  • A polymer neural probe with tunable flexibility

    • A novel polymeric material, off stoichiometry thiol-ene-epoxy (OSTE+), has been evaluated for the fabrication of neural implants. OSTE+ is easily photo-structurable and exhibits mechanical properties suitable for stable implantation of the probe into brain tissue, while being sufficiently soft at physiological temperatures to reduce living tissue damage. The facile processing of OSTE+ allows use in applications where SU-8 or polyimide currently are the materials of choice. Uniquely, OSTE+ has a Young’s modulus of 1.9 GPa at 10 °C decreasing almost two orders of magnitude to 30 MPa at 40 °C, which can be compared to the Young’s modulus of 2.1 GPa for SU-8. We show a probe, with nine gold electrode sites, implanted into 0.5% agar at 40 °C using active cooling during the implantation.

    • Membranous nephropathy is one of the most common causes of nephrotic syndrome in adults. Recent reports suggest that treatment with adrenocorticotropic hormone (ACTH) reduces proteinuria, but the mechanism of action is unknown. Here, we identified gene expression of the melanocortin receptor MC1R in podocytes, glomerular endothelial cells, mesangial cells, and tubular epithelial cells. Podocytes expressed most MC1R protein, which colocalized with synaptopodin but not with an endothelial-specific lectin. We treated rats with passive Heymann nephritis (PHN) with MS05, a specific MC1R agonist, which significantly reduced proteinuria compared with untreated PHN rats (P < 0.01). Furthermore, treatment with MC1R agonists improved podocyte morphology and reduced oxidative stress. In summary, podocytes express MC1R, and MC1R agonism reduces proteinuria, improves glomerular morphology, and reduces oxidative stress in nephrotic rats with PHN. These data may explain the proteinuria-reducing effects of ACTH observed in patients with membranous nephropathy, and MC1R agonists may provide a new therapeutic option for these patients.

  • Effects of Melanocortin 1 Receptor Agonists in Experimental Nephropathies

    • Nephrotic syndrome, characterized by massive proteinuria, is caused by a large group of diseases including membranous nephropathy (MN) and focal segmental glomerulosclerosis (FSGS). Although the underlying mechanisms are beginning to unravel, therapy is unspecific and far from efficient. It has been suggested that adrenocorticotropic hormone (ACTH) has beneficial effects in patients with MN and possibly in other nephrotic diseases. We have previously reported that ACTH may act directly on podocytes through the melanocortin 1 receptor (MC1R). In the present study, we evaluate the effect of highly specific MC1R agonists in two different nephrotic disease models. Experimental MN: Passive Heymann nephritis (PHN) was induced in rats that were treated for four weeks with MS05, a selective MC1R agonist, or saline. The degree of albuminuria was significantly reduced over time and the effect was sustained one week after treatment withdrawal (p<0.05). Experimental FSGS: Based on a dose-response study, two doses of adriamycin were used for induction of nephropathy in Balb/c mice. Mice were treated with either a synthetic MC1R agonist (BMS-470539), with alpha-melanocyte stimulating hormone (alpha-MSH) or with saline. There was no beneficial effect of treatment. In summary, MC1R agonists reduce albuminuria and improve morphology in experimentally induced MN whereas they have no effect in experimental FSGS. The results illustrate the differences in these podocytopathies in terms of signaling mechanisms underlying proteinuria, and progression of disease.

  • Metabolic effects of n-3 PUFA as phospholipids are superior to triglycerides in mice fed a high-fat diet: Possible role of endocannabinoids

    • Background: n-3 polyunsaturated fatty acids, namely docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), reduce the risk of cardiovascular disease and can ameliorate many of obesity-associated disorders. We hypothesised that the latter effect will be more pronounced when DHA/EPA is supplemented as phospholipids rather than as triglycerides. Methodology/Principal Findings: In a 'prevention study', C57BL/6J mice were fed for 9 weeks on either a corn oil-based high-fat obesogenic diet (cHF; lipids ~35% wt/wt), or cHF-based diets in which corn oil was partially replaced by DHA/EPA, admixed either as phospholipids or triglycerides from marine fish. The reversal of obesity was studied in mice subjected to the preceding cHF-feeding for 4 months. DHA/EPA administered as phospholipids prevented glucose intolerance and tended to reduce obesity better than triglycerides. Lipemia and hepatosteatosis were suppressed more in response to dietary phospholipids, in correlation with better bioavailability of DHA and EPA, and a higher DHA accumulation in the liver, white adipose tissue (WAT), and muscle phospholipids. In dietary obese mice, both DHA/EPA concentrates prevented a further weight gain, reduced plasma lipid levels to a similar extent, and tended to improve glucose tolerance. Importantly, only the phospholipid form reduced plasma insulin and adipocyte hypertrophy, while being more effective in reducing hepatic steatosis and low-grade inflammation of WAT. These beneficial effects were correlated with changes of endocannabinoid metabolome in WAT, where phospholipids reduced 2-arachidonoylglycerol, and were more effective in increasing anti-inflammatory lipids such as N-docosahexaenoylethanolamine. Conclusions/Significance: Compared with triglycerides, dietary DHA/EPA administered as phospholipids are superior in preserving a healthy metabolic profile under obesogenic conditions, possibly reflecting better bioavalability and improved modulation of the endocannabinoid system activity in WAT. © 2012 Rossmeisl et al.

    • The main aim of this licentiate thesis is to examine how local actors in a medium-sized Swedish municipality, Värnamo, in their role as decision-makers at the local arena, describe and explain the preconditions and strategies for local economic and social development. The analysis uses semi-structured interviews to reveal the connections between the local actors representations of social and economic structures and their representations of central and future strategic choices. The local actors are represented by a) firm owners or central firm managers, b) local politicians in the municipal executive board, c) civil servants in central positions and d) key actors connected to local and regional organizations. The methodological approach is based on discourse analysis. The overall aim of the study is to understand how local discourses are constructed and communicated. The analysis is deepened by relating the local discourses to relevant theoretical models in contemporary economic geography. The economic-geographical theoretical models used in the analysis focus on industrial districts, clusters and innovation systems. Path-dependency, social embeddedness and creative class are other concepts of importance to understand the formation of the local discourse. The analysis shows how local discourses are affected by the perceived globalization process while being at the same time linked to a) specific historical and environmental factors leading to predominant behavioral norms and values stating that you should be hard working, economical, cooperative and trustworthy and b) the relative location, which is conceived to be central regarding communications and regional service functions. This is combined with an image of a creative and innovative local/regional production system, based on traditions in manufacturing and trade. The foundations in manufacturing and trade are however challenged by new sectors like logistics, design, and higher education in cooperation with universities in the region.

  • LIGATION-BASED MUTATION DETECTION AND RCA IN SURFACE UN-MODIFIED OSTE+ POLYMER MICROFLUIDIC CHAMBERS

    • For the first time, we demonstrate DNA mutation detection in surface un-modified polymeric microfluidic chambers without suffering from bubble trapping or bubble formation. Microfluidic devices were manufactured in off-stoichiometry thiol-ene epoxy (OSTE+) polymer using an uncomplicated and rapid fabrication scheme. The device performance was compared to that of PMMA and PDMS devices. In OSTE+ devices, we were able to perform ligation-based mutation detection and rolling circle amplification (RCA) assays directly on the un-modified surface without suffering from bubble formation or enzyme inhibition during bio-operation at elevated temperatures. In contrast, PMMA, PDMS and COP microfluidic devices required specific surface treatment.
