  • Increased CD4+ T cell lineage commitment determined by CpG methylation correlates with better prognosis in urinary bladder cancer patients

    • BACKGROUND: Urinary bladder cancer is a common malignancy worldwide. Environmental factors and chronic inflammation are correlated with the disease risk. Diagnosis is performed by transurethral resection of the bladder, and patients with muscle invasive disease preferably proceed to radical cystectomy, with or without neoadjuvant chemotherapy. The anti-tumour immune responses, known to be initiated in the tumour and draining lymph nodes, may play a major role in future treatment strategies. Thus, increasing the knowledge of tumour-associated immunological processes is important. Activated CD4+ T cells differentiate into four main separate lineages: Th1, Th2, Th17 and Treg, and they are recognized by their effector molecules IFN-γ, IL-13, IL-17A, and the transcription factor Foxp3, respectively. We have previously demonstrated signature CpG sites predictive for lineage commitment of these four major CD4+ T cell lineages. Here, we investigate the lineage commitment specifically in tumour, lymph nodes and blood and relate them to the disease stage and response to neoadjuvant chemotherapy.RESULTS: Blood, tumour and regional lymph nodes were obtained from patients at time of transurethral resection of the bladder and at radical cystectomy. Tumour-infiltrating CD4+ lymphocytes were significantly hypomethylated in all four investigated lineage loci compared to CD4+ lymphocytes in lymph nodes and blood (lymph nodes vs tumour-infiltrating lymphocytes: IFNG -4229 bp p < 0.0001, IL13 -11 bp p < 0.05, IL17A -122 bp p < 0.01 and FOXP3 -77 bp p > 0.05). Examination of individual lymph nodes displayed different methylation signatures, suggesting possible correlation with future survival. More advanced post-cystectomy tumour stages correlated significantly with increased methylation at the IFNG -4229 bp locus. Patients with complete response to neoadjuvant chemotherapy displayed significant hypomethylation in CD4+ T cells for all four investigated loci, most prominently in IFNG p < 0.0001. Neoadjuvant chemotherapy seemed to result in a relocation of Th1-committed CD4+ T cells from blood, presumably to the tumour, indicated by shifts in the methylation patterns, whereas no such shifts were seen for lineages corresponding to IL13, IL17A and FOXP3.CONCLUSION: Increased lineage commitment in CD4+ T cells, as determined by demethylation in predictive CpG sites, is associated with lower post-cystectomy tumour stage, complete response to neoadjuvant chemotherapy and overall better outcome, suggesting epigenetic profiling of CD4+ T cell lineages as a useful readout for clinical staging.

  • Disturbance of the herbaceous layer after invasion of an eutrophic temperate forest by wild boar

    • Disturbances of the soil and the tree canopy are crucial factors determining the diversity, composition and biomass of the herbaceous layer in forests. This study presents a detailed account of ground vegetation in permanent plots surveyed before and after invasion of wild boar (Sus scrofa) to a temperate deciduous broadleaf forest. Specifically, we aimed to quantify the effect of wild boar rooting on cover, richness and composition of spring ephemerals, summer green herbs and saplings of woody species in relation to tree canopy cover. Rooting frequency in sample plots increased from 0% in 2010 to 61% in 2013. In heavily rooted plots, the mean cover of spring ephemeral geophytes (mainly Anemone nemorosa, A. ranunculoides and Ranunculus ficaria) decreased from 75% to 39% between 2010 and 2013. Species richness of summer green herbs generally increased between 2010 and 2013 and was additionally positively affected by heavy rooting and low canopy cover. Rooting also caused heterogenization of the herbaceous layer and amplified ongoing compositional changes induced by changing light conditions. Frequency and richness of spring ephemeral and woody species remained unchanged. We conclude that overall species richness of the herbaceous layer may increase in the short-term as a result of increased plant recruitment and seed dispersal. However, wild boar rooting can greatly reduce the ground cover of spring ephemerals in eutrophic broadleaf forests, thereby threatening their important ecological function. To avoid long-term losses of characteristic spring flora elements, local population control of wild boar is necessary to reduce abundance and frequency of soil rooting.

  • Influence of Cutting Type and Fertilization in Production of Containerized Poplar Plants

    • Most poplar plantations are planted on marginal agricultural land, but poplar plantations also hold the potential for increased profits compared to plantations of other species on non-agricultural, previously forested land. To date, the establishment of poplar plantations on previously forested land is limited by the production of suitable containerized poplar stock for planting. The objective of this study is to investigate how different cutting quality and fertilizer treatments influence height, diameter, and root biomass growth and root-to-shoot ratio, all important variables for plant establishment. Our results show that fertilization increases plant growth and that single-bud and two-bud cuttings with cutting diameters of 5 to 10 mm can be used in the production of containerized plants. Root biomass was similar between these plant types but the number of roots per plant was higher if two-bud cuttings were used. In contrast to fertilized plants, only one cutting type (two-bud 10 mm) grew to a sufficient height and diameter for use in poplar plantation establishment. Interestingly, the root-to-shoot ratio for this cutting type was 0.16 while the ratio for the same cutting type is 0.11 if fertilized. Together, these results suggest that most types of poplar cuttings can be used to establish poplar plantations if fertilizer is used and that the largest cutting type (two-bud 10 mm) might be more suitable to establish poplar plantations at harsh sites, thus reducing the cost of poplar plant production.

By synthesizing the available evidence, we identify positive outcomes from mixtures including increased biodiversity, water quality, esthetic and recreational values, as well as reduced stand vulnerability to pest and pathogen damage. However, some uncertainties and risks were projected to increase, highlighting the importance of conducting comprehensive interdisciplinary evaluations when assessing the pros and cons of mixtures.

  • The biodiversity contribution of wood plantations: Contrasting the bird communities of Sweden's protected and production oak forests

    • The oak-dominated woodlands and forests of northern Europe have experienced dramatic declines due to agriculture, urbanization, and conifer-dominated production forestry. These losses have had a substantial negative impact on biodiversity due to the large number of forest species which depend on oak and the environments oak-dominated forests provide. Production oak stands may serve as a means of supplementing or complementing the habitat provided by the limited remaining natural oak remnants in this region. Here we evaluate the extent to which oak plantations in temperate southern Sweden provide habitat and resources for bird communities, by surveying and contrasting the bird species composition and diversity found in mature and young production oak stands (5 and 8 replicates respectively) and protected oak-dominated remnant forests (5 replicates). The mature production stands possessed a bird community partially overlapping in bird species composition, and comparable in species richness (34 species) to that found within protected oak forests (39 species). Furthermore, the production oak forests surveyed hosted threatened or near threatened bird species, including black woodpecker (Dryocopus martius), goldcrest (Regulus regulus), starling (Sturnus vulgaris), and yellowhammer (Emberiza citrinella). Though production oak forests cannot replace the habitat provided by protected oak forests, these stands do appear to provide conditions consistent with the habitat and resource requirements of a diverse cross-section of bird species in this region, including species of substantial conservation concern. Production oak forests thus have the capacity to make a positive contribution to biodiversity conservation, as well as providing a diverse range of goods and services to society.   • Concealed by darkness: How stand density can override the biodiversity benefits of mixed forests

    • Higher tree species richness often increases the diversity of other taxonomic groups and is promoted in many managed forest systems. The reason for the higher diversity is due to, for example, inter-specific differences in resource filtering of the trees. Resource filtering also depends on the total density of the tree layer, but the interaction between tree species composition and density is, so far, unexplored. Likewise, few studies have addressed whether mixtures of tree species host higher diversity, or support higher productivity, of other taxa than monocultures of the same tree species. We use a gradient in overstory tree species composition, ranging from pure Norway spruce (Picea abies) to pure Birch (Betula sp.), combined with a gradient from open to closed forest, to assess the joint effects of tree species mixture and forest density on the understory vegetation. The cover and species richness of understory vascular plants increased with an increasing proportion of birch and decreased with increasing forest density, while the cover of bryophytes decreased with an increasing proportion of birch and increasing forest density. There were clear interactions between tree species composition and forest density; the decrease in vascular plants with forest density was stronger in forest dominated by spruce than in forests dominated by birch, and the positive effect of an increasing proportion of birch was smaller in dense than in open forests. There were no indications of mixed forests supporting a higher species richness or plant cover than any of the two monocultures. Additionally, few species (10%) showed tendencies toward a higher probability of occurrence in mixed forests, and most of these also occurred in open pure stands. Our results indicate a potential conflict between goals for diversified forests and increased biomass production, as the positive effects for understory vegetation from mixtures may be lost to concomitant increases in forest density. Furthermore, the limited number of species benefiting from mixtures per se indicates that similar biodiversity benefits for understory vegetation may be obtained at landscape levels from the increased use of broadleaf and lower-density production stands, as from mixtures.

  • Modeling birch seed supply and seedling establishment during forest regeneration

    • The seed ecology and regeneration management of birch species in Fennoscandia has been quite thoroughly investigated through decades of forestry research. Nonetheless, there are few methods for precise predictions of spontaneous natural regeneration. In this study a first framework of a model is presented that combines possible seed supply with seed emergence and seedling survival based on GIS data and the results of past experiments. The birch seed supply was calculated by combining spatial data on standing birch volume with birch seed dispersal distributions. The establishment on a site was further estimated based on the effect parameters of soil moisture conditions and soil scarification. This model shows a reduced variance compared to general means of seed supply and partly explains the variation between sites with the same soil scarification treatments. Even though additional management and ecological variables should be incorporated to increase the model's predictive ability, it could already be of use in practical forestry. (C) 2017 Elsevier B.V.   • Natural Regeneration on Planted Clearcuts—The Easy Way to Mixed Forest?

    • Establishing mixed stands by stimulating natural in combination with planting could potentially ensure adequate stocking and meet shifting demands for forest products. Two long-term experiments were separately established in former Norway spruce (Picea abies L. Karst) stands in southern Sweden to be able to evaluate mixed forest establishment and management. The treatment plots were treated with different intensity of soil scarification (disc trenching, soil inversion and control) to test effects of soil disturbance on regeneration success. Five years later, both experiments were used for a first evaluation, where seedlings of all naturally regenerated tree species were counted together with estimates of the cover and height of other vegetation. This was made in sample plots in symmetric grids. Testing the effects of scarification and distance to seed source on natural regeneration of silver birch (Betula pendula Roth.) and downy birch (B. pubescens Ehrh.) showed that both scarification type and degree of soil disturbance significantly affected seedling counts but only if seed sources were close. In conclusion, conventional management practices for regenerating coniferous forest may be suitable for establishing mixed forest stands with combinations of planted and naturally regenerated seedlings.

    • Early management of the regenerated seedlings shapes the future stand properties. To address these issues, pre-commercial thinning (PCT) and control treatments were applied to planted Norway spruce (Picea abies L. Karst) and naturally regenerated birch (Betula pendula Roth., Betula pubescens Ehrh.) stands in forest experiments in southern Sweden (lat. 56-57 N) containing 1.1-5.5 m tall saplings. The treatments were retention of 1000 or 2000 stems ha(-1) of Norway spruce, with no birch or birch at 1000 stems ha(-1). Treatments were replicated with and without annual removal of birch sprouts from stumps. The periodic annual increment (PAI) over five years was calculated for total stand volume and individual trees. The mean PAI of dominant trees was significantly higher both following all PCT treatments than controls, and following low rather than high-density PCT. Birch retention did not affect growth of the dominant trees but PAI was lower in plots with uncontrolled sprouting. The PAI of birch was significantly higher in low-density Norway spruce plots than in control plots and the high-density plots. The treatment response was significant even in stands with initial heights of only 1-2 m.
