  • Autonomous last-mile delivery robots: a literature review

    • This literature review investigates how self-driving autonomous delivery robots (ADRs) impact last-mile deliveries, add value to the logistics and transport industry, and contribute to creating competitive business models. Autonomous vehicles are still a developing technology and ADRs could possibly be one of the solutions to the last-mile problem, in particular in cities and for urban freight with an increasing number of parcels to deliver. Last-mile delivery is also changing as e-commerce and more demanding customers emerge. Such development, however, faces challenges regarding infrastructure, externalities such as CO2 emissions, and shorter delivery-time requirements. This review, focused on ADRs, reveals four major themes (operations, infrastructure, regulations, and acceptance) through which we explain the barriers and benefits of using ADRs for last-mile deliveries. The review shows that the operations of ADRs can impact last-mile deliveries by lowering costs, optimising the use of time, and reducing externalities. The review also shows that the foundation of last-mile infrastructure would have to change if ADRs are to be used to a greater extent. Regulations for ADRs are still not yet in place, which makes the market somewhat confused. The acceptance of ADRs in society is another challenge because the innovation of ADRs is still new and unfamiliar. Altogether, the use of ADRs for last-mile deliveries shows great potential, based on the promising results of the articles reviewed. However, most studies on ADRs have been theoretical in nature, such as models, which highlights the need for real-world case studies and implementations.

  • Finite element model of mechanical imaging of the breast

    • Purpose: Malignant breast lesions can be distinguished from benign lesions by their mechanical properties. This has been utilized for mechanical imaging in which the stress distribution over the breast is measured. Mechanical imaging has shown the ability to identify benign or normal cases and to reduce the number of false positives from mammography screening. Our aim was to develop a model of mechanical imaging acquisition for simulation purposes. To that end, we simulated mammographic compression of a computer model of breast anatomy and lesions. Approach: The breast compression was modeled using the finite element method. Two finite element breast models of different sizes were used and solved using linear elastic material properties in open-source virtual clinical trial (VCT) software. A spherical lesion (15 mm in diameter) was inserted into the breasts, and both the location and stiffness of the lesion were varied extensively. The average stress over the breast and the average stress at the lesion location, as well as the relative mean pressure over lesion area (RMPA), were calculated. Results: The average stress varied 6.2-6.5 kPa over the breast surface and 7.8-11.4 kPa over the lesion, for different lesion locations and stiffnesses. These stresses correspond to an RMPA of 0.80 to 1.46. The average stress was 20% to 50% higher at the lesion location compared with the average stress over the entire breast surface. Conclusions: The average stress over the breast and the lesion location corresponded well to clinical measurements. The proposed model can be used in VCTs for evaluation and optimization of mechanical imaging screening strategies.

  • Multi-species multicellular life cycles

    • Textbook examples of multicellular organisms vary in their scale and complexity but are typically composed of a single species. The prevalence of entities such as lichens, however, suggest that two different species may be capable of forming a type of multi-species multicellularity-though it may not resemble its clonal counterparts. In this chapter, we consider the possibility of multi-species multicellularity and in particular its origins. Drawing upon previous studies of the evolutionary origins of clonal multicellularity, we focus on the emergence of simple reproducing groups that have the capacity to gain adaptations. We present a framework for organizing these initial multi-species group life cycles based on whether the constituent species are unicellular or multicellular and whether the groups reproduce via fragmentation or cycles of dissociation and re-association. We discuss characteristics of each type of multi-species multicellularity and representative examples to assess their likely evolutionary trajectories. Ultimately, we conclude that the multi-species groups that most resemble textbook multicellular organisms are composed of unicellular and multicellular species and reproduce via cycles of dissociation and re-association.

  • Impact of storage time prior to cryopreservation on mechanical properties of aortic homografts

    • Optimal time spans in homograft procurement are still debatable among tissue banks and needs to be further investigated. Cell viability decreases at longer preparation intervals, but the effect on collagen and elastic fibers has not been investigated to the same extent. These fibers are of importance to the homograft elasticity and strength. The objective of this study was to analyze the mechanical properties of homograft tissue at different time spans in the procurement process. Ten aortic homografts were collected at the Tissue Bank in Lund. Twelve samples were obtained from each homograft, cryopreserved in groups of three after 2–4 days, 7–9 days, 28–30 days, and 60–62 days in antibiotic decontamination. Mechanical testing was performed with uniaxial tensile tests, calculating elastic modulus, yield stress and energy at yield stress. Two randomly selected samples were assessed with light microscopy. Procurement generated a total of 120 samples, with 30 samples in each time group. Elastic modulus and yield stress was significantly higher in samples cryopreserved after 2–4 days (2.7 MPa (2.5-5.0) and 0.78 MPa (0.68-1.0)) compared to 7–9 days (2.2 MPa (2.0-2.6) and 0.53 MPa (0.46–0.69)), p = 0.008 and 0.011 respectively. Light microscopy did not show any difference in collagen and elastin at different time spans. There was a significant decrease in elastic modulus and yield stress after 7 days of decontamination at 4 °C compared to 2–4 days. This could indicate some deterioration of elastin and collagen at longer decontamination intervals. Clinical significance of these findings remains to be clarified.

  • Computer model of mechanical imaging acquisition for virtual clinical trials

    • Malignant breast tumours can be distinguished from benign lesions and normal tissue based on their mechanical properties. Our pilot studies have demonstrated the potential of using Mechanical Imaging (MI) combined with mammography to reduce recalls and false positives in breast cancer screening by more accurately identifying benign lesions. To enable further optimization of MI we propose a computer simulation of the MI acquisition, for use in a Virtual Clinical Trial (VCT) framework. VCTs are computer simulated clinical trials used to efficiently evaluate clinical imaging systems. A linear elastic finite element (FE) model of the breast under dynamic compression was implemented using an open-source FE solver. A spherical tumour (15 mm in diameter) was inserted into the simulated predominantly adipose breast. The location and stiffness of the tumour was varied. The average stress on the compressed breast surface was calculated and compared with the local average stress at the tumour location and the Relative Mean Pressure over lesion Area (RMPA) was calculated. Preliminary results were within a realistic range with an average stress on the breast (tumour) of 5.9-16.6 kPa which is in agreement with published values between 1.0 - 22.5 kPa. This corresponds to RMPA values of 0.96-2.15 depending on stiffness and location of the tumour. This can lead to more detailed validation of various MI acquisition schemes through VCTs before their use in clinical studies.

  • Spatio-temporal evolution of hydroxyapatite crystal thickness at the bone-implant interface

    • A better understanding of bone nanostructure around the bone-implant interface is essential to improve longevity of clinical implants and decrease failure risks. This study investigates the spatio-temporal evolution of mineral crystal thickness and plate orientation in newly formed bone around the surface of a metallic implant. Standardized coin-shaped titanium implants designed with a bone chamber were inserted into rabbit tibiae for 7 and 13 weeks. Scanning measurements with micro-focused small-angle X-ray scattering (SAXS) were carried out on newly formed bone close to the implant and in control mature cortical bone. Mineral crystals were thinner close to the implant (1.8 ± 0.45 nm at 7 weeks and 2.4 ± 0.57 nm at 13 weeks) than in the control mature bone tissue (2.5 ± 0.21 nm at 7 weeks and 2.8 ± 0.35 nm at 13 weeks), with increasing thickness over healing time (+30 % in 6 weeks). These results are explained by younger bone close to the implant, which matures during osseointegration. Thinner mineral crystals parallel to the implant surface within the first 100 µm close to the implant indicate that the implant affects bone ultrastructure close to the implant, potentially due to heterogeneous interfacial stresses, and suggest a longer maturation process of bone tissue and difficulty in binding to the metal. The bone growth kinetics within the bone chamber was derived from the spatio-temporal evolution of bone tissue's nanostructure, coupled with microtomographic imaging. The findings indicate that understanding mineral crystal thickness or plate orientation can improve our knowledge of osseointegration.

  • Multi-scale characterization of the spatio-temporal interplay between elemental composition, mineral deposition and remodelling in bone fracture healing

    • Bone mineralization involves a complex orchestration of physico-chemical responses from the organism. Despite extensive studies, the detailed mechanisms of mineralization remain to be elucidated. This study aims to characterize bone mineralization using an in-vivo long bone fracture healing model in the rat. The spatio-temporal distribution of relevant elements was correlated to the deposition and maturation of hydroxyapatite and the presence of matrix remodeling compounds (MMP-13). Multi-scale measurements indicated that (i) zinc is required for both the initial mineral deposition and resorption processes during mature mineral remodeling; (ii) Zinc and MMP-13 show similar spatio-temporal trends during early mineralization; (iii) Iron acts locally and in coordination with zinc during mineralization, thus indicating novel evidence of the time-events and inter-play between the elements. These findings improve the understanding of bone mineralization by explaining the link between the different constituents of this process throughout the healing time.

  • Description of same-direction car-to-bicycle crash scenarios using real-world data from Sweden, Germany, and a global crash database

    • The overall number of traffic crashes is decreasing, but the number of crashes incurring cyclist injuries is not decreasing at the same pace. Of all car-to-bicycle crashes, same-direction crashes are among the ones with the highest risk of a serious-to-fatal injury. In this study, car-to-bicycle crashes occurring when a passenger car and a bicycle are both traveling in the same direction and on the same road (without a physically separated lane) from four different real-world crash databases were investigated. The focus was on analyzing pre-crash factors such as speed and light conditions, as well as other factors such as impact configurations and cyclist injuries. Three main crash scenarios were identified among the crashes that were studied. The most common one (comprising 65%) was CS1: “continued same-direction” with no intention of turning by either road user. The other two scenarios were CS2: “the bicycle crosses the vehicle's path by turning” (16%) and CS3: “the car crosses the bicycle's path by turning” (19%). The CS1 crashes were divided into three overtaking phases: approaching and steering, passing, and returning, representing 42–44%, 41–44%, and 12–17%, respectively, of the CS1 scenario. The three crash scenarios varied in car and bicycle speeds, road type, and weather and light conditions, as well as in impact points and cyclist injuries. The analysis of different same-direction crash scenarios and overtaking phases in this study offers a novel view of same-direction crashes, providing relevant information for the design of methods for the evaluation of crash avoidance and injury mitigation measures for these scenarios.

  • Secretome screening reveals immunomodulating functions of IFNα-7, PAP and GDF-7 on regulatory T-cells

    • Regulatory T cells (Tregs) are the key cells regulating peripheral autoreactive T lymphocytes. Tregs exert their function by suppressing effector T cells. Tregs have been shown to play essential roles in the control of a variety of physiological and pathological immune responses. However, Tregs are unstable and can lose the expression of FOXP3 and suppressive functions as a consequence of outer stimuli. Available literature suggests that secreted proteins regulate Treg functional states, such as differentiation, proliferation and suppressive function. Identification of secreted proteins that affect Treg cell function are highly interesting for both therapeutic and diagnostic purposes in either hyperactive or immunosuppressed populations. Here, we report a phenotypic screening of a human secretome library in human Treg cells utilising a high throughput flow cytometry technology. Screening a library of 575 secreted proteins allowed us to identify proteins stabilising or destabilising the Treg phenotype as suggested by changes in expression of Treg marker proteins FOXP3 and/or CTLA4. Four proteins including GDF-7, IL-10, PAP and IFNα-7 were identified as positive regulators that increased FOXP3 and/or CTLA4 expression. PAP is a phosphatase. A catalytic-dead version of the protein did not induce an increase in FOXP3 expression. Ten interferon proteins were identified as negative regulators that reduced the expression of both CTLA4 and FOXP3, without affecting cell viability. A transcriptomics analysis supported the differential effect on Tregs of IFNα-7 versus other IFNα proteins, indicating differences in JAK/STAT signaling. A conformational model experiment confirmed a tenfold reduction in IFNAR-mediated ISG transcription for IFNα-7 compared to IFNα-10. This further strengthened the theory of a shift in downstream messaging upon external stimulation. As a summary, we have identified four positive regulators of FOXP3 and/or CTLA4 expression. Further exploration of these Treg modulators and their method of action has the potential to aid the discovery of novel therapies for both autoimmune and infectious diseases as well as for cancer.
