  • A serological type II collagen neoepitope biomarker reflects cartilage breakdown in patients with osteoarthritis

    • Objectives: There is an unmet medical need for biomarkers in OA which can be applied in clinical drug development trials. The present study describes the development of a specific and robust assay measuring type II collagen degradation (T2CM) and discusses its potential as a noninvasive translational biomarker. Methods: A type II collagen specific neoepitope (T2CM) was identified by mass spectrometry and monoclonal antibodies were raised towards the epitope, employed in a chemiluminescence immunoassay. T2CM was assessed in bovine cartilage explants with or without MMP-13 inhibitor, and explant supernatants were analyzed by Western blot. T2CM was measured in plasma samples from one study (n ​= ​48 patients) where OA patients were referred to total knee replacement (TKR). Additionally, T2CM was quantified in serum from OA patients receiving salmon calcitonin treatment (sCT) (n ​= ​50) compared to placebo (n ​= ​57). Results: The T2CM assay was technically robust (13/4 ​% inter/intra-variation) and specific for the type II collagen fragment cleaved by MMP-1 and -13. The MMP-13 inhibitor reduced the T2CM release from bovine cartilage explants receiving catabolic treatment. These results were confirmed by Western blot. In human end-stage OA patients (scheduled for TKR), the T2CM levels were elevated compared to moderate OA (p<0.004). The OA patients receiving sCT had lower levels of T2CM compared to placebo group after 1, 6, and 24 months of treatment (p ​= ​0.0285, p ​= ​0.0484, p ​= ​0.0035). Conclusions: To our knowledge, T2CM is the first technically robust serological biomarker assay which has shown biological relevance in ex vivo models and OA cohorts. This suggests that T2CM may have potential as a translational biomarker for cartilage degradation.

    • This paper presents 'virtual collaborative robot', a virtual reality platform for designing and evaluating collaboration between operators and industrial robots. Within the platform, human-robot collaboration scenarios can be created and a user can interact with a robot without the safety risks that might arise with physical industrial robots. In an initial evaluation of the platform a scenario was implemented combining speech recognition, haptic control, and augmented reality to assemble a car model. The results from this evaluation indicate that the suggested platform can be used to successfully test new applications with the standard equipment of virtual reality headsets.

    • Industry is on the threshold of the fourth industrial revolution where smart factories area necessity to meet customer demands for increasing volumes of individualized products. Within the smart factory, cyber-physical production systems are becoming important to deal with changing production. Human-robot collaboration is an example of a cyber-physical system in which humans and robots share a workspace. By introducing robots and humans into the same working cell, the two can collaborate by allowing the robot to deal with heavy lifting, repetitive, and high accuracy tasks, while the human focuses on tasks that need intelligence, flexibility, and adaptability. There are few such collaborative applications in industry today. In the implementations that actually exist, the robots are mainly working side-by-side with humans rather than truly collaborating. Three main factors that limit the widespread application of human-robot collaboration can be identified: lack of knowledge regarding suitable human-robot collaboration tasks, lack of knowledge regarding efficient communication technologies for enabling interaction between humans and robots when carrying out tasks, and lack of efficient ways to safely analyze and evaluate collaborative tasks.The overall aim of this thesis is to address these problems and facilitate and improve interaction between humans and robots, with a special focus on assembly manufacturing tasks. To fulfill this aim, an assembly workstation for human-robot collaboration has been developed and implemented both physically and virtually. A virtual reality platform called ViCoR has been developed that can be used to investigate, evaluate, and analyze the interaction between humans and robots and thereby facilitate the implementation of new human-robot collaboration cells. The workstation developed has also been used for data collection and experiments during the thesis work, and used to extract knowledge of how the interaction between human and robot can be improved.

  • Flipped or traditional online teaching? : Two different strategies to handle teaching in nursing education during the COVID-19 pandemic.

    • OBJECTIVES: The aim of this study was to investigate differences in academic achievement between two groups of students who were taught the same course online within the nursing program but through two different teaching strategies and to examine the students' attitudes towards flipped classroom.METHODS: Online lectures using Zoom was given to teach a course regarding the immune system and another course was taught the same subject in flipped classroom approach using video lectures followed by seminars. Academic achievement were compared between the groups, and perspectives on flipped classroom were investigated using a questionnaire.RESULTS: The main findings were that participation in flipped classroom seminars had a positive effect on academic achievement (OR 2.3 (CI [1.001-5.1]), and that students preferred the flipped classroom approach over traditional lectures.CONCLUSIONS: This study suggests that a student centered teaching strategy like flipped classroom is an effective way to increase the students' engagement and academic achievements.

    • Kursen Anatomi och fysiologi 15 hp är en fristående kurs som ges på halvfart och helt på distans vid ämnet Medicinsk vetenskap. Ursprungligen gavs kursen inom Biovetenskapligt program på campus och på distans genom filmade campusföreläsningar. Satsningar på distansutbildning görs för att tillgodose kompetensförsörjningen i hela landet så omformningen av kursen i Anatomi och fysiologi från campus till distans ligger helt i linje med detta. Speciellt då detta hela tiden efterfrågats från studenthåll.Kursen ges helt och hållet på distans genom filmade föreläsningar där varje kursavsnitt följs av uppföljande seminarier via Zoom för diskussioner kring materialet utefter studenternas frågeställningar. Detta ger studenterna möjlighet till interaktivitet med varandra och med lärarna. Till varje föreläsning finns tillhörande övningsquiz och detaljerade inlärningsmål som ofta används som grund till seminarierna. Övningsquizen fungerar som hjälpmedel för att studenterna ska kunna bedöma och ta ett större ansvar för sitt eget lärande och dessutom förbättra sina akademiska prestationer. Studenterna examineras kontinuerligt under kursen genom två canvasquiz, ett zoomseminarium kring medicinsk etik och två salstentor.En återkommande kommentar från studenterna är att kursen har en mycket tydlig struktur och upplägg i Canvas. Kursstrukturen i Canvas är tätt kopplad till schemat och därmed blir det tydligt och enkelt för studenterna att följa. Övningsquizen och zoomseminarierna är uppskattade moment som i viss mån kompenserar för den nu uteblivna interaktionen mellan lärare-student och student-student som annars uppstår vid campusföreläsningar. Införandet av övningsquiz har dessutom förbättrat resultatet hos de som genomför examinationerna.

  • Impact of digital literacy on academic achievement : Evidence from an online anatomy and physiology course

    • The increasing prevalence of online courses has highlighted the importance of digital literacy for students. This study investigates the relationship between digital literacy and academic achievement among students who participate in an online course on anatomy and physiology. The study also evaluates how different aspects of digital literacy, such as age and previous education in natural science, affect students’ grades. Using logistic regression analysis, data from five areas of digital literacy that were assessed among participants with different educational backgrounds are analyzed. The results show that some aspects of digital literacy are more crucial for academic success in the online course. Students with a natural science background exhibited higher levels of digital literacy, emphasizing the importance of considering previous education in supporting students’ digital skills in online courses. The study also reveals that students were proficient in self-assessing their own digital literacy, enabling easy evaluation of the collective digital literacy within the course and facilitating targeted interventions for all students, regardless of their initial digital literacy levels. This study underscores the importance of digital literacy in online education. It highlights the specific areas of digital literacy that strongly contribute to academic achievement and emphasizes the positive impact of previous education in natural science on students’ digital skills. These findings suggest that instructors should consider these factors when designing and delivering online courses to ensure equal opportunities for students to enhance their digital literacy.

    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

    • A new method for the fluorine-18 labelling of trifluoromethyl ketones has been developed. This method is based on the conversion of a-COCF3 functional group to a difluoro enol silyl ether followed by halogenation and fluorine-18 labelling. The utility of this new method was demonstrated by the synthesis of fluorine-18 labelled neutrophil elastase inhibitors, which are potentially useful for detection of inflammatory disorders.

    • Rationale: Oxygen is essential for cellular energy metabolism. Neurons are particularly vulnerable to hypoxia. Increasing oxygen supply shortly after stroke onset could preserve the ischemic penumbra until revascularization occurs.Aims: PROOF investigates the use of normobaric oxygen (NBO) therapy within 6 h of symptom onset/notice for brain-protective bridging until endovascular revascularization of acute intracranial anterior-circulation occlusion.Methods and design: Randomized (1:1), standard treatment-controlled, open-label, blinded endpoint, multicenter adaptive phase IIb trial.Study outcomes: Primary outcome is ischemic core growth (mL) from baseline to 24 h (intention-to-treat analysis). Secondary efficacy outcomes include change in NIHSS from baseline to 24 h, mRS at 90 days, cognitive and emotional function, and quality of life. Safety outcomes include mortality, intracranial hemorrhage, and respiratory failure. Exploratory analyses of imaging and blood biomarkers will be conducted.Sample size: Using an adaptive design with interim analysis at 80 patients per arm, up to 456 participants (228 per arm) would be needed for 80% power (one-sided alpha 0.05) to detect a mean reduction of ischemic core growth by 6.68 mL, assuming 21.4 mL standard deviation.Discussion: By enrolling endovascular thrombectomy candidates in an early time window, the trial replicates insights from preclinical studies in which NBO showed beneficial effects, namely early initiation of near 100% inspired oxygen during short temporary ischemia. Primary outcome assessment at 24 h on follow-up imaging reduces variability due to withdrawal of care and early clinical confounders such as delayed extubation and aspiration pneumonia.
