  • Identification of Plant Semiochemicals and Characterization of New Olfactory Sensory Neuron Types in a Polyphagous Pest Moth, Spodoptera littoralis

    • Phytophagous insects use blends of volatiles released from plants to select hosts for feeding and oviposition. To behaviorally analyze complex blends, we need efficient and selective methods for elucidating neuron types, their ligands, and specificity. Gas chromatography-combined single sensillum recordings (GC-SSRs) from antennal olfactory sensilla of female moth, Spodoptera littoralis revealed 38 physiologically active peaks in the headspace volatile blends from both larvae-damaged cotton plants and lilac flowers. Using GC-combined mass spectrometry, 9 new physiologically active compounds were identified from damaged cotton and 11 from lilac compared with earlier electrophysiological studies using antennae of female S. littoralis. We characterized 14 novel classes of olfactory sensory neurons (OSNs). Among these, we found the first 2 ligands for a frequent type of short trichoid sensillum, for which no ligands were identified earlier. By using GC-SSR, a substantial increase in functional classes of OSNs and active compounds, 40% and 34% more, respectively, compared with recent studies using GC-electroantennogram or SSR using single compounds was detected. Compared with the estimated number of corresponding antennal olfactory receptors, the OSN classes now correspond to 83% of a likely maximum. The many specialist OSNs observed may facilitate behavioral confirmation of key plant volatiles in blends.

  • Fresh, dried or smoked? Repellent properties of volatiles emitted from ethnomedicinal plant leaves against malaria and yellow fever vectors in Ethiopia

    • Background: In the search for plant-based mosquito repellents, volatile emanations were investigated from five plant species, Corymbia citriodora, Ocimum suave, Ocimum lamiifolium, Olea europaea and Ostostegia integrifolia, traditionally used in Ethiopia as protection against mosquitoes. Methods: The behaviour of two mosquitoes, the malaria vector Anopheles arabiensis and the arbovirus vector Aedes aegypti, was assessed towards volatiles collected from the headspace of fresh and dried leaves, and the smoke from burning the dried leaves in a two-choice landing bioassay and in the background of human odour. Results: Volatile extracts from the smoke of burning dried leaves were found to be more repellent than those from fresh leaves, which in turn were more repellent to mosquitoes than volatiles from dried leaves. Of all smoke and fresh volatile extracts, those from Co. citriodora (52–76%) and Oc. suave (58–68%) were found to be the most repellent, Os. integrifolia (29–56%) to be intermediate while Ol. europaea (23–40%) and Os. integrifolia (19–37%) were the least repellent. One volatile present in each of the fresh leaf extracts of Co. citriodora, Oc. suave and Os. integrifolia was ß-ocimene. The levels of ß-ocimene reflected the mosquito repellent activity of these three fresh leaf extracts. Female host-seeking mosquitoes responded dose-dependently to ß-ocimene, both physiologically and behaviourally, with a maximal behavioural repulsion at 14% ß-ocimene. ß-ocimene (14%) repels mosquitoes in our 6-minute landing assays comparable to the synthetic insect repellent N,N-diethyl-m-toluamide (10% DEET). Conclusions: Volatiles in the smoke of burning as well as fresh leaves of Co. citriodora and Oc. suave have significant repellent properties against host seeking An. arabiensis and Ae. aegypti mosquitoes. ß-ocimene, present in the fresh leaf headspace of Co. citriodora, Oc. suave and Os. integrifolia, is a significantly effective volatile mosquito repellent in the laboratory. In addition to its repellent properties, ß-ocimene has long approved safe for use in food and cosmetics, making this volatile an intriguing compound to pursue in further tests in the laboratory and field to validate its mosquito repellent activity and potential for use in a commercial product. Also, the landing bioassay with humanised membranes is a potentially useful repellent screening technique that does not require the exposure of humans to the vectors, however further tests in parallel with conventional techniques are advised

  • Evolution of mosquito preference for humans linked to an odorant receptor

    • Female mosquitoes are major vectors of human disease and the most dangerous are those that preferentially bite humans. A 'domestic' form of the mosquito Aedes aegypti has evolved to specialize in biting humans and is the main worldwide vector of dengue, yellow fever, and chikungunya viruses. The domestic form coexists with an ancestral, 'forest' form that prefers to bite non-human animals and is found along the coast of Kenya. We collected the two forms, established laboratory colonies, and document striking divergence in preference for human versus non-human animal odour. We further show that the evolution of preference for human odour in domestic mosquitoes is tightly linked to increases in the expression and ligand-sensitivity of the odorant receptor AaegOr4, which we found recognizes a compound present at high levels inhuman odour. Our results provide a rare example of a gene contributing to behavioural evolution and provide insight into how disease-vectoring mosquitoes came to specialize on humans.

    • Conclusions: The identification of mosquito repellent compounds contributes to deciphering the mechanisms underlying repulsion, supporting the rational design of novel repellents. The three mosquito repellent compounds identified in this study are structurally dissimilar, which may indicate involvement of different sensory neurons in repulsion. Repulsion may well be enhanced through combining different repellent plants (or their synthetic mimics), and can be a locally sustainable part in mosquito control efforts.

    • Mate aphids respond to short (UV) and long (green yellow) wavelength stimuli during host-plant searching behaviour. Although many aphids are attracted to yellow, the bird cherry-oat aphid, Rhopalosiphum padi, is attracted to green. As part of identifying suitable plant species for crop border plants for seed potatoes, the attraction of R. padi to different shades of green in relation to differences in spectral reflectance of three cultivars of a non-host, potato, and two host-plant species, maize and wheat, were determined. Choice experiments were carried out under laboratory conditions to evaluate aphid landing preference for stimuli of different colours. Rhopalosiphum padi alates preferred to land on the colour targets with the highest reflectance. Significantly more alates landed on yellow and lime colour targets with a maximum wavelength reflectance of 46 % from 600-610 rim and 26 % from 525-531 nm, respectively. The peak light reflectance of the crop plants ranged between 12 % (potato; 532-555 nm) and 20 % (wheat; 537-553 nm). The results on aphid landing preference for different colour targets suggest that R. padi will land in higher numbers on the wheat plants, compared to potato, due to their higher peak percentage reflectance. In addition, the study indicates that the wavelength reflectance curves of plants can be used as a characteristic to select possible crop border plants when making choices between seemingly equally suitable plant species/cultivars.

    • Culex quinquefasciatus (the southern house mosquito) is an important mosquito vector of viruses such as West Nile virus and St. Louis encephalitis virus, as well as of nematodes that cause lymphatic filariasis. C. quinquefasciatus is one species within the Culex pipiens species complex and can be found throughout tropical and temperate climates of the world. The ability of C. quinquefasciatus to take blood meals from birds, livestock, and humans contributes to its ability to vector pathogens between species. Here, we describe the genomic sequence of C. quinquefasciatus: Its repertoire of 18,883 protein-coding genes is 22% larger than that of Aedes aegypti and 52% larger than that of Anopheles gambiae with multiple gene-family expansions, including olfactory and gustatory receptors, salivary gland genes, and genes associated with xenobiotic detoxification.
