    • Cyclical wind variability is an ubiquitous but as yet unexplained feature among OB stars. The O7.5 III(n)((f)) star ξ Persei is the brightest representative of this class on the Northern hemisphere. As its prominent cyclical wind properties vary on a rotational time scale (2 or 4 days) the star has been already for a long time a serious magnetic candidate. As the cause of this enigmatic behavior non-radial pulsations and/or a surface magnetic field are suggested. We present a preliminary report on our attempts to detect a magnetic field in this star with high-resolution measurements obtained with the spectropolarimeter Narval at TBL, France during 2 observing runs of 5 nights in 2006 and 5 nights in 2007. Only upper limits could be obtained, even with the longest possible exposure times. If the star hosts a magnetic field, its surface strength should be less than about 300 G. This would still be enough to disturb the stellar wind significantly. From our new data it seems that the amplitude of the known non-radial pulsations has changed within less than a year, which needs further investigation.

  • Fluorescence resonance energy transfer sensors for quantitative monitoring of pentose and disaccharide accumulation in bacteria

    • Background: Engineering microorganisms to improve metabolite flux requires detailed knowledge of the concentrations and flux rates of metabolites and metabolic intermediates in vivo. Fluorescence resonance energy transfer sensors represent a promising technology for measuring metabolite levels and corresponding rate changes in live cells. These sensors have been applied successfully in mammalian and plant cells but potentially could also be used to monitor steady-state levels of metabolites in microorganisms using fluorimetric assays. Sensors for hexose and pentose carbohydrates could help in the development of fermentative microorganisms, for example, for biofuels applications. Arabinose is one of the carbohydrates to be monitored during biofuels production from lignocellulose, while maltose is an important degradation product of starch that is relevant for starch-derived biofuels production. Results: An Escherichia coli expression vector compatible with phage. recombination technology was constructed to facilitate sensor construction and was used to generate a novel fluorescence resonance energy transfer sensor for arabinose. In parallel, a strategy for improving the sensor signal was applied to construct an improved maltose sensor. Both sensors were expressed in the cytosol of E. coli and sugar accumulation was monitored using a simple fluorimetric assay of E. coli cultures in microtiter plates. In the case of both nanosensors, the addition of the respective ligand led to concentration-dependent fluorescence resonance energy transfer responses allowing quantitative analysis of the intracellular sugar levels at given extracellular supply levels as well as accumulation rates. Conclusion: The nanosensor destination vector combined with the optimization strategy for sensor responses should help to accelerate the development of metabolite sensors. The new carbohydrate fluorescence resonance energy transfer sensors can be used for in vivo monitoring of sugar levels in prokaryotes, demonstrating the potential of such sensors as reporter tools in the development of metabolically engineered microbial strains or for real-time monitoring of intracellular metabolite during fermentation.

  • Diffuse Interstellar Bands in NGC 1448

    • We present spectroscopic VLT/UVES observations of two emerging supernovae, the Type Ia SN 2001el and the Type II SN 2003hn, in the spiral galaxy NGC 1448. Our high resolution and high signal-to-noise spectra display atomic lines of Ca II, Na I, Ti II and K I in the host galaxy. In the line of sight towards SN 2001el, we also detect over a dozen diffuse interstellar bands (DIBs) within NGC 1448. These DIBs have strengths comparable to low reddening galactic lines of sight, albeit with some variations. In particular, a good match is found with the line of sight towards the σ type diffuse cloud (HD 144217). The DIBs towards SN 2003hn are significantly weaker, and this line of sight has also lower sodium column density. The DIB central velocities show that the DIBs towards SN 2001el are closely related to the strongest interstellar Ca II and Na I components, indicating that the DIBs are preferentially produced in the same cloud. The ratio of the λ 5797 and λ 5780 DIB strengths (r ˜ 0.14) suggests a rather strong UV field in the DIB environment towards SN 2001el. We also note that the extinction estimates obtained from the sodium lines using multiple line fitting agree with reddening estimates based on the colors of the Type Ia SN 2001el.
