogether, the study included 33 GK rats aged 12 and 20 months, and 36 age-matched Wistar rats as controls. All underwent test of glucose tolerance and nerve (sciatic) conduction velocity (NCV) showing that the diabetic rats had significantly higher blood glucose levels and lower NCV confirming the presence of diabetes and neuropathy. Skeletal features: Radiologic analysis of bone entailed X-ray, Dual Energy X-ray Absorptiometry (DEXA) and peripheral Quantitative Computed Tomography (pQCT). In diabetic rats, the length of humerus and height of vertebrae was reduced by 8%. The long bones exhibited endosteal erosion of the diaphyses up to 18% and periosteal expansion up to 8%. The vertebrae and metaphyses of long bones showed a decrease up to 24% in areal bone mineral density (BMD), whereas no decrease was seen in the diaphyses. Cross-sectional measurements by pQCT showed a decrease in volumetric BMD ranging from 33 to 62%, which exclusively pertained to trabecular bone (vertebrae, metaphyses), whereas volumetric BMD of the cortical bone of diaphyses was only marginally affected. The results indicate that juxta-articular bone in diabetes is substantially weaker, whereas diaphyseal cortical bone may be even stronger. Over all, the observations suggest that the diabetic skeleton is characterized by regional changes, which cannot be explained by systemic factors like calcium regulating hormones. Local bone turn-over is regulated by complex mechanisms involving cytokines, prostaglandins, growth factors and, also neuropeptides. Further analysis focused on the insulin-like growth factor (IGF) system and neuronal mediators in bone. IGF system: Immunoassays of IGF-I were done on serum, ankle samples and cortical preparations. In addition, the inhibitory IGF-I binding proteins, IGFBP-1 and -4 were analysed in serum. In diabetic rats, serum IGF-I was reduced by 18%, while IGFBP-1 and IGFBP-4 were increased by 89 and 20%, respectively. This complies with the lower BMD in the diabetic rats. In cortical bone, IGF-I was reduced by 38%, whereas no change was seen in ankles. The loss of IGF-I in cortical bone represents a novel finding. Given the cortical expansion observed in diabetic rats, the opposite was expected. Conceivably, loss of IGF-I results in endosteal erosion, which is compensated by periosteal expansion. Neuropeptides: The analyses focused on two sensory mediators, i.e. substance P (SP) and calcitonin gene- related peptide (CGRP), and one autonomic, i.e. neuropeptide Y (NPY). Immunohistochemistry was applied to ankles and tibial diaphyses, whereas radioimmunoassay (RIA) was used for separate preparations of periosteum, cortex and bone marrow from femur and tibia, whole ankles, dorsal root ganglia (DRG) and lumbar spinal cord. The morphological analysis showed SP, CGRP and NPY positive nerve fibers in bone and joints, which mostly were blood vessel related, although free terminals were also seen. In addition, NPY-positive hematopoietic cells were observed in the bone marrow. RIA revealed a significant decrease of CGRP, albeit not of SP, in DRG (-26%) and spinal cord (-29%) in the diabetic rats. As for bone, only NPY was significantly reduced, most evidently in bone marrow (-66%), but also in cortical bone (-36%) and ankles (-29%). Given the bone anabolic effects of CGRP and NPY, loss of these neuropeptides may prove, at least partly, to underlie the trabecular osteopenia and endocortical erosion observed in diabetic rats. Conclusion: The skeleton of diabetic rats with type-2 diabetes and neuropathy is characterized by regional changes of size, form, mineral content and density and concomitantly with regional abnormalities of the IGF- system and neuropeptides suggesting that also local factors beyond systemic play an important role in the development of diabetic osteopathy.

  • A neurolinguistic study of South Swedish word accents : Electrical brain potentials in nouns and verbs

    • The brain response to words with correct and incorrect word accent–suffix combinations in South Swedish was investigated using electroencephalography (EEG). Accent 1 yielded an increased brain response (‘preactivation negativity’) that has previously been interpreted as reflecting preactivation of suffixes. Preactivation is greater for accent 1 due to its association with a limited set of suffixes, whereas accent 2 is default for compound words. The tonal realization of the word accent opposition in South Swedish is practically the mirror image of that in Central Swedish, where a similar preactivation negativity has been found. Therefore, the brain response is unlikely to result from a difference in acoustic features between the word accents. Invalidly cued suffixes yielded brain response pattern showing increased processing load of the unexpected suffix (negative electric potential) followed by its reprocessing (positivity ‘P600’).

  • Customer Preferences for Delivery Service Attributes in Attended Home Delivery

    • Retailers face increasing competitive pressure to determine how best to deliver products purchased online to the end customer. Grocery retailers often require attended home delivery where the customer must be present to receive the delivery. For attended home delivery to function, the retailer and customer must agree on a delivery time slot that works for both parties. Using online data from a grocery retailer, we observe customer preferences for three delivery service attributes associated with each time slot: speed, precision, and timing. We define speed as the expected time between the placement of an order and its delivery, precision as the duration of the offered time slot, and timing as the availability of choices across times of the day and days of the week. We show that customers not only value speed as an attribute of delivery service but that precision and timing are also key drivers of the customer's time slot selection process. We also observe substantial customer heterogeneity in the willingness of customers to pay for time slots. Customers that differ in their loyalty to the retailer, basket value, basket size, and basket composition exhibit distinct differences in their willingness to pay. We show that retailers with the capability to tailor their time slot offerings to specific customer segments have the potential to generate approximately 9% more shipping revenue than those who cannot. Our findings inform practitioners seeking to design competitive fulfillment strategies and academics customer behavior in the attended home context.

    • One of the challenges for next generation DNA sequencing is to have a robust, stable, and reproducible nanodevice. In this work, we propose how to improve the sensing of DNA nucleobase using functionalized graphene nanogap as a solid state device. Two types of edge functionalization, namely, either hydrogen or nitrogen, were considered. We showed that, independent of species involved in the edge passivation, the highest-to-lowest order of the nucleobase transmissions is not altered, but the intensity is affected by several orders of magnitude. Our results show that nitrogen edge tends to p-dope graphene, and most importantly, it contributes with resonance states close to the Fermi level, which can be associated with the increased conductance. Finally, the translocation process of nucleobases passing through the nanogap was also investigated by varying their position from a certain height (from +3 to -3 angstrom) with respect to the graphene sheet to show that nitrogen-terminated sheets have enhanced sensitivity, as moving the nucleobase by approximately 1 angstrom reduces the conductance by up to 3 orders of magnitude.

  • Ubiquitous broad-line emission and the relation between ionized gas outflows and Lyman continuum escape in Green Pea galaxies

    • We report observational evidence of highly turbulent ionized gas kinematics in a sample of 20 Lyman continuum (LyC) emitters (LCEs) at low redshift (z ∼ 0.3). Detailed Gaussian modeling of optical emission line profiles in high-dispersion spectra consistently shows that both bright recombination and collisionally excited lines can be fitted as one or two narrow components with intrinsic velocity dispersion of σ ∼ 40 − 100 km s−1, in addition to a broader component with σ ∼ 100 − 300 km s−1, which contributes up to ∼40% of the total flux and is preferentially blueshifted from the systemic velocity. We interpret the narrow emission as highly ionized gas close to the young massive star clusters and the broader emission as a signpost of unresolved ionized outflows, resulting from massive stars and supernova feedback. We find a significant correlation between the width of the broad emission and the LyC escape fraction, with strong LCEs exhibiting more complex and broader line profiles than galaxies with weaker or undetected LyC emission. We provide new observational evidence supporting predictions from models and simulations; our findings suggest that gas turbulence and outflows resulting from strong radiative and mechanical feedback play a key role in clearing channels through which LyC photons escape from galaxies. We propose that the detection of blueshifted broad emission in the nebular lines of compact extreme emission-line galaxies can provide a new indirect diagnostic of Lyman photon escape, which could be useful to identify potential LyC leakers in the epoch of reionization with the JWST.

    • Accurate estimation of the state-of-health (SOH) is vital to the life management of lithium-ion batteries (LIBs). This paper proposes a fusion-type SOH estimation method by combining the model-based feature extraction and data-based state estimate. Particularly, a novel model-based voltage construction method is proposed to eliminate the unfavorable numerical condition and reshape the disturbance-free incremental capacity (IC) curves. Leveraging the modified IC curves, a set of informative features-of-interest are extracted and evaluated, while eventually several cautiously-selected ones are used to estimate the SOH of LIB accurately. Furthermore, the impact of model order on the estimation performance is scrutinized, to give insights into the parameterization in practical applications. Long-term cycling tests on different types of LIB cells are used for evaluation. The proposed method is validated with a good robustness to the cell inconsistency, temperature uncertainty, noise corruption, and a satisfied generality to different battery chemistries.
