  • Whole-genome sequence association analysis of blood proteins in a longitudinal wellness cohort

    • Background The human plasma proteome is important for many biological processes and targets for diagnostics and therapy. It is therefore of great interest to understand the interplay of genetic and environmental factors to determine the specific protein levels in individuals and to gain a deeper insight of the importance of genetic architecture related to the individual variability of plasma levels of proteins during adult life. Methods We have combined whole-genome sequencing, multiplex plasma protein profiling, and extensive clinical phenotyping in a longitudinal 2-year wellness study of 101 healthy individuals with repeated sampling. Analyses of genetic and non-genetic associations related to the variability of blood levels of proteins in these individuals were performed. Results The analyses showed that each individual has a unique protein profile, and we report on the intra-individual as well as inter-individual variation for 794 plasma proteins. A genome-wide association study (GWAS) using 7.3 million genetic variants identified by whole-genome sequencing revealed 144 independent variants across 107 proteins that showed strong association (P < 6 x 10(-11)) between genetics and the inter-individual variability on protein levels. Many proteins not reported before were identified (67 out of 107) with individual plasma level affected by genetics. Our longitudinal analysis further demonstrates that these levels are stable during the 2-year study period. The variability of protein profiles as a consequence of environmental factors was also analyzed with focus on the effects of weight loss and infections. Conclusions We show that the adult blood levels of many proteins are determined at birth by genetics, which is important for efforts aimed to understand the relationship between plasma proteome profiles and human biology and disease.

    • Barley malt, a product of controlled germination, has been shown to produce high levels of butyric acid in the cecum and portal serum of rats and may therefore have anti-inflammatory effects. The aim of the study was to investigate how four barley malts, caramelized and colored malts, 50-malt and 350-malt, differing in functional characteristics concerning beta-glucan content and color, affect short-chain fatty acids (SCFA), barrier function and inflammation in the hindgut of rats fed high-fat diets. Male Wistar rats were given malt-supplemented high-fat diets for four weeks. Low and high-fat diets containing microcrystalline cellulose were incorporated as controls. All diets contained 70 g kg(-1) dietary fiber. The malt-fed groups were found to have had induced higher amounts of butyric and propionic acids in the hindgut and portal serum compared with controls, while cecal succinic acid only increased to a small extent. Fat increased the mRNA expression of tight junction proteins and Toll-like receptors (TLR) in the small intestine and distal colon of the rats, as well as the concentration of some amino acids in the portal plasma, but malt seemed to counteract these adverse effects to some extent. However, the high content of advanced glycation end-products (AGE) in caramelized malt tended to prohibit the positive effects on occludin in the small intestine and plasma amino acids seen with the other malt products. In conclusion, malting seems to be an interesting process for producing foods with positive health effects, but part of these effects may be destroyed if the malt contains a high content of AGE.

  • Enhancement of few-cycle light fields for relativistic nanophotonics

    • Pulses of light that are both ultrashort and ultraintense are often generated using optical parametric amplifiers (OPA). These are capable of driving highly non-linear interactions with matter, which are interesting when studying the fundamental laws of our universe. Furthermore, they are also used in many scientific and industrial applications, such as particle accelerators, inertial-confinement nuclear fusion, and medical diagnostics and treatment. This thesis explores the diagnostic and optimization of pulses of light with extreme properties and utilizes them to drive electron acceleration.The applied light pulses with very short duration (<5 fs) and high peak power (>10 TW) are sensitive to develop spatio-temporal aberrations. These are color-dependent distortions that can significantly degrade the pulse properties, like peak-intensity, and affect their applicability. Furthermore, in most cases they are not easy to correctly diagnose, with current tools failing to provide widely applicable solutions. In this thesis, we describe a new type of spatio-temporal coupling that is especially relevant for optical parametric synthesizers (OPS), systems that coherently combine multiple OPA stages. To do this, we have contributed to the development of two methods for the characterization of such aberrations, the so-called simplified-INSIGHT and HASO multispectral. These enabled us to further improve the structure of our OPS and laser systems.We also explored the applicability of light pulses to drive relativistic electron acceleration in vacuum. To this end, an injection system using nanotips is presented, capable of inserting electrons spatially in the focus and temporally in the most intense light-cycle. This way, vacuum laser accelerated electrons of up to 14 MeV were detected using a tight focusing configuration (f#1) and their properties characterized. Furthermore, we investigated the dependence of the acceleration process when the focusing geometry is relaxed (f#3). This resulted in the unexpected outcome of similar electron energies in both cases, although the intensity was ten times reduced. This indicates that the decrease in accelerating field strength is compensated by longer acceleration lengths, which is not predicted by currently existing analytical models.

  • Simulation of Unsteady Propeller Blade Loads Using OpenFOAM

    • Unsteady propeller blade loads are important to consider in the design of the propulsion system in order to avoid problems with noise and vibrations. This is normally done through an analysis of the average wake field flowing into the propulsor in order to correctly match the different components of the propulsion system. As computational resources increase, it now starts to become possible to better study the impact on the natural wake flow unsteadiness and how this influences the behaviour of the propeller. This work is a feasibility study to investigate different computational aspects regarding the flow around a propeller operating in a ship wake. The simulations are performed in OpenFOAM 2.2.x using LES, Large Eddy Simulation.The interest concerns both how the sliding mesh implementation in OpenFOAM performs and how large variation in the propeller flow that the resolved transient wake field incurs.
