  • Regulatory and Functional Connection of Microphthalmia-Associated Transcription Factor and Anti-Metastatic Pigment Epithelium Derived Factor in Melanoma

    • Pigment epithelium-derived factor (PEDF), a member of the serine protease inhibitor superfamily, has potent anti-metastatic effects in cutaneous melanoma through its direct actions on endothelial and melanoma cells. Here we show that PEDF expression positively correlates with microphthalmia-associated transcription factor ( MITF) in melanoma cell lines and human samples. High PEDF and MITF expression is characteristic of low aggressive melanomas classified according to molecular and pathological criteria, whereas both factors are decreased in senescent melanocytes and naevi. Importantly, MITF silencing down-regulates PEDF expression in melanoma cell lines and primary melanocytes, suggesting that the correlation in the expression reflects a causal relationship. In agreement, analysis of Chromatin immunoprecipitation coupled to high throughput sequencing (ChIP-seq) data sets revealed three MITF binding regions within the first intron of SERPINF1, and reporter assays demonstrated that the binding of MITF to these regions is sufficient to drive transcription. Finally, we demonstrate that exogenous PEDF expression efficiently halts in vitro migration and invasion, as well as in vivo dissemination of melanoma cells induced by MITF silencing. In summary, these results identify PEDF as a novel transcriptional target of MITF and support a relevant functional role for the MITF-PEDF axis in the biology of melanoma.

  • Measurement of prompt J/psi and beauty hadron production cross sections at mid-rapidity in pp collisions at root s=7 TeV

    • The ALICE experiment at the LHC has studied J/psi production at mid-rapidity in pp collisions at root s = 7 TeV through its electron pair decay on a data sample corresponding to an integrated luminosity L-int = 5.6 nb(-1). The fraction of J/psi from the decay of long-lived beauty hadrons was determined for J/psi candidates with transverse momentum p(t) > 1,3 GeV/c and rapidity vertical bar y vertical bar < 0.9. The cross section for prompt J/psi mesons, i.e. directly produced J/psi and prompt decays of heavier charmonium states such as the psi(2S) and chi(c) resonances, is sigma(prompt J/psi) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 8.3 +/- 0.8(stat.) +/- 1.1 (syst.)(-1.4)(+1.5) (syst. pol.) mu b. The cross section for the production of b-hadrons decaying to J/psi with p(t) > 1.3 GeV/c and vertical bar y vertical bar < 0.9 is a sigma(J/psi <- hB) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 1.46 +/- 0.38 (stat.)(-0.32)(+0.26) (syst.) mu b. The results are compared to QCD model predictions. The shape of the p(t) and y distributions of b-quarks predicted by perturbative QCD model calculations are used to extrapolate the measured cross section to derive the b (b) over bar pair total cross section and d sigma/dy at mid-rapidity.

  • Underlying Event measurements in pp collisions at root s=0.9 and 7 TeV with the ALICE experiment at the LHC

    • We present measurements of Underlying Event observables in pp collisions at root s = 0 : 9 and 7 TeV. The analysis is performed as a function of the highest charged-particle transverse momentum p(T),L-T in the event. Different regions are defined with respect to the azimuthal direction of the leading (highest transverse momentum) track: Toward, Transverse and Away. The Toward and Away regions collect the fragmentation products of the hardest partonic interaction. The Transverse region is expected to be most sensitive to the Underlying Event activity. The study is performed with charged particles above three different p(T) thresholds: 0.15, 0.5 and 1.0 GeV/c. In the Transverse region we observe an increase in the multiplicity of a factor 2-3 between the lower and higher collision energies, depending on the track p(T) threshold considered. Data are compared to PYTHIA 6.4, PYTHIA 8.1 and PHOJET. On average, all models considered underestimate the multiplicity and summed p(T) in the Transverse region by about 10-30%.

  • Curiosity's rover environmental monitoring station : Overview of the first 100 sols

    • In the first 100 Martian solar days (sols) of the Mars Science Laboratory mission, the Rover Environmental Monitoring Station (REMS) measured the seasonally evolving diurnal cycles of ultraviolet radiation, atmospheric pressure, air temperature, ground temperature, relative humidity, and wind within Gale Crater on Mars. As an introduction to several REMS-based articles in this issue, we provide an overview of the design and performance of the REMS sensors and discuss our approach to mitigating some of the difficulties we encountered following landing, including the loss of one of the two wind sensors. We discuss the REMS data set in the context of other Mars Science Laboratory instruments and observations and describe how an enhanced observing strategy greatly increased the amount of REMS data returned in the first 100 sols, providing complete coverage of the diurnal cycle every 4 to 6 sols. Finally, we provide a brief overview of key science results from the first 100 sols. We found Gale to be very dry, never reaching saturation relative humidities, subject to larger diurnal surface pressure variations than seen by any previous lander on Mars, air temperatures consistent with model predictions and abundant short timescale variability, and surface temperatures responsive to changes in surface properties and suggestive of subsurface layering.   • Fixed and Mobile Convergence: Needs and Solutions

    • The drivers of Fixed and Mobile Convergence (FMC) are discussed. A reference framework for FMC proposed by European project COMBO is then presented. Some use cases of FMC are described, showing the needs for mutualization and convergence of fixed and mobile broadband networks. Five network scenarios providing technical solutions to FMC use cases are proposed. They target an optimal and seamless quality of experience for the end user together with an optimized network infrastructure ensuring increased performance, flexibility, reduced cost and reduced energy consumption.

  • Smart Cities at the forefront of the future internet

    • Smart cities have been recently pointed out by M2M experts as an emerging market with enormous potential, which is expected to drive the digital economy forward in the coming years. However, most of the current city and urban developments are based on vertical ICT solutions leading to an unsustainable sea of systems and market islands. In this work we discuss how the recent vision of the Future Internet (FI), and its particular components, Internet of Things (IoT) and Internet of Services (IoS), can become building blocks to progress towards a unified urban-scale ICT platform transforming a Smart City into an open innovation platform. Moreover, we present some results of generic implementations based on the ITU-T’s Ubiquitous Sensor Network (USN) model. The referenced platform model fulfills basic principles of open, federated and trusted platforms (FOTs) at two different levels: the infrastructure level (IoT to support the complexity of heterogeneous sensors deployed in urban spaces), and at the service level (IoS as a suit of open and standardized enablers to facilitate the composition of interoperable smart city services). We also discuss the need of infrastructures at the European level for a realistic large-scale experimentally-driven research, and present main principles of the unique-in-the-world experimental test facility under development within the SmartSantander EU project.

    • Angular correlations between charged trigger and associated particles are measured by the ALICE detector in p-Pb collisions at a nucleon-nucleon centre-of-mass energy of 5.02 TeV for transverse momentum ranges within 0.5 < P-T,P-assoc < P-T,P-trig < 4 GeV/c. The correlations are measured over two units of pseudorapidity and full azimuthal angle in different intervals of event multiplicity, and expressed as associated yield per trigger particle. Two long-range ridge-like structures, one on the near side and one on the away side, are observed when the per-trigger yield obtained in low-multiplicity events is subtracted from the one in high-multiplicity events. The excess on the near-side is qualitatively similar to that recently reported by the CMS Collaboration, while the excess on the away-side is reported for the first time. The two-ridge structure projected onto azimuthal angle is quantified with the second and third Fourier coefficients as well as by near-side and away-side yields and widths. The yields on the near side and on the away side are equal within the uncertainties for all studied event multiplicity and p(T) bins, and the widths show no significant evolution with event multiplicity or p(T). These findings suggest that the near-side ridge is accompanied by an essentially identical away-side ridge. (c) 2013 CERN. Published by Elsevier B.V. All rights reserved.

  • Predicting seminal quality with artificial intelligence methods

    • Fertility rates have dramatically decreased in the last two decades, especially in men. It has been described that environmental factors, as well as life habits, may affect semen quality. Artificial intelligence techniques are now an emerging methodology as decision support systems in medicine. In this paper we compare three artificial intelligence techniques, decision trees, Multilayer Perceptron and Support Vector Machines, in order to evaluate their performance in the prediction of the seminal quality from the data of the environmental factors and lifestyle. To do that we collect data by a normalized questionnaire from young healthy volunteers and then, we use the results of a semen analysis to asses the accuracy in the prediction of the three classification methods mentioned above. The results show that Multilayer Perceptron and Support Vector Machines show the highest accuracy, with prediction accuracy values of 86% for some of the seminal parameters. In contrast decision trees provide a visual and illustrative approach that can compensate the slightly lower accuracy obtained. In conclusion artificial intelligence methods are a useful tool in order to predict the seminal profile of an individual from the environmental factors and life habits. From the studied methods, Multilayer Perceptron and Support Vector Machines are the most accurate in the prediction. Therefore these tools, together with the visual help that decision trees offer, are the suggested methods to be included in the evaluation of the infertile patient. (C) 2012 Elsevier Ltd.   • Geographic variation in body mass of first-year Reed Warblers Acrocephalus scirpaceus in Iberia

    • On their route to tropical Africa, European trans-Saharan migrants must cross two major geographical barriers, the Mediterranean Sea and the Sahara Desert, which necessitates the accumulation of large fuel loads. While northern Africa is the chief region where most migrants gain fuel for the Sahara crossing, Iberia is a target area to gain fuel before the sea crossing existing between Europe and Africa. Despite the large body of studies approach- ing the question of fuel accumulation before geographic barriers, it is still poorly known which factors apart from distance to a certain barrier shape the geographical pattern of fuel reserves. To investigate this question in detail we used data of first-year Reed Warblers from 12 localities within Iberia during the autumn migration period of 2009. We run linear models to analyze the effects of location in Iberia, date, and body size on body mass variation at each migratory flyway (eastern, central and western Iberia). Flight ranges from each site were also calculated. Our results showed that Reed Warblers in Iberia had the necessary fuel needed to arrive in northern Africa but not to tropical Africa. However, body mass patterns varied depending on the geographical region (eastern, central or west- ern Iberia). Date did not affect body mass in central and western Iberia, but in eastern Ibe- ria heavier birds tended to pass later. Thus, the factors shaping body mass of Reed War- blers in Iberia before the sea crossing to Africa seemed to be more complex than just the distance to this geographical barrier, with underlying stopover quality-associated factors possibly playing a relevant role.

  • A replication study confirms the association of TNFSF4 (OX40L) polymorphisms with systemic sclerosis in a large European cohort

    • Objectives The aim of this study was to confirm the influence of TNFSF4 polymorphisms on systemic sclerosis (SSc) susceptibility and phenotypic features. Methods A total of 8 European populations of Caucasian ancestry were included, comprising 3014 patients with SSc and 3125 healthy controls. Four genetic variants of TNFSF4 gene promoter (rs1234314, rs844644, rs844648 and rs12039904) were selected as genetic markers. Results A pooled analysis revealed the association of rs1234314 and rs12039904 polymorphisms with SSc (OR 1.15, 95% CI 1.02 to 1.31; OR 1.18, 95% CI 1.08 to 1.29, respectively). Significant association of the four tested variants with patients with limited cutaneous SSc (lcSSc) was revealed (rs1234314 OR 1.22, 95% CI 1.07 to 1.38; rs844644 OR 0.91, 95% CI 0.83 to 0.99; rs844648 OR 1.10, 95% CI 1.01 to 1.20 and rs12039904 OR 1.20, 95% CI 1.09 to 1.33). Association of rs1234314, rs844648 and rs12039904 minor alleles with patients positive for anti-centromere antibodies (ACA) remained significant (OR 1.23, 95% CI 1.10 to 1.37; OR 1.12, 95% CI 1.01 to 1.25; OR 1.22, 95% CI 1.07 to 1.38, respectively). Haplotype analysis confirmed a protective haplotype associated with SSc, lcSSc and ACA positive subgroups (OR 0.88, 95% CI 0.82 to 0.96; OR 0.88, 95% CI 0.80 to 0.96; OR 0.86, 95% CI 0.77 to 0.97, respectively) and revealed a new risk haplotype associated with the same groups of patients (OR 1.14, 95% CI 1.03 to 1.26; OR 1.20, 95% CI 1.08 to 1.35; OR 1.23, 95% CI 1.07 to 1.42, respectively). Conclusions The data confirm the influence of TNFSF4 polymorphisms in SSc genetic susceptibility, especially in subsets of patients positive for lcSSc and ACA.
