  • c-Myc antagonizes the effect of p53 on apoptosis and p21WAF1 transactivation in K562 leukemia cells
  • 2000
  • Ingår i: Oncogene. - : Springer Science and Business Media LLC. - 0950-9232 .- 1476-5594. ; 19, s. 2194-2204
  • Tidskriftsartikel (refereegranskat)abstract
    • c-myc protooncogene positively regulates cell proliferation and overexpression of c-myc is found in many solid tumors and leukemias. In the present study we used the K562 human myeloid leukemia cell line as a model to study the functional interaction between c-Myc and p53. Using two different methods, we generated K562 transfectant cell lines with conditional expression of either c-Myc or p53. The cells expressed the p53Vall35 mutant, which adopts a wild-type conformation at 32 degrees C, while c-Myc induction was achieved with a zinc-inducible expression vector. We found that p53 in wild-type conformation induces growth arrest and apoptosis of K562. Expression of c-Myc significantly attenuated apoptosis and impaired the transcriptional activity of p53 on p21WAF1, Bax and cytomegalovirus promoters. The impairment of p21WAF1 transactivation by c-Myc was confirmed by transfection of a c-Myc-estrogen receptor fusion protein and by induction of c-myc by zinc in transfected cells. Also, p53-mediated up-regulation of p21WAF1 mRNA protein were significantly reduced by c-Myc, while Bax levels were unaffected. Consistently, c-Myc increased cyclin-dependent kinase 2 activity in K562 cells expressing p53 in wild-type conformation. These results suggest that c-Myc overexpression may antagonize the pro-apoptotic function of p53, thus providing a molecular mechanism for the frequently observed deregulation of c-myc in human cancer.

    • Sweden is nearly unique in providing and publicly funding meals for all schoolchildren. The current debate shows that the policy of providing school meals is universally accepted. That school meals are taken for granted makes the subject particularly interesting to problematize from a historical perspective, perhaps especially in light of the protracted implementation of the policy. The Swedish Rikadag voted in favor of providing school meals for every child in 1946. The aim in this thesis is to discuss the notions, ideas and political visions about children's food and eating from about 1900 to the end of the 1940's. The debate on school meals and child nutrition is focused to describe an important change in society in a way other than by concentrating more specifically on children or families. When the discussion on public health revealed the shortcomings in the knowledge and traditions of working-class families and the school system demanded that pupils pay attention and concentrate, children's hungry bellies were no longer a private matter. Malnutrition - nutrition that was faulty in quality or inadequate in quantity - became an important aspect of the welfare policy in a way that within its framework diffused and moved the border between private and public responsibility. Notions about children and their living conditions - the very construction of children's place in social and political contexts- that which we call childhood- had impact on the means by. which a reform like that of universal school meals could be implemented.

  • Language impairment in Swedish bilingual children: a comparison between bilingual and monolingual children in Malmö.
  • 2002
  • Ingår i: Acta Pædiatrica. - : Wiley. - 1651-2227 .- 0803-5253. ; 91:2, s. 229-234
  • Tidskriftsartikel (refereegranskat)abstract
    • In this study two groups of children were compared, 192 bilingual and 246 monolingual, referred over a 12-mo period to University Hospital in Malmö for suspected language impairment. Compared with monolingual children, bilingual children ran a significantly lower risk (RR = 0.76, p < 0.0001) of being referred by a child health centre, and a significantly higher risk (RR= 1.54, p < 0.0103) of being referred after 5 y of age. In the bilingual group there was also a significantly higher risk of parental refusal of assessment (RR = 2.35, p < 0.0016) and of the children being diagnosed with a severe language impairment (RR = 1.87, p < 0.0009). The risk for bilingual children with severe language impairment being discharged owing to non-attendance was high (RR = 6.20, p < 0.0002) compared to monolingual children. The risk increased with severity of language impairment. CONCLUSION: Bilingual children displayed a higher risk of a diagnosis of severe language impairment and of being discharged for non-attendance. The problem of low compliance among bilingual parents, despite strong indications of parental distress, must be addressed.
