    • A novel mutation in exon 9 of tau, I260V, is associated with a clinical syndrome consistent with frontotemporal dementia with extensive tau pathology; however, neurofibrillary tangles and Pick bodies are absent. Significantly, Sarkosyl- insoluble tau extracted from affected brain tissue consisted almost exclusively of four-repeat isoforms. Consistent with these findings, in vitro biochemical assays demonstrated that the I260V mutation causes a selective increase in tau aggregation and a decrease in tau-induced microtubule assembly with four-repeat isoforms only. The contrasting pathology and biochemical effects of this mutation suggest a different disease mechanism from the other exon 9 mutations and demonstrates the critical role for the first microtubule-binding domain in tau-promoted microtubule assembly and the pathogenic aggregation of tau.

    • The biosynthesis and processing of proinsulin was investigated in the diabetic Goto-Kakizaki (GK) rat. Immunofluorescence microscopy comparing GK and Wistar control rat pancreata revealed marked changes in the distribution of alpha-cells and pronounced beta-cell heterogeneity in the expression patterns of insulin, prohormone convertases PC1, PC2, carboxypeptidase E (CPE) and the PC-binding proteins 7B2 and ProSAAS. Western blot analyses of isolated islets revealed little difference in PC1 and CPE expression but PC2 immunoreactivity was markedly lower in the GK islets. The processing of the PC2-dependent substrate chromogranin A was reduced as evidenced by the appearance of intermediates. No differences were seen in the biosynthesis and post-translational modification of PC1, PC2 or CPE following incubation of islets in 16.7 mM glucose, but incubation in 3.3 mM glucose resulted in decreased PC2 biosynthesis in the GK islets. The rates of biosynthesis, processing and secretion of newly synthesized (pro)insulin were comparable. Circulating insulin immunoreactivity in both Wistar and GK rats was predominantly insulin 1 and 2 in the expected ratios with no (pro)insulin evident. Thus, the marked changes in islet morphology and PC2 expression did not impact the rate or extent of proinsulin processing either in vitro or in vivo in this experimental model.

    • Methods of VUV electron beam ion trap (EBIT) spectroscopy are applied to the study of Pm-like tungsten (W13+). These data show that theory appears well capable of dealing with these multi-electron (61) ions, at least for high ionization stages. A comparison of other spectroscopic methods applied to the study of other ions of the Pm I sequence is also given, and finally a search for the Pm-like W lines at the ASDEX Upgrade Tokamak is mentioned.     • The electron transfer process in the interaction of a slow highlycharged ion with a metal surface has been studied employing amicrocapillary foil as a target. The combination of the target and avisible light spectroscopy allows us to directly evaluate the initial(n,ℓ) distributions of the first transferred electrons. It wasfound that observed lines were primarily attributed to Δn=1transitions with relatively high angular momenta ( ℓi≳4 ) . A cascade analysis revealed that the electron is selectivelytransferred to n≈ qin +1 states with a narrowdistribution width of δn≈2 (full width at half maximum). Theaverage initial principal quantum number is consistent with theprediction of the classical over-barrier model.
