  • Effects of lifestyle factors on concentrations of salivary cortisol in healthy individuals
  • 2009
  • Ingår i: Scandinavian Journal of Clinical & Laboratory Investigation. - : Informa UK Limited. - 1502-7686 .- 0036-5513. ; 69:2, s. 242-250
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective . Salivary cortisol is widely used in occupational health research. However, many ordinary daily activities can influence the concentrations of cortisol and the interpretation of field studies. The aim of the present study was to evaluate the effect of lifestyle factors on salivary cortisol in everyday settings. Material and methods . Healthy employees participated in one or more sub-studies on the effect of eating a vegetable salad versus protein-rich mid-day meal (n=40), drinking coffee and smoking (n=12), drinking alcohol (n=32), awakening at different times (n=29) and exercising (n=21). Cortisol in saliva was measured by radioimmunoassay (RIA). Results . When eating a mid-day meal, salivary cortisol was increased by 10 % (CI -1 % to 24 %) 1 h after eating compared to before eating in the case of both types of meal. Salivary cortisol increased by 80 % (CI 9 % to 199 %) after exercising compared to before exercise. The relative awakening response was approximately 100 % when using an alarm clock on both work-days and days off. However, the awakening response was 39 % (CI 10 % to 75 %) on a day off with spontaneous awakening. No effects of alcohol, coffee or smoking were observed. Discussion . In field studies, the biological variation in salivary cortisol may be reduced by restricting physical exercise and in collecting pre-meal samples. However, the protein content of food and moderate consumption of alcohol had no effect on concentrations of cortisol. Differences in relative awakening responses on work-days and days off are related to time and mode of awakening.

  • Intercalibration of classical and molecular techniques for identification of Alexandrium fundyense (Dinophyceae) and estimation of cell densities
  • 2007
  • Ingår i: Harmful Algae. - : Elsevier BV. - 1568-9883. ; 6:1, s. 56-72
  • Tidskriftsartikel (refereegranskat)abstract
    • A workshop with the aim to compare classical and molecular techniques for phytoplankton enumeration took place at Kristineberg Marine Research Station, Sweden, in August 2005. Seventeen different techniques - nine classical microscopic-based and eight molecular methods - were compared. Alexandrium fundyense was the target organism in four experiments. Experiment 1 was designed to determine the range of cell densities over which the methods were applicable. Experiment 2 tested the species specificity of the methods by adding Alexandrium ostenfeldii, to samples containing A. fundyense. Experiments 3 and 4 tested the ability of the methods to detect the target organism within a natural phytoplankton community. Most of the methods could detect cells at the lowest concentration tested, 100 cells L-1, but the variance was high for methods using small volumes, such as counting chambers and slides. In general, the precision and reproducibility of the investigated methods increased with increased target cell concentration. Particularly molecular methods were exceptions in that their relative standard deviation did not vary with target cell concentration. Only two of the microscopic methods and three of the molecular methods had a significant linear relationship between their cell count estimates and the A. fundyense concentration in experiment 2, where the objective was to discriminate that species from a morphologically similar and genetically closely related species. None of the investigated methods were affected by the addition of a natural plankton community background matrix in experiment 3. The results of this study are discussed in the context of previous intercomparisons and the difficulties in defining the absolute, true target cell concentration. (c) 2006 Elsevier B.V. • Settling blooms of filamentous cyanobacteria as food for meiofauna assemblages.
  • 2008
  • Ingår i: Limnology and Oceanography. - 0024-3590 .- 1939-5590. ; 53:6, s. 2636-2643
  • Tidskriftsartikel (refereegranskat)abstract
    • Summer blooms of filamentous nitrogen-fixing cyanobacteria in the Baltic Sea are normally dominated by Aphanizomenon sp. and the toxin-producing Nodularia spumigena. In a 2-week laboratory experiment, we followed the uptake by representative benthic meiofauna species of C-14-labeled organic carbon from blooms, each dominated by one of these cyanobacteria. Natural bloom material was collected and labeled by incubation with (NaHCO3)-C-14. Uptake of cyanobacterial carbon was recorded for the major meiofauna taxa living in the first-centimeter layer, namely ostracods, harpacticoids, and nematodes. The uptake rates were within the range found for diatoms in other studies, indicating that cyanobacteria may be an important food resource for the meiobenthos. The uptake of cyanobacterial carbon varied significantly among species, even within the same class. The ostracod Candona neglecta showed the highest uptake values, whereas two other ostracod species took up very little of the label. There was no significant difference in utilization of carbon from Aphanizomenon sp. and N. spumigena and no reduction in the abundance of the meiofaunal taxa analyzed compared to unexposed controls, indicating that Baltic meiofaunal assemblages in general experience no mortality when exposed to settled cyanobacteria, even the hepatotoxic N. spumigena.

  • Host selection in Tomicus piniperda L. : Composition of monoterpene hydrocarbons in relation to attack frequency in the shoot feeding phase
  • 2006
  • Ingår i: Zeitschrift fur Naturforschung - Section C Journal of Biosciences. - : Walter de Gruyter GmbH. - 0939-5075. ; 61:5-6, s. 439-444
  • Tidskriftsartikel (refereegranskat)abstract
    • The aim of this study was to investigate the host selection capacity of the pine shoot beetle, Tomicus piniperda, in the shoot-feeding phase and analyze the chiral and non-chiral host volatiles by means of GC-MS and 2D-GC in five Pinus species originating from France (Pinus sylvestris, P. halepensis, P. nigra laricio, P. pinaster maritima, P. pinaster mesogeensis). Dominating monoterpenes were (-)-α-pinene, (+)-α-pinene, (-)-β-pinene and (+)-3-carene. The amounts of the enantiomers varied considerably within and among the species. In a principal component analysis-plot, based on the absolute amounts of 18 monoterpene hydrocarbons, separation of the pine species into two groups was obtained. P. halepensis and P. sylvestris were grouped according to the amount of (+)-α-pinene and (+)-3-carene, while P. nigra laricio, P. pinaster maritima and P. pinaster mesogeensis were grouped according to (-)-α-pinene and (-)-β-pinene. P. nigra laricio was the species most attacked and P. halepensis the one least attacked by T. piniperda.

  • Molecular receptive ranges of olfactory receptor neurones responding selectively to terpenoids, aliphatic green leaf volatiles and aromatic compounds, in the strawberry blossom weevil Anthonomus rubi
  • 2005
  • Ingår i: Chemoecology. - : Springer Science and Business Media LLC. - 0937-7409 .- 1423-0445. ; 15:4, s. 211-226
  • Tidskriftsartikel (refereegranskat)abstract
    • An important question in insect-plant interactions is which of the numerous plant compounds contribute to the perception of odour qualities in herbivorous insects and are likely to be used as cues in host-searching behaviour. In order to identify which plant-produced volatiles the strawberry blossom weevil Anthonomus rubi detects, we have used electrophysiological recordings from single olfactory neurones linked to gas chromatography and mass spectrometry. We here present 15 receptor neurone types specialised for naturally produced compounds present in the host and nonhost plants and two types for two aggregation pheromone components. The active compounds were terpenoids, aromatic and aliphatic esters, alcohols and aldehydes, some of which are induced by feeding activity of the weevils. The neurones were characterised by a strong response to one or two primary odorants and weaker responses to a few others having similar chemical structure. With one exception, the molecular receptive range of each neurone type was within one chemical group. Enantiomers of linalool separated on a chiral column activated two neurone types with different enantioselectivity. Inhibition by linalool of another neurone type, excited by alpha-pinene, indicated an additional mechanism for coding the information about this compound. Altogether, detection of 54 compounds by olfactory receptor neurones is shown, of which 40 have been chemically identified in this study. Thus A. rubi has the ability to detect a large number of odorants that may be used in host selection behaviour.

  • Antifeedants in the feces of the pine weevil Hylobius abietis : Identification and biological activity
  • 2006
  • Ingår i: Journal of Chemical Ecology. - : Springer Science and Business Media LLC. - 0098-0331 .- 1573-1561. ; 32:5, s. 943-957
  • Tidskriftsartikel (refereegranskat)abstract
    • Egg-laying females of the pine weevil, Hylobius abietis (L.), regularly deposit feces adjacent to each egg. Egg cavities are gnawed in the bark of roots of recently dead conifer trees. After egg deposition, the cavity is sealed by feces and a plug of bark fragments. Root bark containing egg cavities with feces is avoided as food by pine weevils, which indicates the presence of natural antifeedants. Here we present the first results of the isolation and chemical analyses of antifeedant compounds in the feces of H. abietis. In feeding bioassays, methanol extracts of the feces revealed strong antifeedant properties. Methanol extracts were fractionated by medium-pressure liquid chromatography and the antifeedant effects were mainly found in the fractions of highest polarity. Volatile compounds in the active fractions were identified by gas chromatography-mass spectrometry (GC-MS) and the nonvolatile compounds were characterized by pyrolysis-GC-MS. Based on mass spectra, a number of compounds with various chemical structures were selected to be tested for their antifeedant properties. Antifeedant effects were found among compounds apparently originating from lignin: e.g., a methylanisol, guaiacol, veratrol, dihydroxybenzenes, and dihydroconiferyl alcohol. A weak effect by fatty acid derivatives was found. The types of naturally occurring antifeedant compounds identified in this study may become useful for the protection of planted conifer seedlings against damage by H. abietis.
