  • Heterozygosity for a Loss-of-Function Mutation in GALNT2 Improves Plasma Triglyceride Clearance in Man

    • Genome-wide association studies have identified GALNT2 as a candidate gene in lipid metabolism, but it is not known how the encoded enzyme ppGalNAc-T2, which contributes to the initiation of mucin-type O-linked glycosylation, mediates this effect. In two probands with elevated plasma high-density lipoprotein cholesterol and reduced triglycerides, we identified a mutation in GALNT2. It is shown that carriers have improved postprandial triglyceride clearance, which is likely attributable to attenuated glycosylation of apolipoprotein (apo) C-III, as observed in their plasma. This protein inhibits lipoprotein lipase (LPL), which hydrolyses plasma triglycerides. We show that an apoC-III-based peptide is a substrate for ppGalNAc-T2 while its glycosylation by the mutant enzyme is impaired. In addition, neuraminidase treatment of apoC-III which removes the sialic acids from its glycan chain decreases its potential to inhibit LPL. Combined, these data suggest that ppGalNAc-T2 can affect lipid metabolism through apoC-III glycosylation, thereby establishing GALNT2 as a lipid-modifying gene.

  • Sound and vibration : effects on infants' heart rate and heart rate variability during neonatal transport

    • Aim: To measure the effect of sound and whole-body vibration on infants' heart rate and heart rate variability during ground and air ambulance transport.Methods: Sixteen infants were transported by air ambulance with ground ambulance transport to and from the airports. Whole-body vibration and sound levels were recorded and heart parameters were obtained by ECG signal.Results: Sound and whole-body vibration levels exceeded the recommended limits. Mean whole-body vibration and sound levels were 0.19m/s(2) and 73dBA, respectively. Higher whole-body vibration was associated with a lower heart rate (p<0.05), and higher sound level was linked to a higher heart rate (p=0.05). The heart rate variability was significantly higher at the end of the transport than at the beginning (p<0.01). Poorer physiologic status was associated with lower heart rate variability (p<0.001) and a lower heart rate (p<0.01). Infants wearing earmuffs had a lower heart rate (p<0.05).Conclusions: Sound and whole-body vibration during neonatal transport exceed recommended levels for adults and sound seem to have a more stressful effect on the infant than vibrations. Infants should wear earmuffs during neonatal transport because of the stress reducing effect.

  • Core-valence double photoionization of the CS2 molecule

    • Double photoionization spectra of the CS2 molecule have been recorded using the TOF-PEPECO technique in combination with synchrotron radiation at the photon energies h nu=220, 230, 240, 243, and 362.7 eV. The spectra were recorded in the S 2p and C 1s inner-shell ionization regions and reflect dicationic states formed out of one inner-shell vacancy and one vacancy in the valence region. MCSCF calculations were performed to model the energies of the dicationic states. The spectra associated with a S 2p vacancy are well structured and have been interpreted in some detail by comparison to conventional S 2p and valence photoelectron spectra. The lowest inner-shell-valence dicationic state is observed at the vertical double ionization energy 188.45 eV and is associated with a (2p(3/2))(-1)(2 pi(g))(-1) double vacancy. The spectrum connected to the C 1s vacancy shows a distinct line at 310.8 eV, accompanied by additional broad features at higher double ionization energies.     • Sex hormone-binding globulin (SHBG) is a glycoprotein responsible for the transport and biologic availability of sex steroid hormones, primarily testosterone and estradiol. SHBG has been associated with chronic diseases including type 2 diabetes (T2D) and with hormone-sensitive cancers such as breast and prostate cancer. We performed a genome-wide association study (GWAS) meta-analysis of 21,791 individuals from 10 epidemiologic studies and validated these findings in 7,046 individuals in an additional six studies. We identified twelve genomic regions (SNPs) associated with circulating SHBG concentrations. Loci near the identified SNPs included SHBG (rs12150660, 17p13.1, p=1.8×10(-106)), PRMT6 (rs17496332, 1p13.3, p=1.4×10(-11)), GCKR (rs780093, 2p23.3, p=2.2×10(-16)), ZBTB10 (rs440837, 8q21.13, p=3.4×10(-09)), JMJD1C (rs7910927, 10q21.3, p=6.1×10(-35)), SLCO1B1 (rs4149056, 12p12.1, p=1.9×10(-08)), NR2F2 (rs8023580, 15q26.2, p=8.3×10(-12)), ZNF652 (rs2411984, 17q21.32, p=3.5×10(-14)), TDGF3 (rs1573036, Xq22.3, p=4.1×10(-14)), LHCGR (rs10454142, 2p16.3, p=1.3×10(-07)), BAIAP2L1 (rs3779195, 7q21.3, p=2.7×10(-08)), and UGT2B15 (rs293428, 4q13.2, p=5.5×10(-06)). These genes encompass multiple biologic pathways, including hepatic function, lipid metabolism, carbohydrate metabolism and T2D, androgen and estrogen receptor function, epigenetic effects, and the biology of sex steroid hormone-responsive cancers including breast and prostate cancer. We found evidence of sex-differentiated genetic influences on SHBG. In a sex-specific GWAS, the loci 4q13.2-UGT2B15 was significant in men only (men p=2.5×10(-08), women p=0.66, heterogeneity p=0.003). Additionally, three loci showed strong sex-differentiated effects: 17p13.1-SHBG and Xq22.3-TDGF3 were stronger in men, whereas 8q21.12-ZBTB10 was stronger in women. Conditional analyses identified additional signals at the SHBG gene that together almost double the proportion of variance explained at the locus. Using an independent study of 1,129 individuals, all SNPs identified in the overall or sex-differentiated or conditional analyses explained ∼15.6% and ∼8.4% of the genetic variation of SHBG concentrations in men and women, respectively. The evidence for sex-differentiated effects and allelic heterogeneity highlight the importance of considering these features when estimating complex trait variance.

  • Electronically sensors system to warn drivers if a reindeer is close to the road or the railway

    • In Norway, Sweden and Finland approximately 9.000 reindeer are killed every year in traffic accidents (road and railway). In field experiments two different electronically sensor system were used. The purpose is to warn car drivers and railway engineers that reindeer are close to, or on the traffic system, and reduce the numbers of accident. The information system developed by Telespor is based on GPS/GSM radio collars. These collars use SMS to inform that the reindeer are close to the traffic system. In addition to the warning, the reindeer owners get information about the reindeer land use throughout the whole year. The collars are expensive (approx. 250 euros/unit) and need battery change every year. At Umeå University a system based on wireless sensor nodes attached to the reindeer collars is developed. This node will send out a periodic warning radio beacon. On road sticks a warning device will be attached containing a radio receiver and LED flashlights that will warn if a radio beacon is received. To further increase the warning distance it is possible to use communication between the roadside warning devices and let several devices flash their LED lights when a reindeer is detected. The radio collar (approx. 15 euros/unit) will have a theoretical battery life of more than 10 years. The radio range is around 500 meters between the radio collar and the roadside warning device, which is more than sufficient.

  • Improved Representation of Marine Stratocumulus Cloud Shortwave Radiative Properties in the CMIP5 Climate Models

    • The radiative properties of subtropical marine stratocumulus clouds are investigated in an ensemble of current-generation global climate models from phase 5 of the Climate Model Intercomparison Project (CMIP5). Using a previously documented method for determining regional mean cloud albedo, the authors find a closer agreement with observations in the CMIP5 models as compared to the previous generation of models (phase 3 of CMIP). The multimodel average indicates regional mean, monthly mean cloud albedos ranging from 0.32 to 0.5 among 26 models and five regions, to be compared with satellite observations that indicate a range from 0.32 to 0.39 for the same five regions. The intermodel spread in cloud fraction gives rise to a spread in albedo. Within models, there is a tendency for large cloud fraction to be related to low cloud albedo and vice versa, a relationship that dampens the intermodel variability in total albedo. The intramodel variability in albedo, for a given cloud fraction, is found to be up to twice as large in magnitude in models as in satellite observations. The reason for this larger variability in models is not settled, but possible contributing factors may be imperfect representation in the models of cloud type distribution or of sensitivity to meteorological variability or aerosols. Changes in aerosol loading are found to be the likely cause of an increase in cloud albedo over time. The radiative effect of such a scene brightening in marine stratocumulus cloud regions, from preindustrial times to present day, is estimated to be up to -1W m(-2) for the global ocean, but there are no observations to verify this number.

  • The Importance of Representing Mixed-Phase Clouds for Simulating Distinctive Atmospheric States in the Arctic

    • Observations from the Surface Heat Budget of the Arctic Ocean experiment (SHEBA) suggest that the Arctic Basin is characterized by two distinctly different preferred atmospheric states during wintertime. These states appear as two peaks in the frequency distribution of surface downwelling longwave radiation (LWD), representing radiatively clear and opaque conditions. Here, the authors have investigated the occurrence and representation of these states in the widely used ECMWF Interim Re-Analysis (ERA-Interim) dataset. An interannually recurring bimodal distribution of LWD values is not a clearly observable feature in the reanalysis data. However, large differences in the simulated liquid water content of clouds in ERA-Interim compared to observations are identified and these are linked to the lack of a radiatively opaque peak in the reanalysis. Using a single-column model, dynamically controlled by data from ERA-Interim, the authors show that, by tuning the glaciation speed of supercooled liquid clouds, it is possible to reach a very good agreement between the model and observations from the SHEBA campaign in terms of LWD. The results suggest that the presence of two preferred Arctic states, as observed during SHEBA, is a recurring feature of the Arctic climate system during winter [December–March (DJFM)]. The mean increase in LWD during the Arctic winter compared to ERA-Interim is 15 W m−2. This has a substantial bearing on climate model evaluation in the Arctic as it indicates the importance of representing Arctic states in climate models and reanalysis data and that doing so could have a significant impact on winter ice thickness and surface temperatures in the Arctic.

    • This paper proposes an intuitive wireless sensor/actuator based communication network for human animal interaction for a digital zoo. In order to enhance effective observation and control over wild life, we have built a wireless sensor network. 25 video transmitting nodes are installed for animal behavior observation and experimental vibrotactile collars have been designed for effective control in an animal park. The goal of our research is two-folded. Firstly, to provide an interaction between digital users and animals, and monitor the animal behavior for safety purposes. Secondly, we investigate how animals can be controlled or trained based on vibrotactile stimuli instead of electric stimuli. We have designed a multimedia sensor network for human animal machine interaction. We have evaluated the effect of human animal machine state communication model in field experiments.

    • In this paper a conceptual model for a social media game will be illustrated. The idea is to find a way to integrate our physical and digital world in a way that will involve the environment around us containing both artifacts and people and become a natural part of our daily lives. We want to show that by integrate ubiquitous information and social media in a game idea, it is possible to create a concept for a social media game application that could be used at a tourist attraction, e.g. a zoo. The value that is received from this application is both educative for the visitors and marketing for the tourist attraction. The conceptual model of the social media game builds upon the foundations of internet of things for the future version of a tourist attraction, such as the Digital Zoo.

    • In this study Moderate Resolution Imaging Spectroradiometer (MODIS) Aqua retrievals of aerosol optical thickness (AOT) at 555 nm are compared to Sun photometer measurements from Svalbard for a period of 9 years. For the 642 daily coincident measurements that were obtained, MODIS AOT generally varies within the predicted uncertainty of the retrieval over ocean (Delta AOT = +/- 0.03 +/- 0.05 . AOT). The results from the remote sensing have been used to examine the accuracy in estimates of aerosol optical properties in the Arctic, generated by global climate models and from in situ measurements at the Zeppelin station, Svalbard. AOT simulated with the Norwegian Earth System Model/Community Atmosphere Model version 4 Oslo global climate model does not reproduce the observed seasonal variability of the Arctic aerosol. The model overestimates clear-sky AOT by nearly a factor of 2 for the background summer season, while tending to underestimate the values in the spring season. Furthermore, large differences in all-sky AOT of up to 1 order of magnitude are found for the Coupled Model Intercomparison Project phase 5 model ensemble for the spring and summer seasons. Large differences between satellite/ground-based remote sensing of AOT and AOT estimated from dry and humidified scattering coefficients are found for the subarctic marine boundary layer in summer.
