  • Altered cytoplasmic-to-nuclear ratio of survivin is a prognostic indicator in breast cancer

    • Purpose: Survivin (BIRC5) is a promising tumor biomarker. Conflicting data exist on its prognostic effect in breast cancer. These data may at least be partly due to the manual interpretation of immunohistochemical staining, especially as survivin can be located in both the nucleus and cytoplasm. Quantitative determination of survivin expression using image analysis offers the opportunity to develop alternative scoring models for survivin immunohistochemistry. Here, we present such a model. Experimental Design: A breast cancer tissue microarray containing 102 tumors was stained with an anti-survivin antibody. Whole-slide scanning was used to capture high-resolution images. These images were analyzed using automated algorithms to quantify the staining. Results: Increased nuclear, but not cytoplasmic, survivin was associated with a reduced overall survival (OS; P = 0.038) and disease-specific survival (P = 0.0015). A high cytoplasmicto-nuclear ratio (CNR) of survivin was associated with improved OS (P = 0.005) and disease-specific survival (P = 0.05). Multivariate analysis revealed that the survivin CNR was an independent predictor of CIS (hazard ratio, 0.09; 95% confidence interval, 0.01-0.76; P = 0.027). A survivin CNR of >5 correlated positively with estrogen receptor (P = 0.019) and progesterone receptor (P = 0.033) levels, whereas it was negatively associated with Ki-67 expression (P = 0.04), p53 status (P = 0.005), and c-myc amplification (P = 0.016). Conclusion: Different prognostic information is supplied by nuclear and cytoplasmic survivin in breast cancer. Nuclear survivin is a poor prognostic marker in breast cancer. Moreover, CNR of survivin, as determined by image analysis, is an independent prognostic factor.

  • Understanding the phylogeographic patterns of European hedgehogs, Erinaceus concolor and E. europaeus using the MHC.

    • The genome of the European hedgehog, Erinaceus concolor and E. europaeus, shows a strong signal of cycles of restriction to glacial refugia and postglacial expansion. Patterns of expansion, however, differ for mitochondrial DNA (mtDNA) and preliminary analysis of nuclear markers. In this study, we determine phylogeographic patterns in the hedgehog using two loci of the major histocompatibility complex (MHC), isolated for the first time in hedgehogs. These genes show long persistence times and high polymorphism in many species because of the actions of balancing selection. Among 84 individuals screened for variation, only two DQA alleles were identified in each species, but 10 DQB alleles were found in E. concolor and six in E. europaeus. A strong effect of demography on patterns of DQB variability is observed, with only weak evidence of balancing selection. While data from mtDNA clearly subdivide both species into monophyletic subgroups, the MHC data delineate only E. concolor into distinct subgroups, supporting the preliminary findings of other nuclear markers. Together with differences in variability, this suggests that the refugia history and/or expansion patterns of E. concolor and E. europaeus differ.

  • Expression of the cytoskeleton linker protein ezrin in human cancers

    • Expression of the metastasis-associated protein, ezrin, in over 5,000 human cancers and normal tissues was analyzed using tissue microarray immunohistochemistry. Ezrin staining was compared between cancers and their corresponding normal tissues, between cancers of epithelial and mesenchymal origin, in the context of the putative inhibitor protein, merlin, and against clinicopathological data available for breast, lung, prostate cancers and sarcomas. Ezrin was found in most cancers and normal tissues at varying levels of intensity. In general ezrin was expressed at higher levels in sarcomas than in carcinomas. By normalizing the expression of ezrin in each cancer using ezrin expression found in the corresponding normal tissue, significant associations between ezrin were found in advancing histological grade in sarcomas (P = 0.02) and poor outcome in breast cancer (P = 0.025). Clinicopathologic associations were not changed by simultaneous assessment of ezrin and merlin in each patient sample for the cancer types examined. These data support a role for ezrin in the biology of human cancers and the need for additional studies in breast cancer and sarcoma patients that may validate ezrin as a marker of cancer progression and as a potential target for cancer therapy.

  • Benthic nutrient fluxes along an estuarine gradient: influence of the pinnid bivalve Atrina zelandica in summer

    • Benthic nutrient fluxes (BNF) can supply 30 to 100% of the nutrient requirements of benthic and pelagic algae in an estuary, and can, thus, potentially sustain benthic and pelagic primary production within the estuarine food web. While BNF can be influenced by microbial processes, epibenthic suspension-feeding bivalves have the potential to alter fluxes by their influence on the community composition of surrounding macrofauna and benthic boundary conditions, and their feeding activities. In Mahurangi Harbour, New Zealand, the large suspension feeding pinnid Atrina zelandica (hereafter referred to as Atrina) occupies large areas of the harbour floor. Consequently, Atrina have the potential to substantially influence the BNF and, thus, primary production, and the food supply to the filter feeding community within the harbour, including the rack-farmed Pacific oyster aquaculture industry. Mahurangi Harbour is almost always isohaline, but exhibits a strong gradient in suspended sediment concentration, which declines from head to mouth. As Atrina increase their rate of pseudofaeces production with increases in suspended sediment concentration, we conducted in situ light and dark paired benthic chamber experiments with and without Atrina at 4 stations along this turbidity gradient, to determine their effect on BNF. Our results showed substantially greater BNF from Atrina beds than bare sediments. We also found greater net BNF (difference between Atrina beds and bare sediment) in the less turbid water under dark conditions, but enhanced water column nutrient supply in the more turbid water in light, due to Atrina excretion of ammoniacal nitrogen (NH4-N). On an areal basis, we estimate that BNF from Atrina beds may account for up to 80% of the nutrient supply for pelagic primary production and, thus, they are of major importance to the sustainability of aquaculture in this harbour.

    • Vegetation patterns of subalpine gully-head mires were investigated in the flat-topped Lammerlaw and Lammermoor Ranges, South Island, New Zealand. Two intensively studied mires each consist of a series of peaty terraces and scarps. Terraces may contain pools, elongated downslope in the narrow, lower altitude mire, but across slope in the broader, upper mire. A crest occurs on some terrace lips, and marginal "spillways" (channel-like zones) occur down some scarps. Some mires have drained by subsurface pipes. Vegetation analysis distinguished between grassland or herbfield on gully sides, vegetation of mire margins, showing aspect differences on the steeper, lower mire, and the vegetation of gully floors, including oligotrophic mire centre vegetation and species-poor pools. The crests, though warmer, bore no special vegetation type. Mineral soil beneath the peat indicates a previous non-mire vegetation, which has subsequently paludified. Scarp slumps indicate downslope creep of organic material. Peat fissures, and mineral, vegetation, and erosion dams all appear to have initiated development of some pools. Mires are designated gully-head ribbon fens. Patterning appears to be accentuated because of the mires' gully-head location on broad-topped ranges, and drainage of soligenous water from upslope gully sides. These apparently unique fens give insight into patterning in aapa mires, and merit special conservation.

  • Monitoring and quantification of inclusion body formation in Escherichia coli by multi-parameter flow cytometry

    • Multi-parameter flow cytometry was used to monitor the formation of promegapoietin (PMP) inclusion bodies during a high cell density Escherichia coli fed-batch fermentation process. Inclusion bodies were labelled with a primary antibody and then with a secondary fluorescent antibody. Using this method it was possible to detect PMP inclusion body formation with a high specificity and it was possible to monitor the increased accumulation of the protein with process time (6-48 mg PMP/g CDW) whilst highlighting population heterogeneity.
