  • Characterization of humoral responses to soluble trimeric HIV gp140 from a clade A Ugandan field isolate

    • Trimeric soluble forms of HIV gp140 envelope glycoproteins represent one of the closest molecular structures compared to native spikes present on intact virus particles. Trimeric soluble gp140 have been generated by several groups and such molecules have been shown to induce antibodies with neutralizing activity against homologous and heterologous viruses. In the present study, we generated a recombinant trimeric soluble gp140, derived from a previously identified Ugandan A-clade HIV field isolate (gp140(94UG018)). Antibodies elicited in immunized rabbits show a broad binding pattern to HIV envelopes of different clades. An epitope mapping analysis reveals that, on average, the binding is mostly focused on the C1, C2, V3, V5 and C5 regions. Immune sera show neutralization activity to Tier 1 isolates of different clades, demonstrating cross clade neutralizing activity which needs to be further broadened by possible structural modifications of the clade A gp140(94UG018). Our results provide a rationale for the design and evaluation of immunogens and the clade A gp140(94UG018) shows promising characteristics for potential involvement in an effective HIV vaccine with broad activity.

    • Approaches exploiting trait distribution extremes may be used to identify loci associated with common traits, but it is unknown whether these loci are generalizable to the broader population. In a genome-wide search for loci associated with the upper versus the lower 5th percentiles of body mass index, height and waist-to-hip ratio, as well as clinical classes of obesity, including up to 263,407 individuals of European ancestry, we identified 4 new loci (IGFBP4, H6PD, RSRC1 and PPP2R2A) influencing height detected in the distribution tails and 7 new loci (HNF4G, RPTOR, GNAT2, MRPS33P4, ADCY9, HS6ST3 and ZZZ3) for clinical classes of obesity. Further, we find a large overlap in genetic structure and the distribution of variants between traits based on extremes and the general population and little etiological heterogeneity between obesity subgroups.

    • AIMS: A higher intake of fruits and vegetables has been associated with a lower risk of ischaemic heart disease (IHD), but there is some uncertainty about the interpretation of this association. The objective was to assess the relation between fruit and vegetable intake and risk of mortality from IHD in the European Prospective Investigation into Cancer and Nutrition (EPIC)-Heart study. METHODS AND RESULTS: After an average of 8.4 years of follow-up, there were 1636 deaths from IHD among 313 074 men and women without previous myocardial infarction or stroke from eight European countries. Participants consuming at least eight portions (80 g each) of fruits and vegetables a day had a 22% lower risk of fatal IHD [relative risk (RR) = 0.78, 95% confidence interval (CI): 0.65-0.95] compared with those consuming fewer than three portions a day. After calibration of fruit and vegetable intake to account for differences in dietary assessment between the participating centres, a one portion (80 g) increment in fruit and vegetable intake was associated with a 4% lower risk of fatal IHD (RR = 0.96, 95% CI: 0.92-1.00, P for trend = 0.033). CONCLUSION: Results from this large observational study suggest that a higher intake of fruits and vegetables is associated with a reduced risk of IHD mortality. Whether this association is causal and, if so, the biological mechanism(s) by which fruits and vegetables operate to lower IHD risks remains unclear.

  • Use of snus and acute myocardial infarction: pooled analysis of eight prospective observational studies

    • The use of snus (also referred to as Scandinavian or Swedish moist smokeless tobacco), which is common in Sweden and increasing elsewhere, is receiving increasing attention since considered a tobacco smoke "potential reduction exposure product". Snus delivers a high dose of nicotine with possible hemodynamic effects, but its impact on cardiovascular morbidity and mortality is uncertain. The aim of this study was to investigate whether snus use is associated with risk of and survival after acute myocardial infarction (AMI). Data from eight prospective cohort studies set in Sweden was pooled and reanalysed. The relative risk of first time AMI and 28-day case-fatality was calculated for 130,361 men who never smoked. During 2,262,333 person-years of follow-up, 3,390 incident events of AMI were identified. Current snus use was not associated with risk of AMI (pooled multivariable hazard ratio 1.04, 95 % confidence interval 0.93 to 1.17). The short-term case fatality rate appeared increased in snus users (odds ratio 1.28, 95 % confidence interval 0.99 to 1.68). This study does not support any association between use of snus and development of AMI. Hence, toxic components other than nicotine appear implicated in the pathophysiology of smoking related ischemic heart disease. Case fatality after AMI is seemingly increased among snus users, but this relationship may be due to confounding by socioeconomic or life style factors.

  • Continuum thinking and the contexts of personal information management

    • Introduction. Recent personal information management literature has underlined the significance of the contextuality of personal information and its use. The present article discusses the applicability of the records continuum model and its generalisation, continuum thinking, as a theoretical framework for explicating the overlap and evolution of the diverse contexts of personal information and their implications for personal information management practices. Method. This article is based on three empirical case studies on 1) the personal information management practices of sound artists (qualitative semi-structured interviews), 2) graduate students' management of scholarly articles (web survey of Swedish students) and 3) the management of personal archives at Swedish archival institutions (qualitative semi-structured interviews). Analysis. The empirical material was analysed using the constant comparative method (case 1), non-parametric statistical tests including Friedman's test, Kruskal-Wallis analysis, Wilcoxon-Signed Rank test, Mann-Whitney-U test and Chi-square test in SPSS version 20.0 (case 2), and thematic analysis (case 3). Results. The continuum approach and the conceptualisation of diverse contextual aspects of personal information management as axes on a Giddensian spatio-temporal continuum can help to understand the contextual changes and continuities of personal information management and use. Conclusions. The records continuum model provides a useful theoretical basis for explicating personal information management as a process of Giddensian time-space structuration similarly to how Upward argued that the theory is useful in the context of explicating the record-keeping process and, in more general terms, providing a framework for theorising particular types of document-centric management processes.

    • This paper details the project ―Borderland  Stories,‖  which focuses on national identity in various borderland regions of the Russian Federation. The authors point out that the contradiction between the pronunciation of national identity in the borderlands and the practical aspects of borders makes it particularly exciting to focus on national identity formation and daily life in the borderlands, especially in a huge country, such as the Russian Federation, that borders many different countries. Collection and analysis of the material are being conducted in 2014 and the first results are due for presentation in 2015.

    • This article examines how students' common-sense conceptualizations of “nation,” specifically the Swedish nation, operate in teaching situations in which a critical constructivist theory of “nation” is part of the curriculum. Taking its point of departure from discussions of conceptual change, this article examines how students negotiate common-sense notions and how these notions affect students' learning processes. We examine group discussions from two different undergraduate courses at a Swedish university. The article discusses obstacles and opportunities for students to understand the concept of “nation” from a constructivist perspective and to challenge the concept of “nation” as a given and natural entity. We find several discursive repertoires in the material. Three of these repertoires are scrutinized to discover how common-sense notions work. The repertories are multifaceted and can function in ways that facilitate as well as block denaturalization. The results show that common-sense understandings remain with the students even if they learn to account for constructivist perspectives. Furthermore, the results demonstrate that students' understandings of scholarly discussions of “nation” are complex and that resisting the content of a specific theory may, in some cases, be productive in terms of learning.
