  • On the Mechanism of MoSi2 Pesting in the Temperature Range 400–500°C

    • The oxidation of a MoSi2 composite was studied in the temperature range 400–500°C. The peak pesting temperature was identified and the oxidation kinetics was observed up to a holding time of 4000 hours. The reaction kinetics was tracked using thermogravimetric analysis :IS well iis oxide thickness nieasurenients. A detailed analysis of the morphology and composition of the oxide was performed using SEM and EDX. The peak pesting temperature was found to be 470°C. The reaction kinetics in static air could be described with two consecutive parabolic oxidation curves, one up to 500 hours and another between 500–4000 hours. With a higher water vapor coiitent the kinetics showed linear behwior. MoO3 evaporation took place during the entire duration of the oxidation. The oxide appeared to grow by inward diffusion of oxygen to the oxidehulk interface.

    • The oxidation characteristics of a MoSi2-based composite within the temperature range of 400–550 °C were investigated. The effects of temperature and water vapour on oxidation were examined. The oxidation kinetics were studied using a thermobalance, while the morphology and composition of the oxides were examined using XRD, ESEM/EDX, and SEM/EDX.The peak oxidation rates in dry O2 and View the MathML source were found to occur at temperatures of approximately 510 and 470 °C, respectively. Within the temperature range of accelerated oxidation (400–500 °C), the oxidation rate in View the MathML source was substantially higher than that in dry O2. At higher temperatures, the oxidation rate decreased, and the magnitude of the decrease was steeper and occurred at a lower temperature for View the MathML source (510 °C) than for O2 (550 °C). Furthermore, the rate of depletion of molybdenum (Mo) from the oxide scales during oxidation increased with increasing temperature and water vapour content. It appears that Mo loss is a key process influencing the protective properties of the oxide layer on the MoSi2 composite. A potential mechanism for the different oxidation behaviours in O2 and View the MathML source is proposed.

    • The oxidation of a MoSi2 composite was studied in dry air, oxygen and oxygensaturated with 10% water vapour at 450°C. The kinetics were investigated using TGA as well as oxide thickness measurements. Detailed analyses were performed on the morphology and composition of the oxide using XRD, ESEM, SEM, and EDX. It is shown that the oxidation rate increases drastically in the presence of water vapour, and the growth of Mo03 crystals on the oxide surface increases considerably. The different regions in the oxide cross-section are Mo-depleted compared with the corresponding regions in the bulk when oxidised in oxygen saturated with 10% water vapour. However, the samples oxidised in dry oxygen only shows Mo-depletion in some outer parts of the oxide. Accelerated growth of the MoSi2-oxide layer during exposure in 02+10%H20 compared to that in 02 can be related to the fact that more volatile Mo-species form in the presence of water vapour, resulting in a substantial loss of Mo03 from the inner part of the oxide. The voids left behind are not healed by the silica at this low temperature, which leaves the oxide with an open structure. As a result, the oxidation rate increases.

    • Boken "Tyska turister i Sverige" syftar till att öka kunskapen om tysk turism i Sverige - i ett historiskt perspektiv och i ett nutidsperspektiv. Ambitionen är inte att ge en helhetsbild. I stället inriktar sig boken på ett antal områden där ETOURs forskning i större eller mindre omfattning haft fokus på tysk turism. Det gäller fjällturism, fritidshusturism, miljöturism samt charterturism till norra Sverige. Detta kompletterat med färska studier bland tyska researrangörer och konsumenter samt en allmän bakgrund till tyskar och dess resande hoppas vi ska ge en värdefull bild av "Tyska turister i Sverige". Förhoppningen är också att den forskning och de studier som finns i boken mer allmänt ska kunna bidra till att öka kunskapen hos svenska turistföretag för att de ska bli framgångsrika på olika resemarknader. Dieter K. Müller, tidigare verksam vid ETOUR, nu lektor vid Umeå universitet, tar i de inledande avsnitten upp mer allmänna aspekter , t.ex. om tyskars syn på Sverige, varför just tyskar reser mest i världen och varför många tyskar väljer Sverige som turistmål. Dieter skriver i nästa kapitel om tyskarnas intresse för svenska fritidshus. Nästa kapitel handlar om de tyskägda stugorna i Norrland, skrivet av Robert Pettersson vid ETOUR. Den tyska fjällturisten är rubriken på ett avsnitt av Peter Fredman och Christine Lundberg, båda verksamma på ETOUR. Bodil Frey är författare till det avsnitt som handlar om de tyska gäster som kommer till vårt land enbart för att uppleva naturen. Interjuver av tyska researrangörer och konsumenter redovisas i de två avslutande avsnitten i boken. Flera av bokens kapitel avslutas med tips och slutsatser som kan vara till nytta för turistföretagare i deras strävan att bli framgångsrika på den tyska utresemarknaden.

  • Construction and characterization of affibody-Fc chimeras produced in Escherichia coli

    • Affibody-Fc chimeras were constructed by genetic fusion between different affibody affinity proteins with prescribed specificities and an Fc fragment derived from human I-G. Using affibody ligands previously selected for binding to respiratory syncytial virus (RSV) surface protein G and Thermus aquaticus (Taq) DNA polymerase, respectively, affibody-Fc fusion proteins showing spontaneous Fc fragment-mediated homodimerization via disulfide bridges were produced in Escherichia coli and affinity purified on protein A Sepharose from bacterial periplasms at yields ranging between 1 and 6 mg/l culture, Further characterization of the chimeras using biosensor technology showed that the affibody moieties have retained high selectivities for their respective targets after fusion to the Fc fragment. Avidity effects in the target binding were observed for the affibody-Fc chimeras compared to monovalent affibody fusion proteins, indicating that both affibody moieties in the chimeras were accessible and contributed in the binding. Fusion of a head-to-tail dimeric affibody moiety to the Fc fragment resulted in tetravalent affibody constructs which showed even more pronounced avidity effects. In addition, the Fc moiety of the chimeras was demonstrated to be specifically recognized by anti-human IgG antibody enzyme conjugates. One application for this class of artificial antibodies was demonstrated in a western blotting experiment in which one of the anti-RSV surface protein G affibody-Fc chimeras was demonstrated to be useful for specific detection of the target protein in a complex background consisting of a total E. coli lysate. The results show that through the replacement of the Fab portion of an antibody for an alternative binding domain based on a less complicated structure, chimeric proteins compatible with bacterial production routes containing both antigen recognition domains and Fc domains can be constructed. Such artificial antibodies should be interesting alternatives to, for example, whole antibodies or scFv-Fc fusions as detection devices and in diagnostic or therapeutic applications.

  • Alternative splicing of the human MUC2 gene.

    • Human colon cancers differ in amounts of MUC2 mucin synthesized. However, it is unclear whether MUC2 encodes a single protein. When clones of human colon cancer cells were assayed with antibodies against the TR2 mucin repeat or non-TR2 epitopes, differences in relative expression of MUC2 proteins suggested multiple immunoreactive forms. RT-PCR analysis detected the established 15kbp MUC2 cDNA and a novel form (designated MUC2.1) lacking the MUC2 TR2 repeat. Sequencing of cDNA and genomic DNA indicated that MUC2.1 results from an alternate splice donor. RT-PCR with splice-junction spanning primers confirmed the expression of MUC2.1 mRNA. Anti-MUC2.1 antibody stained colon cancer cells and normal colon in a pattern different from TR2-specific antibody. The presence of MUC2.1 mucin may help us to explain previous conflicting reports that have attempted to correlate the relative abundance of MUC2 protein and/or mRNA with the biological behavior of colon cancer cells.
