  • 3000 Years of past regional and local land-use and land-cover change in the southeastern Swedish coastal area : Early human-induced increases in landscape openness as a potential nutrient source to the Baltic Sea coastal waters

    • Reconstructions of past land use and related land-cover changes at local and regional scales are needed to evaluate the potential long-term impacts of land use on the coastal waters of the Baltic Sea. In this purpose, we selected the Gamleby area at the Swedish Baltic Sea coast for a case study. We use a new, high resolution pollen record from a small lake (Lillsjön) located 3.6 km NNW of the bay Gamlebyviken and detailed analysis of the available archeological data to reconstruct local land-use changes over the last 3000 years. To estimate land-cover change at local (2–3 km radius area) and regional (50 km radius area) scales we use four additional, published pollen records from two small and two large lakes (25–70 km S of Lillsjön) and the Landscape Reconstruction Algorithm, a pollen-vegetation modeling scheme. Results show that regional and local (small lakes Lillsjön and Hyttegöl) land-cover changes are comparable over the last 1500 years (Late Iron Age to present), and that landscape openness was much larger locally than regionally (difference of 20–40% cover over the last 500 years). The periods of largest potential impacts on the Gamlebyviken Bay from regional and local land use are 200–950 CE (Late Iron Age) and 1450 CE to present, and of lowest potential impacts 950–1450 CE. The question on whether the large landscape openness 1150–50 BCE and significant afforestation 50 BCE–200 CE reconstructed for Lillsjön’s area are characteristic of the Gamlebyviken region will require additional pollen records in the catchment area.

  • A head-to-head comparison of myocardial strain by fast-strain encoding and feature tracking imaging in acute myocardial infarction

    • BackgroundMyocardial infarction (MI) is a major cause of heart failure. Left ventricular adverse remodeling is common post-MI. Several studies have demonstrated a correlation between reduced myocardial strain and the development of adverse remodeling. Cardiac magnetic resonance (CMR) with fast-strain encoding (fast-SENC) or feature tracking (FT) enables rapid assessment of myocardial deformation. The aim of this study was to establish a head-to-head comparison of fast-SENC and FT in post-ST-elevated myocardial infarction (STEMI) patients, with clinical 2D speckle tracking echocardiography (2DEcho) as a reference. MethodsThirty patients treated with primary percutaneous coronary intervention for STEMI were investigated. All participants underwent CMR examination with late gadolinium enhancement, cine-loop steady-state free precession, and fast-SENC imaging using a 1.5T scanner as well as a 2DEcho. Global longitudinal strain (GLS), segmental longitudinal strain (SLS), global circumferential strain (GCS), and segmental circumferential strain (SCS) were assessed along with the MI scar extent. ResultsThe GCS measurements from fast-SENC and FT were nearly identical: the mean difference was 0.01 (2.5)% (95% CI - 0.92 to 0.95). For GLS, fast-SENC values were higher than FT, with a mean difference of 1.8 (1.4)% (95% CI 1.31-2.35). Tests of significance for GLS did not show any differences between the MR methods and 2DEcho. Average strain in the infarct-related artery (IRA) segments compared to the remote myocardium was significantly lower for the left anterior descending artery and right coronary artery culprits but not for the left circumflex artery culprits. Fast-SENC displayed a higher area under the curve for detecting infarcted segments than FT for both SCS and SLS. ConclusionGLS and GCS did not significantly differ between fast-SENC and FT. Both showed acceptable agreement with 2DEcho for longitudinal strain. Segments perfused by the IRA showed significantly reduced strain values compared to the remote myocardium. Fast-SENC presented a higher sensitivity and specificity for detecting infarcted segments than FT.

    • For a series of conventional soda-lime-silicate glasses with increasing Al2O3 content, we investigated the thermal, mechanical, and structural properties before and after K+-for-Na+ ion-exchange strengthening by exposure to molten KNO3. The Al-for-Si replacement resulted in increased glass network polymerization and lowered compactness. The glass transition temperature (T (g)), hardness (H) and reduced elastic modulus (E (r)), of the pristine glasses enhanced monotonically for increasing Al2O3 content. H and E (r) increased linearly up to a glass composition with roughly equal stoichiometric amounts of Na2O and Al2O3 where a nonlinear dependence on Al2O3 was observed, whereas H and E (r) of the chemically strengthened (CS) glasses revealed a strictly linear dependence. T (g), on the other hand, showed linear increase with Al-for-Si for pristine glasses while for the CS glasses a linear to nonlinear trend was observed. Solid-state Al-27 nuclear magnetic resonance (NMR) revealed the sole presence of AlO4 groups in both the pristine and CS glasses. Na-23 NMR and wet-chemical analysis manifested that all Al-bearing glasses had a lower and near-constant K+-for-Na+ ion exchange ratio than the soda-lime-silicate glass. Differential thermal analysis of CS glasses revealed a "blurred " glass transition temperature (T (g)) and an exothermic step below T (g); the latter stems from the relaxation of residual compressive stresses. The nanoindentation-derived hardness at low loads and n(M O-(2))/n(Al2O3) & AP; 1 for the CS glasses, which is attributed to an increased elastic energy recovery that is linked to the glass compactness.

  • Actuating the European Energy System Transition: Indicators for Translating Energy Systems Modelling Results into Policy-Making

    • In this paper, we define indicators, with a focus on the electricity sector, that translate the results of energy systems modelling to quantitative entities that can facilitate assessments of the transitions required to meet stringent climate targets. Such indicators, which are often overlooked in model scenario presentations, can be applied to make the modelling results more accessible and are useful for managing the transition on the policy level, as well as for internal evaluations of modelling results. We propose a set of 13 indicators related to: 1) the resource and material usages in modelled energy system designs; 2) the rates of transition from current to future energy systems; and 3) the energy security in energy system modelling results. To illustrate its value, the proposed set of indicators is applied to energy system scenarios derived from an electricity system investment model for Northern Europe. We show that the proposed indicators are useful for facilitating discussions, raising new questions, and relating the modelling results to Sustainable Development Goals and thus facilitate better policy processes. The indicators presented here should not be seen as a complete set, but rather as examples. Therefore, this paper represents a starting point and a call to other modellers to expand and refine the list of indicators.

  • Tensions experienced and balancing strategies used in Agile Software Development environments

    • To achieve efficiency in rapidly changing environments, working methods that promote change and flexibility are needed. Thus, the Agile ways of working (i.e. Agile values, principles, and frameworks) are today the most common approach to developing software and something that is currently spreading to many other industries and organizations outside of the traditional IT sector. However, it is challenging to combine the business-driven need for predictability and planning with the Agile ways of working, e.g. changeability, self-management and step-by-step development and delivery; therefore, several tensions can arise at the team and organizational levels. Tensions can, for example, be experienced if the team members’ experiences are rooted in traditional project environments with stable processes and predefined requirements based on detailed planning. This dissertation aims to contribute to our knowledge of Agile Software Development (ASD) by examining the contradictions and tensions in ASD environments, as well as how to balance and react to these tensions. The dissertation responds to the great need for conceptual development in the field of Information Systems. The target groups are both researchers and practitioners.The research reported on in this dissertation is based on one conceptual study and two empirical studies. The studies reported on in four appended papers jointly contribute toward answering three research questions (RQs):RQ 1: What is the nature of the tensions experienced in ASD environments?RQ 2: What are the sources of the tensions experienced in ASD environments?RQ 3: What ambidextrous strategies can be used to both balance and react to the tensions found in ASD environments?This dissertation brings together concepts from academic domains of knowledge (organizational theories of [paradoxical] tensions and ambidexterity and Activity Theory derived from socio-psychological theory) in order to provide a new insight into the complexity of ASD.This dissertation contributes by discussing and visualizing how patterns of contradictions, categorized tensions and ambidextrous (i.e. the ability to manage two seemingly contradictory activities) strategies have been compiled and connected. This dissertation also contributes by describing how three models that can be used to develop the concept of “shared mental models”, which is so important to team-based learning.

    • Purpose: Treating localized prostate cancer (PC) with combination radiotherapy consisting of external beam radiotherapy (EBRT) and high-dose-rate brachytherapy (HDR-BT) has been proven to result in better disease outcome than EBRT only. We aimed to evaluate the incidence of toxicities due to combination therapy and identify parameters correlated to acute or late urinary, rectal, and erectile toxicities.Material and methods: Data on symptoms and tumor/treatment parameters were collected from 359 patients treated between 2008 and 2018 with EBRT (42 Gy in 14 fractions) and HDR-BT (14.5 Gy in one fraction) for localized PC, at the Örebro University Hospital. Urinary, rectal, and erectile symptoms were presented descriptively, and bivariate analyses for correlation between grade ≥ 2 toxicity and potential predictors were performed. To evaluate prognostic models, multivariable analyses were applied.Results: Urinary toxicity grade ≥ 2 was observed in 154 patients (47% of patients without pre-existing symptoms grade ≥ 2), of which 15 were grade 3. Rectal toxicity grade 2 was observed in 22 (6%) patients. Any grade erectile dysfunction was evident in all patients without pre-existing dysfunction (n = 103), whereas only 7 recovered completely. In bivariate analyses age was correlated with higher risk of acute urinary toxicity, and irradiated volume was associated with both urinary and rectal toxicities. However, we found no multivariable model of clinical and statistical significance to predict the risk of urinary or rectal toxicities.Conclusions: In our study cohort, the severity of toxicities was in general mild or moderate and temporary, whereas the incidence of severe toxicity was considerably low. Although we found no predictive models for toxicities, our findings are reassuring that this treatment approach as curative therapy for localized PC is well-tolerated.

  • Liquid biopsy as an option for predictive testing and prognosis in patients with lung cancer

    • Background: The aim of this study was to investigate the clinical value of liquid biopsy as a primary source for variant analysis in lung cancer. In addition, we sought to characterize liquid biopsy variants and to correlate mutational load to clinical data.Methods: Circulating cell-free DNA was extracted from plasma from patients with lung cancer (n = 60) and controls with benign lung disease (n = 16). Variant analysis was performed using the AVENIO ctDNA Surveillance kit and the results were correlated to clinical and variant analysis data from tumor tissue or cytology retrieved from clinical routine diagnostics.Results: There were significantly more variants detected in lung cancer cases compared to controls (p = 0.011), but no difference between the histological subgroups of lung cancer was found (p = 0.465). Furthermore, significantly more variants were detected in patients with stage IIIb-IV disease compared to patients with stage I-IIIa (median 7 vs 4, p = 0.017). Plasma cfDNA mutational load was significantly associated with overall survival (p = 0.010). The association persisted when adjusted for stage and ECOG performance status (HR: 3.64, 95% CI 1.37-9.67, p = 0.009). Agreement between tumor and plasma samples significantly differed with stage; patients with stage IIIb-IV disease showed agreement in 88.2% of the cases with clinically relevant variants, compared to zero cases in stage I-IIIa (p = 0.004). Furthermore, one variant in EGFR, two in KRAS, and one in BRAF were detected in plasma but not in tumor samples.Conclusion: This study concludes that in the vast majority of advanced NSCLC patients a reliable variant analysis can be performed using liquid biopsy from plasma. Furthermore, we found that the number of variants in plasma is associated with prognosis, possibly indicating a strategy for closer follow up on this crucial patient group.
