  • Nanocrystalline diamond sensor targeted for selective CRP detection : An ATR-FTIR spectroscopy study

    • Protein immobilization on functionalized fluorine- terminated nanocrystalline (NCD) films was studied by attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy using an immobilization protocol developed to specifically bind C-reactive protein (CRP). Using an ATR- FTIR spectroscopy method employing a force-controlled anvil-type configuration, three critical steps of the ex situ CRP immobilization were analyzed. First, the NCD surface was passivated by deposition of a copolymer layer consisting of polyethylene oxide and polypropylene oxide. Second, a synthetic modified polypeptide binder with high affinity to CRP was covalently attached to the polymeric film. Third, CRP dissolved in aqueous buffer in concentrations of 10–20 μg/ mL was added on the functionalized NCD surface. Both the amide I and II bands, due to the polypeptide binder and CRP, were clearly observed in ATR-FTIR spectra. CRP amide I bands were extracted from difference spectra and yielded bands that agreed well with the reported amide I band of free (non-bonded) CRP in solution. Thus, our results show that CRP retains its secondary structure when it is attached to the polypeptide binders. Compared to previous IR studies of CRP in solution, about 200 times lower concentration was applied in the present study.

  • Mikrobiologiska risker vid odling av kryddor på friland och hantering till färdig produkt

    • Under primärproduktionen och vidareförädling via torkning av kryddor kan kontamination ske med sjukdomsframkallande mikroorganismer, vilket kan orsaka att personer som konsumerar produkter blir sjuka. Det är inte säkert att vidare behandling av kryddorna såsom värme sker innan konsumtion och därmed sker ingen reduktion av mikroorganismer. Kryddorna kan, om de är råvara i andra produkter och om de är kontaminerade med sjukdomsframkallande mikroorganismer, kontaminera de nya produkterna. För att minska sannolikheten att detta sker måste smittkällor och spridningsvägar kontrolleras och styras i möjligaste mån. Detta innebär att det måste finnas effektiva grundförutsättningar, det vill säga rutiner för hur odling, bevattning, gödsling, skörd, torkning, lagring med mera utförs för att minimera den mikrobiologiska kontamineringen och tillväxten av mikroorganismer på kryddorna. Om man med dessa grundförutsättningar som bas i processen utför en faroanalys och implementerar åtgärder för att styra farorna enligt HACCP-metodiken får man säkra kryddor som inte medför någon fara för slutkonsumenten oavsett användning.

  • Transfer and Decontamination of S. aureus in Transmission Routes Regarding Hands and Contact Surfaces

    • Hand hygiene, cleaning and disinfection are pre-requirements for hygiene management in hospital settings and the food industry. In order to facilitate risk management, different contamination scenarios and interventions need to be evaluated. In the present study data on transfer rates and reductions of Staphylococcus aureus were provided in an experimental set-up using artificial skin. Using this methodology, test persons were not exposed with pathogenic bacteria. An exposure assessment model was developed and applied to evaluate different contamination routes and hygiene interventions. The transfer rates of S. aureus from inoculated VITRO-SKIN® to fomites were calculated from blotting series. The VITRO-SKIN® was more prone to spread bacteria than fomites. When different surfaces were cleaned, the reduction of S. aureus varied between <1 and 7 log CFU. It could not be concluded that a certain coupon material, cleaning agent, cleaning wipe, soiling or humidity consistently resulted in a high or low reduction of S. aureus. The reduction of S. aureus and E. coli during hand washing was evaluated on artificial skin, VITRO-SKIN®. The reduction of E. coli on VITRO-SKIN® was similar to the log reduction obtained when washing human hands. The S. aureus count on a human hand was both calculated in different scenarios describing different contamination routes starting from a contaminated hand using the exposure assessment model, and measured on an experimental setup using VITRO-SKIN® for validation. A linear relationship was obtained between the analysed level of S. aureus and the calculated level. However, the calculated levels of S. aureus on the VITRO-SKIN® in the scenarios were 1–1.5 log lower than the analysed level. One of the scenarios was used to study the effect of interventions like hand washing and cleaning of surfaces.

    • PurposeThe purpose of this paper is to develop and test a concept of auditor as consigliere in family firms, that captures additional functions to monitoring, those of advice, mediating, and conveying.Design/methodology/approachThe concept is tested through a survey conducted on 309 Swedish auditors.FindingsThe data indicate that the consigliere role is generally not emphasized, indicating that auditors primarily perform the monitoring role of the audit. However, the authors do find indications of the auditor performing the consigliere role, through performing the advisory and mediating functions and, to a smaller degree, the conveying function.Research limitations/implicationsThe survey is limited in response rate and in separating governance situations from consigliere functions.Practical implicationsWith reservation for professional independence, the auditor as consigliere could be part of the governance of the family firm, but should be trained for this activity.Social implicationsRegulators should pay attention to the consigliere role when, for example, stipulating compulsory rotation of auditors.Originality/valueThe paper shows that the auditor is more than a monitor in family firms. The consigliere role, even if not at all dominating, has to be considered, at least in family firms.

    • Three Physiologically Based Pharmacokinetic software packages (GI-Sim, Simcyp (R) Simulator, and GastroPlus (TM)) were evaluated as part of the Innovative Medicine Initiative Oral Biopharmaceutics Tools project (OrBiTo) during a blinded "bottom-up" anticipation of human pharmacokinetics. After data analysis of the predicted vs. measured pharmacokinetics parameters, it was found that oral bioavailability (F-oral) was underpredicted for compounds with low permeability, suggesting improper estimates of intestinal surface area, colonic absorption and/or lack of intestinal transporter information. Foralwas also underpredicted for acidic compounds, suggesting overestimation of impact of ionisation on permeation, lack of information on intestinal transporters, or underestimation of solubilisation of weak acids due to less than optimal intestinal model pH settings or underestimation of bile micelle contribution. F-oral was overpredicted for weak bases, suggesting inadequate models for precipitation or lack of in vitro precipitation information to build informed models. Relative bioavailability was underpredicted for both high logP compounds as well as poorly water-soluble compounds, suggesting inadequate models for solubility/dissolution, underperforming bile enhancement models and/or lack of biorelevant solubility measurements. These results indicate areas for improvement in model software, modelling approaches, and generation of applicable input data. However, caution is required when interpreting the impact of drug-specific properties in this exercise, as the availability of input parameters was heterogeneous and highly variable, and the modellers generally used the data "as is" in this blinded bottom-up prediction approach.

  • Manager competences in logistics and supply chain practice

    • In the 30 years since its inception, the field, profession, and practice of logistics and supply chain management have undergone profound business transformation. This study uses shadowing and practice theory to explore the nature of manager competence in logistics and supply chain management. The results suggest that logistics and supply chain managers use business managerial, generic, and behavioral competences in practice rather than supply chain management expertise. Although the existing literature depicts competences as discrete and factor-based, the findings further reveal how managers use combinations of competences that create synergistic effects. The findings imply that the level of competence in practice extends beyond the sum of individual competences. In particular, company experience is a distinct key competence that managers constantly use in combination with other competences, and thereby has a significant effect on manager competences. The results produce four propositions for future research.

    • In Italy, the prosperous city of Reggio Emilia has become famous worldwide for its pre- schools and infant -toddler centres, attracting study groups from around the world keen to learn more about the innovative art-based processes that constitutes the Reggio Emilia Approach. We have seen in the other chapters in Part 2 of this book, how different early childhood education and care approaches have developed in response to different social and political agendas. Markedly, the socio-political pedagogical tradition of the Reggio Emilia Approach arose specifically from the municipal socialism of public life in and around the province of Reggio Emilia. Within this context the Reggio Emilia Approach is a form of progressive early childhood education shaped by Loris Malaguzzi, a psychologist and educator influenced by Dewey's model of progressive education, Montessori’s prepared environment, Vygotsky’s social cultural theories and the psychological theories of Piaget, Erikson, Bronfenbrenner, and Bruner (Cagliari et al., 2016; Rinaldi, 2006). At the same time, though, the protagonists of the approach continue to formulate new interpretations and new hypotheses and ideas about learning and teaching through their daily observations and practice of learning along with children (Gandini, 2003). This chapter begins by considering the influences that shaped the development of the Reggio Emilia Approach starting with both a geographical and historical perspective as the two are linked.
