    • A 27-year-old woman (gravida 2, para 1) was transported to the regional university hospital in gestational week (GW) 34 + 4 due to a three-day history of fever, abdominal pain and reduced foetal movements. She had developed a dry cough one day prior to the admission (Figure S1). The woman, was slightly overweight (BMI 27 kg/m2 ) but otherwise healthy. She had normal antenatal check-ups and an obstetric ultrasound at GW 32 + 2 showed a normal foetal weight deviation of +8%1 .

  • Autoplant—Autonomous Site Preparation and Tree Planting for a Sustainable Bioeconomy

    • Sustainable forestry requires efficient regeneration methods to ensure that new forests are established quickly. In Sweden, 99% of the planting is manual, but finding labor for this arduous work is difficult. An autonomous scarifying and planting machine with high precision, low environmental impact, and a good work environment would meet the needs of the forest industry. For two years, a collaborative group of researchers, manufacturers, and users (forest companies) has worked together on developing and testing a new concept for autonomous forest regeneration (Autoplant). The concept comprises several subsystems, i.e., regeneration and route planning, autonomous driving (path planning), new technology for forest regeneration with minimal environmental impact, automatic plant management, crane motion planning, detection of planting spots, and follow-up. The subsystems were tested separately and integrated together during a field test at a clearcut. The concept shows great potential, especially from an environmental perspective, with significantly reduced soil disturbances, from approximately 50% (the area proportion of the area disturbed by disc trenching) to less than 3%. The Autoplant project highlights the challenges and opportunities related to future development, e.g., the relation between machine cost and operating speed, sensor robustness in response to vibrations and weather, and precision in detecting the size and type of obstacles during autonomous driving and planting.

  • Generalization of Deep Learning in Digital Pathology : Experience in Breast Cancer Metastasis Detection

    • Simple Summary Pathology is a cornerstone in cancer diagnostics, and digital pathology and artificial intelligence-driven image analysis could potentially save time and enhance diagnostic accuracy. For clinical implementation of artificial intelligence, a major question is whether the computer models maintain high performance when applied to new settings. We tested the generalizability of a highly accurate deep learning model for breast cancer metastasis detection in sentinel lymph nodes from, firstly, unseen sentinel node data and, secondly, data with a small change in surgical indication, in this case lymph nodes from axillary dissections. Model performance dropped in both settings, particularly on axillary dissection nodes. Retraining of the model was needed to mitigate the performance drop. The study highlights the generalization challenge of clinical implementation of AI models, and the possibility that retraining might be necessary. Poor generalizability is a major barrier to clinical implementation of artificial intelligence in digital pathology. The aim of this study was to test the generalizability of a pretrained deep learning model to a new diagnostic setting and to a small change in surgical indication. A deep learning model for breast cancer metastases detection in sentinel lymph nodes, trained on CAMELYON multicenter data, was used as a base model, and achieved an AUC of 0.969 (95% CI 0.926-0.998) and FROC of 0.838 (95% CI 0.757-0.913) on CAMELYON16 test data. On local sentinel node data, the base model performance dropped to AUC 0.929 (95% CI 0.800-0.998) and FROC 0.744 (95% CI 0.566-0.912). On data with a change in surgical indication (axillary dissections) the base model performance indicated an even larger drop with a FROC of 0.503 (95%CI 0.201-0.911). The model was retrained with addition of local data, resulting in about a 4% increase for both AUC and FROC for sentinel nodes, and an increase of 11% in AUC and 49% in FROC for axillary nodes. Pathologist qualitative evaluation of the retrained model s output showed no missed positive slides. False positives, false negatives and one previously undetected micro-metastasis were observed. The study highlights the generalization challenge even when using a multicenter trained model, and that a small change in indication can considerably impact the model s performance.

    • This study aims to develop knowledge about the teaching of literary history in upper secondary school. Sub-study I explores: What content is foregrounded when upper secondary school teachers of Swedish introduce literary history? And what significance can different teaching content have for sparking interest? Sub-study II explores: What teaching content sparks students’ interest in literary history? And what other aspects of the teaching spark their interest in literary history according to the students themselves? The study is framed by didactics and curriculum theory and also theories about interest and teaching. The material consists of video recordings from ten different lessons when upper secondary teachers were introducing literary history in the course Swedish 2. The experiences of upper secondary school students of the teaching were collected using questionnaires and interviews. All students were enrolled in university preparation programmes in a medium-sized Swedish city. The results of the analysis in Sub-study I show a wide range of potential literary history teaching content. The study thus contributes new empirical knowledge about what forms (in terms of its content) literary history can take in its teaching. In the past, Swedish as a subject has been discussed to a large extent in relation to the conceptions of the subject of Swedish as a skills subject, as a cultural heritage subject, and as an experience-based subject. The results of the study are discussed in relation to these conceptions but make further contributions by concretizing the variety of teaching content that occurs in classrooms. The results of the analysis in Sub-study II show that upper secondary school students’ interest in literary history can be sparked in relation to different content in the teaching such as students experiences; intertextuality; similarities and differences between different periods; epochs, authors and works, and aesthetic elements. The analysis also revealed aspects that could be linked to the teachers’ ways of leading and organising the teaching. These were passion and engagement; content legitimation; interaction and participation; variety; structure and delimitation and grades or de-emphasis on performance. The many themes identified in Sub-study II contribute to revealing different ways of sparking interest in literary history. This is an important result in view of the fact that previous research has shown that students lack interest in literary history teaching. Together, the two sub-studies contribute to revealing new empirical knowledge about literary history teaching. Finally, there is a reflective discussion which illuminate the study in relation to different perspectives on classroom management.

    • To develop highly competitive products, companies need to understand customer needs (CNs) by effectively gathering and analysing customer data. With the advances in Information Technology, customer data comes not only from surveys and focus groups but also from social media and networking sites. Few studies have focused on developing algorithms that are devised exclusively to help to understand customer needs from big opinion data. Topic mining, aspect-based sentiment analysis and word embedding are some of the techniques adopted to identify CNs from text data. However, most of them do not consider the possibility that part of the customer data analysed is already known by companies. With the aim to continuously enhance company understanding of CNs, this paper presents an autonomous methodology for automatically classifying a set of text data (customer sentences) as referring to known or unknown CN statements by the company. For verification purposes, an example regarding a set of customer answers from an open survey questionnaire regarding the climate system of a car is illustrated. Results indicate that the proposed methodology helps companies to validate and update the customer need database with an average of 90 % precision and 60 % recall.

    • B7-H3 (CD276), an immune checkpoint protein, is a promising molecular target for immune therapy of malignant tumours. Sufficient B7-H3 expression level is a precondition for successful therapy. Radionuclide molecular imaging is a powerful technique for visualization of expression levels of molecular targets in vivo. Use of small radiolabelled targeting proteins would enable high-contrast radionuclide imaging of molecular targets if adequate binding affinity and specificity of an imaging probe could be provided. Affibody molecules, small engineered affinity proteins based on a non-immunoglobulin scaffold, have demonstrated an appreciable potential in radionuclide imaging. Proof-of principle of radionuclide visualization of expression levels of B7-H3 in vivo was demonstrated using the [99mTc]Tc-AC12-GGGC Affibody molecule. We performed an affinity maturation of AC12, enabling selection of clones with higher affinity. Three most promising clones were expressed with a -GGGC (triglycine-cysteine) chelating sequence at the C-terminus and labelled with technetium-99m (99mTc). 99mTc-labelled conjugates bound to B7-H3-expressing cells specifically in vitro and in vivo. Biodistribution in mice bearing B7-H3-expressing SKOV-3 xenografts demonstrated improved imaging properties of the new conjugates compared with the parental variant [99mTc]Tc-AC12-GGGC. [99mTc]Tc-SYNT-179 provided the strongest improvement of tumour-to-organ ratios. Thus, affinity maturation of B7-H3 Affibody molecules could improve biodistribution and targeting properties for imaging of B7-H3-expressing tumours.

    • Radionuclide molecular imaging could provide an accurate assessment of the expression of molecular targets in disseminated cancers enabling stratification of patients for specific therapies. B7-H3 (CD276) is a transmembrane protein belonging to the B7 superfamily. This protein is overexpressed in different types of human malignancies and such upregulation is generally associated with a poor clinical prognosis. In this study, targeting properties of an Affibody-based probe, AC12, containing a -GGGC amino acid sequence as a chelator (designated as AC12-GGGC) labelled with technetium-99m (Tc-99m) were evaluated for imaging of B7-H3-expressing tumours. AC12-GGGC was efficiently labelled with Tc-99m. [Tc-99m]Tc-AC12-GGGC bound specifically to B7-H3 expressing cells in vitro with affinities in nanomolar range. In mice bearing B7-H3-expressing xenografts, [Tc-99m]Tc-AC12-GGGC showed tumour uptake of 2.1 +/- 0.5 %ID/g at 2 h after injection. Its clearance from blood, normal organs and tissues was very rapid. This new targeting agent, [Tc-99m]Tc-AC12-GGGC, provided high tumour-to-blood ratio already at 2 h (8.2 +/- 1.9), which increased to 11.0 +/- 0.5 at 4 h after injection. Significantly (p < 0.05) higher tumour-to-liver and higher tumour-to-bone ratios at 2 h in comparison with 4 h after injection were observed. Thus, [Tc-99m]Tc-AC12-GGGC could be a promising candidate for further development.
