  • A 28-day toxicity study with highly purified PCB 180 in Sprague–Dawley rats
  • 2008
  • Ingår i: Toxicology Letters. - : Elsevier. ; , s. 205-
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • PCB 180 is a persistent and accumulative heptachlorinated non-dioxin-like polychlorinated biphenyl (NDL-PCB) abundantly present in food and the environment. Risk characterisation of NDL-PCBs has been confounded by the presence of highly potent dioxin-like (DL) impurities. We used therefore highly purified PCB 180 (DL impurities 2.7 ng TEqWHO/g PCB 180) for the 28-day toxicity study in adult rats. Groups of 5 males and 5 females were given total doses of 3, 10, 30, 100, 300, 1000 or 1700 mg/kg PCB 180 by gavage. Total dose was divided into 6 daily loading doses and 3 weekly maintenance doses. Bodyweight development was retarded at 1700 mg/kg during loading dosing, but recovered by the end of the study. Hepatic effects are reported separately (Roos et al., ibid.). Thyroid weight was increased in males. Plasma free T4 was dose-dependently decreased (maximally by 69% in males and 50% in females) in association with thyroid follicle depletion, but T3 was not affected. Increased thyroid follicle cell vacuolization is suggestive of thyroid activation. In males, vacuolization and extracellular deposits were observed in the frontal part of pituitary. Activation of adrenal cortex is demonstrated by vacuolization and hypertrophy observed in cells of zona fasciculata of the adrenal cortex. Erythrocyte parameters (red blood cell count, haematocrit, haemoglobin) were dose-dependently decreased in both genders. The results demonstrate a distinct toxicological profile of PCB 180 with some similarities with that of DL compounds, but clearly lower potency.Acknowledgement: Funded by the European Commission (ATHON, FOOD-CT-2005-022923

  • Att köra eller inte köra : Hur de äldre, åldrande och bilkörning har diskuterats i svensk transportpolitik
  • 2008
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • The objective of this study is to analyse the formation of older and ageing drivers in Swedish transportation policy. Documents from the Swedish Government, the Riksdag and, in a few cases, public authorities and newspapers are analysed. The discourse-analytical framework is inspired by Foucault’s archaeology, which is used as a tool box. The study identifies/constructs three periods in the formation of older and ageing drivers. In the first period (1934–1967) there is a discursive gap between statements about driver selection and traffic safety. Driving in ‘old age’ is a problem for medical expertise and mandatory tests are proposed. The proposals are rejected, as older people’s driving is not a problem in the discourse of traffic safety. In the second period (1967–1991) ‘older drivers’ is made up as a category of interest. ‘Older drivers’ become an accident population in the discourse of traffic safety due to changes in statistical practices. In the third period (1991–2006) a discourse of older peo-ple’s mobility is appearing alongside the discourses of traffic safety and driver selection. Moreover, the study traces how the statements of older and ageing drivers are ordered by more general ideas of the aged and ageing. Double discourses of ageing are followed by a dichotomisation in ‘healthy’ and ‘sick older drivers’ in the 1990s. The idea of an abrupt exit in old age alters to a more gradual exit, in its turn followed by challenging the very idea of exit.

  • Ålderism - ett fruktbart begrepp?
  • 2008

  • Tidskriftsartikel (refereegranskat)abstract
    • Ageism has gained growing attention in Sweden the last decade. Age is even discussed to be included in the discrimination legislation. Still, the concept has not been of much sociological interest. This article is an overview of the concept ageism and how it has been discussed in social gerontology. In the article it is argued that the overall focus has been on overtly expressions such as stereotypes and discriminating behavior while underlying structures and processes of power and power relations have not got enough attention. As a result the concept of ageism has become limited as an analytical tool. Thus, in order to develop the potential of the concept it seems crucial to explicate and theorise power relations. To accomplish this goal, i.e. to focus on how age based power relations are negotiated, challenged and reproduced in processes and institutionalised practise it is suggested that ageism might have to be complemented with other concepts, such as age coding.

  • Intra- and extracellular signaling by endothelial neuregulin-1
  • 2007
  • Ingår i: Experimental Cell Research. - : Elsevier BV. - 0014-4827 .- 1090-2422. ; 313:13, s. 2896-2909
  • Tidskriftsartikel (refereegranskat)abstract
    • Suppression of tumor growth by inhibition of ErbB receptor signaling is well documented. However, relatively little is known about the ErbB signaling system in the regulation of angiogenesis, a process necessary for tumor growth. We have previously shown that heparin-binding EGF-like growth factor (HB-EGF) is expressed by vascular endothelial cells (EC) and promotes endothelial recruitment of vascular smooth muscle cells (SMC). To assess whether other members of the EGF-family regulate angiogenesis, the expression of 10 EGF-like growth factors in primary ECs and SMCs was analyzed. In addition to HB-EGF, neuregulin-1 (NRG-1) was expressed in ECs in vitro and in vivo. Endothelial NRG-1 was constitutively processed to soluble extracellular and intracellular signaling fragments, and its expression was induced by hypoxia. NRG-1 was angiogenic in vivo in mouse corneal pocket and chicken chorioallantoic membrane (CAM) assays. However, consistent with the lack of NRG-1 receptors in several primary EC lines, NRG-1 did not directly stimulate cellular responses in cultured ECs. In contrast, NRG-1 promoted EC responses in vitro and angiogenesis in CAM in vivo by mechanisms dependent on VEGF-A and VEGFR-2. These results indicate that NRG-1 is expressed by ECs and regulates angiogenesis by mechanisms involving paracrine up-regulation of VEGF-A.

  • Äldre i transportsystemet : mobilitet, design och träningsproblematik
  • 2007
  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • Generally, more elderly will be travelling and be out on the roads as active road-users in the future. Research exists on the travelling habits of the elderly; but more in-depth knowledge on the elderly's preferences as license-holders, drivers, road-users and actors in public transport is required. The aim of this report is to give an overview of previous research as well as to indicate a number of directions for future research on the mobility of the elderly as actors within the transport system. The work has a clear multidisciplinary approach, with knowledge from social science, behavioural science and technical research on transport and the elderly. However, the main weight lays on social science and behavioural science issues. The report is divided into eleven chapters: 1) contains a short background, purpose and method questions; 2) discusses the project's scientific and social relevance; 3) provides theoretical background and theoretical concepts; 4) mentions previous research on the elderly as car drivers; 5) is a chapter on license-less vehicles; 6) discusses traffic and road design for the elderly; 7) discusses the elderly as pedestrians and bicycle road-users; 8) is about the elderly in public transport and 9) is about the training of elderly drivers. Chapter 10) consists of a final discussion and chapter 11) summarises point by point the need for research on issues which have come to light in the report
