  • Meta-analysis of genome-wide association studies discovers multiple loci for chronic lymphocytic leukemia

    • Chronic lymphocytic leukemia (CLL) is a common lymphoid malignancy with strong heritability. To further understand the genetic susceptibility for CLL and identify common loci associated with risk, we conducted a meta-analysis of four genome-wide association studies (GWAS) composed of 3,100 cases and 7,667 controls with follow-up replication in 1,958 cases and 5,530 controls. Here we report three new loci at 3p24.1 (rs9880772, EOMES, P = 2.55 x 10(-11)), 6p25.2 (rs73718779, SERPINB6, P = 1.97 x 10(-8)) and 3q28 (rs9815073, LPP, P = 3.62 x 10(-8)), as well as a new independent SNP at the known 2q13 locus (rs9308731, BCL2L11, P = 1.00 x 10(-11)) in the combined analysis. We find suggestive evidence (P<5 x 10(-7)) for two additional new loci at 4q24 (rs10028805, BANK1, P = 7.19 x 10(-8)) and 3p22.2 (rs1274963, CSRNP1, P = 2.12 x 10(-7)). Pathway analyses of new and known CLL loci consistently show a strong role for apoptosis, providing further evidence for the importance of this biological pathway in CLL susceptibility.

  • Genetic studies of body mass index yield new insights for obesity biology.

    • Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

  • Genetically predicted longer telomere length is associated with increased risk of B-cell lymphoma subtypes

    • Evidence from a small number of studies suggests that longer telomere length measured in peripheral leukocytes is associated with an increased risk of non-Hodgkin lymphoma (NHL). However, these studies may be biased by reverse causation, confounded by unmeasured environmental exposures and might miss time points for which prospective telomere measurement would best reveal a relationship between telomere length and NHL risk. We performed an analysis of genetically inferred telomere length and NHL risk in a study of 10 102 NHL cases of the four most common B-cell histologic types and 9562 controls using a genetic risk score (GRS) comprising nine telomere length-associated single-nucleotide polymorphisms. This approach uses existing genotype data and estimates telomere length by weighing the number of telomere length-associated variant alleles an individual carries with the published change in kb of telomere length. The analysis of the telomere length GRS resulted in an association between longer telomere length and increased NHL risk [four B-cell histologic types combined; odds ratio (OR) = 1.49, 95% CI 1.22-1.82, P-value = 8.5 x 10(-5)]. Subtype-specific analyses indicated that chronic lymphocytic leukemia or small lymphocytic lymphoma (CLL/SLL) was the principal NHL subtype contributing to this association (OR = 2.60, 95% CI 1.93-3.51, P-value = 4.0 x 10(-10)). Significant interactions were observed across strata of sex for CLL/SLL and marginal zone lymphoma subtypes as well as age for the follicular lymphoma subtype. Our results indicate that a genetic background that favors longer telomere length may increase NHL risk, particularly risk of CLL/SLL, and are consistent with earlier studies relating longer telomere length with increased NHL risk.

  • ActiveNavigator: Toward Real-Time Knowledge Capture and Feedback in Design Workspaces

    • Knowledge capture and reuse systems, such as interactive table surfaces or smart whiteboards, have long enabled designers to review and revisit the knowledge artifacts generated by their creative work. Advances in data sensing and computation now allow near real-time analysis and feedback to be added this toolbox. In this paper, we outline our exploratory application of real-time speaker identification, audio transcription, linguistic analysis, and proactive content retrieval to design team meetings. We highlight the potential benefits and limitations of tools available to collect and analyze real-time design interactions and identify areas of future exploration for engineering educators and designers. In addition, we consider the implications of these tools for design research; automatic data analysis makes it possible to instrument several design workspaces simultaneously, increasing the chance of capturing critical moments, and increasing the opportunity to draw comparisons and contrasts across teams.

  • A principal component meta-analysis on multiple anthropometric traits identifies novel loci for body shape

    • Large consortia have revealed hundreds of genetic loci associated with anthropometric traits, one trait at a time. We examined whether genetic variants affect body shape as a composite phenotype that is represented by a combination of anthropometric traits. We developed an approach that calculates averaged PCs (AvPCs) representing body shape derived from six anthropometric traits (body mass index, height, weight, waist and hip circumference, waist-to-hip ratio). The first four AvPCs explain >99% of the variability, are heritable, and associate with cardiometabolic outcomes. We performed genome-wide association analyses for each body shape composite phenotype across 65 studies and meta-analysed summary statistics. We identify six novel loci: LEMD2 and CD47 for AvPC1, RPS6KA5/C14orf159 and GANAB for AvPC3, and ARL15 and ANP32 for AvPC4. Our findings highlight the value of using multiple traits to define complex phenotypes for discovery, which are not captured by single-trait analyses, and may shed light onto new pathways.

    • Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms.

  • Horse, Butler or Elevator? Metaphors and enactment as a catalyst for exploring interaction with autonomous technology

    • As technology becomes increasingly intelligent and progressively gains agency, the relationship between system and human is redefined. Conventional interaction design methodologies cannot fully encompass the emerging new types of relationships, and new methods are necessary to address interaction at early stages in the design process. Both design metaphors and enactment techniques have been suggested as a way forward, and this paper explores whether a combination of the two can support the design of interaction with future autonomous systems. In three workshops, 27 participants in total utilised this combination of methods to design the interaction with an autonomous vehicle. The analysis of the workshops shows that the combination of the methods manages to support the imagining and design, where the metaphors aided the creation of a joint conceptual vision of the relationship, and the enactment created tangible experiences and contextualisation of the design concepts. Nine guidelines for the use of the methods when designing intelligent systems are defined, based on the insights from the workshops.

    • Development of autonomous vehicles is progressing. As automation levels increase, the roles of both the driver and the vehicle are changing, meaning that they need to forge a new relationship to each other as the vehicle gains more agency. We believe this requires approaches that address that relationship early in the design process. One such approach is choosing a metaphor as a guiding principle for the interaction to set the preconditions for the relationship. Another approach is early evaluation of designs between system concept prototypes and the user. The aim of this one-day workshop is to explore the use of metaphors and evaluation though enactment in the design of human-vehicle interaction. This will be done through a short concept development process, where participants are asked to reflect on the process. Outcomes will be an evolved understanding of using the design approaches, as well as identified collaboration and research needs.
