  • Marked response to cabazitaxel in prostate cancer xenografts expressing androgen receptor variant 7 and reversion of acquired resistance by anti-androgens

    • Background Taxane treatment may be a suitable therapeutic option for patients with castration-resistant prostate cancer and high expression of constitutively active androgen receptor variants (AR-Vs). The aim of the study was to compare the effects of cabazitaxel and androgen deprivation treatments in a prostate tumor xenograft model expressing high levels of constitutively active AR-V7. Furthermore, mechanisms behind acquired cabazitaxel resistance were explored. Methods Mice were subcutaneously inoculated with 22Rv1 cells and treated with surgical castration (n = 7), abiraterone (n = 9), cabazitaxel (n = 6), castration plus abiraterone (n = 8), castration plus cabazitaxel (n = 11), or vehicle and/or sham operation (n = 23). Tumor growth was followed for about 2 months or to a volume of approximately 1000 mm(3). Two cabazitaxel resistant cell lines; 22Rv1-CabR1 and 22Rv1-CabR2, were established from xenografts relapsing during cabazitaxel treatment. Differential gene expression between the cabazitaxel resistant and control 22Rv1 cells was examined by whole-genome expression array analysis followed by immunoblotting, immunohistochemistry, and functional pathway analysis. Results Abiraterone treatment alone or in combination with surgical castration had no major effect on 22Rv1 tumor growth, while cabazitaxel significantly delayed and in some cases totally abolished 22Rv1 tumor growth on its own and in combination with surgical castration. The cabazitaxel resistant cell lines; 22Rv1-CabR1 and 22Rv1-CabR2, both showed upregulation of the ATP-binding cassette sub-family B member 1 (ABCB1) efflux pump. Treatment with ABCB1 inhibitor elacridar completely restored susceptibility to cabazitaxel, while treatment with AR-antagonists bicalutamide and enzalutamide partly restored susceptibility to cabazitaxel in both cell lines. The cholesterol biosynthesis pathway was induced in the 22Rv1-CabR2 cell line, which was confirmed by reduced sensitivity to simvastatin treatment. Conclusions Cabazitaxel efficiently inhibits prostate cancer growth despite the high expression of constitutively active AR-V7. Acquired cabazitaxel resistance involving overexpression of efflux transporter ABCB1 can be reverted by bicalutamide or enzalutamide treatment, indicating the great clinical potential for combined treatment with cabazitaxel and anti-androgens.

    • Prostate cancer is a multifocal disease, but if and how individual prostate tumours influence each other is largely unknown. We therefore explored signs of direct or indirect tumour–tumour interactions in experimental models and patient samples. Low‐metastatic AT1 and high‐metastatic MatLyLu (MLL) Dunning rat prostate cancer cells were injected into separate lobes of the ventral prostate of immunocompetent rats. AT1 tumours growing in the same prostate as MLL tumours had increased tumour size and proliferation compared to AT1 tumours growing alone. In addition, the vasculature and macrophage density surrounding the AT1 tumours were increased by MLL tumour closeness. In patient prostatectomy samples, selected to contain an index tumour [tumour with the highest grade, International Society of Urological Pathology (ISUP) grade 1, 2, 3 or 4] and a low‐grade satellite tumour (ISUP grade 1), cell proliferation in low‐grade satellite tumours gradually increased with increasing histological grade of the index tumour. The density of blood vessels and CD68+ macrophages also increased around the low‐grade satellite tumour if a high‐grade index tumour was present. This suggests that high‐grade tumours, by changing the prostate microenvironment, may increase the aggressiveness of low‐grade lesions in the organ. Future studies are needed to explore the mechanisms behind tumour–tumour interactions and their clinical importance.

Boken berör också angreppssätt för att arbeta användarcentrerat med digital medieutveckling och design, samt berör projektledning för utveckling av digital medieteknik. Med denna intro­duktion till medieteknik som ämne visar boken på ett antal framväxande områden för vidare forskning inom medieteknik.Introduktion till medieteknik är sammansatt så att den med fördel kan ­användas som kurslitteratur i utbildning inom områden som medieteknik, ­medie­­produktion, design för digitala medier eller motsvarande. Boken kan läsas som en helhet, alternativt som fristående kapitel.

    • Background: Sleeping difficulties have become increasingly prevalent among adolescents and have negative consequences on their health, well-being, and education.Aim and methods: The overall aim of this thesis was to explore adolescents’ sleep and the factors that impede and promote their sleep. Study I describes the prevalence of self-reported sleep duration and sleeping difficulties and explores their associations with school stress, self-perception, and technology use among adolescents based on a questionnaire of n = 937 adolescents, aged 15–16. Study II describes reasons for sleeping difficulties as perceived by adolescents based on an open-ended question answered by n = 475 adolescents, aged 15–16. Study III illuminates adolescents’ lived experiences of sleeping difficulties based on narrative interviews with n = 16 adolescents, aged 14–15. Study IV explores adolescents’ suggestions on how their sleep could be supported, based on eight focus groups with n = 43 adolescents, aged 15–16.Results: Fifty-five percent of the adolescents slept less than the recommended eight hours per night. School stress and technology use were associated with short sleep duration, whereas school stress and self-perception were associated with sleeping difficulties (I). Stress, technology use, poor sleep habits, existential thoughts, needs, and suffering were all perceived as reasons for sleeping difficulties (II). Having sleeping difficulties makes it challenging to go through the night and cope with the following day. It also implies a feeling of being trapped by circumstances (III). To improve sleep, adolescents suggested receiving support from involved parents, achieving knowledge about on the “whys” and “hows” of sleep, and being guided into finding balance (IV).Conclusions: Sleeping difficulties affect adolescents’ daily lives and constitute a pressing health issue. In order to deal with sleeping difficulties, adolescents need to be encouraged to narrate their experiences and circumstances, be respectfully listened to and supported by their parents, school nurses, and other significant adults.

  • Tackling Digital Competence - Probing Toolkits in Teacher Education

    • This study presents results from a programming day initiative that was developed to increase digital competence among teacher students. The results show an increased understanding of materials and toolkits available, but also gather data on the teacher students own perceived digital competence and how it changed through a hands-on workshop setting. The presented case exemplifies how even a limited deployment of toolkits in teachers’ education can help problematize complex issues and provide a low threshold entry to demands around digitization.

  • The Immeasurable Math Homework : When digital education resources are put to use

    • This ongoing case study examines the use of a subscription-based digital educational resource for mathematics learning. In this case, the digital resource Skolplus (https://skolplus.se/), is used by over 60% of Sweden's primary schools and contains both exercises, games, and printed material for the primary school year FK-6. In this study, we explore how the application is perceived by teachers, parents, and students (ages 7-8). We are specifically interested in what kind of digital features our user groups perceive as important and useful. Likewise, we want to know which expectations of use were not incorporated and why. Teachers' intentions are compared with parents' descriptions of what children do in Skolplus when they use it at home, as well as where dissonance occurs during use.While previous research has shown that digital technologies enable for example representation of objects to improve mathematical learning processes, see for example Young (2017), less attention has been paid to the use of subscription-based digital solutions. One of those that has drawn attention specifically to primary schools' use of online platforms for learning mathematics is Darragh and Franke (2021) showing that "...schools face pressure to provide mathematics programs that are at the same time traditional, modern, high-tech and balanced." Based on previous research, as well as data collected during this ongoing case study, preliminary findings include the use of subscription-based solutions for mathematics learning and the role of homework. The study aims to contribute to technology-related mathematics teaching.

  • What does teachers' digtal vulnerabiltiy look like?

    • In an increasingly digitalized society cyberbullying of children and young people constitutes a high-profile problem yet there are also reports where teachers become vulnerable in the digital environment. This study aims to explore teachers' experiences of what we introduce as digital vulnerability concerning the teaching profession. An online questionnaire was developed and distributed to teachers via teachers' Facebook groups. The questionnaire was based on open-ended questions where teachers were asked to tell in their own words about their experiences of digital vulnerability, including in which situations, how they affected them, and what support they received. The data analysis was conducted based on an iterative process by qualitative content analysis and the use of ideal type situations. The purpose of the analysis was to create narratives that reflect a reduction to the most important characteristics of different experiences of teachers vulnerable in the digital environment. The contribution of the paper includes empirical examples and a theorization about teachers' digital vulnerability by ideal type situations.
