  • Insights from past trends in exergy efficiency and carbon intensity of electricity : Portugal, 1900–2014

    • We use the societal exergy analysis to identify periods and factors controlling efficiency dilution and carbon deepening of electricity in Portugal from 1900 to 2014. Besides estimating the carbon intensity of electricity production, we propose a new indicator, the carbon intensity of electricity use, which quantifies CO 2 /kWh of electricity derived useful exergy. Results show final to useful efficiency dilution until World War I (50% to 30%) due to a decrease in share of the high-efficiency transport sector and from mid-1940s to 1960 and mid-1990s onwards (58% to 47% and 47% to 40%) due to an increase in share of the low efficiency commercial and residential sector. Decarbonization from 1900 to mid-1960s, with carbon intensities of electricity production and use dropping respectively from 12.8 to 0.2 and from 33.6 to 0.4 kg CO 2 /kWh due to an increase in thermoelectricity efficiencies and an increase in share of hydro. Then, a period of carbon deepening until 1990 with carbon intensities tripling due to a shift in shares from hydro to thermoelectricity and more recently a period of decarbonization with carbon intensities decreasing to 0.35 and 0.9 kg CO 2 /kWh, due to the increase in renewable electricity despite a dilution in final to useful efficiency.

  • TMEM16A calcium-activated chloride currents in supporting cells of the mouse olfactory epithelium

    • Glial-like supporting (or sustentacular) cells are important constituents of the olfactory epithelium that are involved in several physiological processes such as production of endocannabinoids, insulin, and ATP and regulation of the ionic composition of the mucus layer that covers the apical surface of the olfactory epithelium. Supporting cells express metabotropic P2Y purinergic receptors that generate ATP-induced Ca2+ signaling through the activation of a PLC-mediated cascade. Recently, we reported that a subpopulation of supporting cells expresses also the Ca2+-activated Cl− channel TMEM16A. Here, we sought to extend our understanding of a possible physiological role of this channel in the olfactory system by asking whether Ca2+ can activate Cl− currents mediated by TMEM16A. We use whole-cell patch-clamp analysis in slices of the olfactory epithelium to measure dose–response relations in the presence of various intracellular Ca2+ concentrations, ion selectivity, and blockage. We find that knockout of TMEM16A abolishes Ca2+-activated Cl− currents, demonstrating that TMEM16A is essential for these currents in supporting cells. Also, by using extracellular ATP as physiological stimuli, we found that the stimulation of purinergic receptors activates a large TMEM16A-dependent Cl− current, indicating a possible role of TMEM16A in ATP-mediated signaling. Altogether, our results establish that TMEM16A-mediated currents are functional in olfactory supporting cells and provide a foundation for future work investigating the precise physiological role of TMEM16A in the olfactory system.

  • Endoscopic closure of transmural bladder wall perforations.

    • BACKGROUND: Traditionally, intraperitoneal bladder perforations caused by trauma or iatrogenic interventions have been treated by open or laparoscopic surgery. Additionally, transvesical access to the peritoneal cavity has been reported to be feasible and useful for natural orifice translumenal endoscopic surgery (NOTES) but would be enhanced by a reliable method of closing the vesicotomy. OBJECTIVE: To assess the feasibility and safety of an endoscopic closure method for vesical perforations using a flexible, small-diameter endoscopic suturing kit in a survival porcine model. DESIGN, SETTING, AND PARTICIPANTS: This pilot study was performed at the University of Minho, Braga, Portugal, using six anesthetized female pigs. INTERVENTIONS: Closure of a full-thickness longitudinal incision in the bladder dome (up to 10 mm in four animals and up to 20 mm in two animals) with the endoscopic suturing kit using one to three absorbable stitches. MEASUREMENTS: The acute quality of sealing was immediately tested by distending the bladder with methylene-blue dye under laparoscopic control (in two animals). Without a bladder catheter, the animals were monitored daily for 2 wk, and a necropsy examination was performed to check for the signs of peritonitis, wound dehiscence, and quality of healing. RESULTS AND LIMITATIONS: Endoscopic closure of bladder perforation was carried out easily and quickly in all animals. The laparoscopic view revealed no acute leak of methylene-blue dye after distension of the bladder. After recovery from anaesthesia, the pigs began to void normally, and no adverse event occurred. Postmortem examination revealed complete healing of vesical incision with no signs of infection or adhesions in the peritoneal cavity. No limitations have yet been studied clinically. CONCLUSIONS: This study demonstrates the feasibility and the safety of endoscopic closure of vesical perforations with an endoscopic suturing kit in a survival porcine model. This study provides support for further studies using endoscopic closure of the bladder which may lead to a new era in management of bladder rupture and adoption of the transvesical port in NOTES procedures.

    • TMEM16A/ANO1 is a calcium-activated chloride channel expressed in several types of epithelia and involved in various physiological processes, including proliferation and development. During mouse embryonic development, the expression of TMEM16A in the olfactory epithelium is dynamic. TMEM16A is expressed at the apical surface of the entire olfactory epithelium at embryonic day E12.5 while from E16.5 its expression is restricted to a region near the transition zone with the respiratory epithelium. To investigate whether TMEM16A plays a role in the development of the mouse olfactory epithelium, we obtained the first immunohistochemistry study comparing the morphological properties of the olfactory epithelium and nasal glands in TMEM16A-/- and TMEM16A+/+ littermate mice. A comparison between the expression of the olfactory marker protein and adenylyl cyclase III shows that genetic ablation of TMEM16A did not seem to affect the maturation of olfactory sensory neurons and their ciliary layer. As TMEM16A is expressed at the apical part of supporting cells and in their microvilli, we used ezrin and cytokeratin 8 as markers of microvilli and cell body of supporting cells, respectively, and found that morphology and development of supporting cells were similar in TMEM16A-/- and TMEM16A+/+ littermate mice. The average number of supporting cells, olfactory sensory neurons, horizontal and globose basal cells were not significantly different in the two types of mice. Moreover, we also observed that the morphology of Bowman’s glands, nasal septal glands and lateral nasal glands did not change in the absence of TMEM16A. Our results indicate that the development of mouse olfactory epithelium and nasal glands does not seem to be affected by the genetic ablation of TMEM16A.

  • Improving the robustness of societal exergy accounting : from primary energy to energy services

    • Results of useful exergy accounting at the societal (i.e. national or global) level are potentially important for policy purposes, such as the development of energy or GHG emission scenarios, and the determination of the major energy inefficiencies (and thus improvement potentials) within a country. However, useful exergy societal studies commonly differ in their accounting methodology, which affect the results. Such differences include the starting point (primary or final energy), the method used to compute primary exergy, the classes of useful exergy categories considered, the definition of second law efficiencies, the inclusion (or not) of exergy inputs related with non-energy uses, and the inclusion or not of muscle (and human) work. To help bring a more consistent approach to useful exergy accounting at the societal level, we review the methodologies of past studies, highlighting the differences and discussing their advantages and disadvantages in each case. Based on this, we select our preferred options, leading to a proposed common methodology that can be used to build national and international series of useful exergy.

    • Studies of societal exergy use have the common aim of tracing the flow of exergy along society, and are used to gain insights into the efficiency of energy use and linkages to economic growth. However, their methodological approaches vary greatly, with significant impacts on results. Therefore, we make a review of past studies to identify, synthesize and discuss methodological differences, to contribute to a more consistent and robust approach to societal exergy accounting. Issues that should be taken into account when making methodological options are discussed and key insights are presented: (1) For mapping of primary inputs and useful exergy categories, the inclusion of all natural resources is more consistent but it has the cost of not being able to distinguish the various energy end-uses in the production of materials. (2) To estimate primary electricity, none of the methods currently used is able to capture simultaneously the efficiency of the renewable energy sector, the environmental impact and the efficiency of energy use in society. (3) To estimate final-to-useful exergy conversion efficiencies, standard thermodynamic definitions should be used because the use of proxies fails to distinguish between increases in exergy efficiency and increases in the efficiency of providing energy services.

  • On the right track? Energy use, carbon emissions, and intensities of world rail transportation, 1840–2020

    • The history of rail transport can offer valuable insights for future energy transitions due to its importance in promoting clean mobility. There is a complex interplay between the evolution of the railway network, fuel consumption, efficiency, energy service, and CO2 emissions that requires further exploration. We developed a dataset that covers energy use in all stages of rail transportation, as well as the length of track, energy service, and CO2 emissions at the world scale. To deal with missing data we utilized machine learning techniques for the first time in a historical energy reconstruction study. Our analysis reveals that for world rail transport (1) the final-to-useful efficiency has increased by 30-fold from 1840 to 2020, mainly due to the replacement of steam trains with diesel and electric ones, (2) the peak in final energy use occurred in the 1940s, while useful energy use and transport service continue to grow, (3) there was a reduction in the energy (carbon) intensity from approximately 20 to 0.2 MJ/tkm (2 to 0.02 kg CO2/tkm) between 1840 and 2010, due not only to the increase in final-to-useful efficiency but also to rising occupancy, better operating conditions, and reduced losses by the passive system.
