  • Single-cell meta-analysis of SARS-CoV-2 entry genes across tissues and demographics

    • Angiotensin-converting enzyme 2 (ACE2) and accessory proteases (TMPRSS2 and CTSL) are needed for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) cellular entry, and their expression may shed light on viral tropism and impact across the body. We assessed the cell-type-specific expression of ACE2, TMPRSS2 and CTSL across 107 single-cell RNA-sequencing studies from different tissues. ACE2, TMPRSS2 and CTSL are coexpressed in specific subsets of respiratory epithelial cells in the nasal passages, airways and alveoli, and in cells from other organs associated with coronavirus disease 2019 (COVID-19) transmission or pathology. We performed a meta-analysis of 31 lung single-cell RNA-sequencing studies with 1,320,896 cells from 377 nasal, airway and lung parenchyma samples from 228 individuals. This revealed cell-type-specific associations of age, sex and smoking with expression levels of ACE2, TMPRSS2 and CTSL. Expression of entry factors increased with age and in males, including in airway secretory cells and alveolar type 2 cells. Expression programs shared by ACE2(+)TMPRSS2(+) cells in nasal, lung and gut tissues included genes that may mediate viral entry, key immune functions and epithelial-macrophage cross-talk, such as genes involved in the interleukin-6, interleukin-1, tumor necrosis factor and complement pathways. Cell-type-specific expression patterns may contribute to the pathogenesis of COVID-19, and our work highlights putative molecular pathways for therapeutic intervention. An integrated analysis of over 100 single-cell and single-nucleus transcriptomics studies illustrates severe acute respiratory syndrome coronavirus 2 viral entry gene coexpression patterns across different human tissues, and shows association of age, smoking status and sex with viral entry gene expression in respiratory cell populations.

  • CMB-S4 : Forecasting Constraints on Primordial Gravitational Waves

    • CMB-S4—the next-generation ground-based cosmic microwave background (CMB) experiment—is set to significantly advance the sensitivity of CMB measurements and enhance our understanding of the origin and evolution of the universe. Among the science cases pursued with CMB-S4, the quest for detecting primordial gravitational waves is a central driver of the experimental design. This work details the development of a forecasting framework that includes a power-spectrum-based semianalytic projection tool, targeted explicitly toward optimizing constraints on the tensor-to-scalar ratio, r, in the presence of Galactic foregrounds and gravitational lensing of the CMB. This framework is unique in its direct use of information from the achieved performance of current Stage 2–3 CMB experiments to robustly forecast the science reach of upcoming CMB-polarization endeavors. The methodology allows for rapid iteration over experimental configurations and offers a flexible way to optimize the design of future experiments, given a desired scientific goal. To form a closed-loop process, we couple this semianalytic tool with map-based validation studies, which allow for the injection of additional complexity and verification of our forecasts with several independent analysis methods. We document multiple rounds of forecasts for CMB-S4 using this process and the resulting establishment of the current reference design of the primordial gravitational-wave component of the Stage-4 experiment, optimized to achieve our science goals of detecting primordial gravitational waves for r > 0.003 at greater than 5σ, or in the absence of a detection, of reaching an upper limit of r < 0.001 at 95% CL.

    • The use of a wide range of methods for incorporating nitrogen atoms on robust catalysts has given rise to fundamental advances in the field of energy conversion and storage. Recently, nitrogen incorporation has proven to be able to fine-tune the electron densities of exposed active sites to create high-performance electrocatalysts. The preservation of a strong interface between the local atomic coordination of nitrogen atoms on bare carbon, single metal atoms, transition metal oxides, metal chalcogenides, and MXenes during synthesis plays an important role in producing an efficient electrocatalysts. In addition, the ability of nitrogen atoms to bind with carbon or metal atoms can be influenced by processing conditions. In this regard, this review is the first comprehensive overview of the range of synthetic strategies to form nitrogen incorporated catalysts and assess their chemical, structural, physical electronic property modification and their influence on electrocatalytic ORR, OER, and HER performance. This review will describe how specific strategies have been utilized to realise effective electrocatalytic systems, including the energy conversion of nitrogen incorporated catalysts, structural coordination, and material optimization. Finally, the main challenges to be considered in future investigations in order to initiate new research efforts in this promising research area are discussed.

  • Influence of Solutionising Time on the Dendrite Morphology and Mechanical Behaviour of Al-Si-Mg-Ni Hypoeutectic Alloy

    • The existing work deals with the development of Al-Si-Mg-Ni hypoeutectic alloys and to investigate the impact of solutionising time on their morphology and how these alloys behave mechanically. Al, Si, Mg, Ni of required weight percentages were melted into a clay graphite crucible and the mixture was poured into a permanent mould from which cast rods of ø32 mm ×156 mm in length were obtained. The cast rods were solutionised at 538 °C for 2, 4, 6, 8 and 10 h, respectively. Further, the alloys were then quenched into 30 °C water and subsequently aged at 155 °C for 12 h. It is perceived from the investigation that, as the solutionising time increases, a refinement in the grain of the developed alloy occurs and the hardness and tensile properties were seen increasing. Moreover, ductility was found to be low for the alloy solutionised at 10 h as compared to the LM25 alloy. The scanning electron images of the fractured surface revealed that the increase of solutionising time from 2 to 10 h led to a much smaller extent of the observance of dimples on the fractograph. Furthermore, the fractured surface of the alloy solutionised for 10 h depicted the presence of micro-voids, which confirms the ceasing of ductile behaviour because of longer solutionising time.

    • BackgroundCytosine modifications in DNA such as 5-methylcytosine (5mC) underlie a broad range of developmental processes, maintain cellular lineage specification, and can define or stratify types of cancer and other diseases. However, the wide variety of approaches available to interrogate these modifications has created a need for harmonized materials, methods, and rigorous benchmarking to improve genome-wide methylome sequencing applications in clinical and basic research. Here, we present a multi-platform assessment and cross-validated resource for epigenetics research from the FDA’s Epigenomics Quality Control Group.ResultsEach sample is processed in multiple replicates by three whole-genome bisulfite sequencing (WGBS) protocols (TruSeq DNA methylation, Accel-NGS MethylSeq, and SPLAT), oxidative bisulfite sequencing (TrueMethyl), enzymatic deamination method (EMSeq), targeted methylation sequencing (Illumina Methyl Capture EPIC), single-molecule long-read nanopore sequencing from Oxford Nanopore Technologies, and 850k Illumina methylation arrays. After rigorous quality assessment and comparison to Illumina EPIC methylation microarrays and testing on a range of algorithms (Bismark, BitmapperBS, bwa-meth, and BitMapperBS), we find overall high concordance between assays, but also differences in efficiency of read mapping, CpG capture, coverage, and platform performance, and variable performance across 26 microarray normalization algorithms.ConclusionsThe data provided herein can guide the use of these DNA reference materials in epigenomics research, as well as provide best practices for experimental design in future studies. By leveraging seven human cell lines that are designated as publicly available reference materials, these data can be used as a baseline to advance epigenomics research.

    • In the present research, hybrid Au-ZnO one-dimensional (1-D) nanostructures were grown on silicon substrates with an Al-doped ZnO (AZO) seed layer (Ultrasonic Spray Pyrolysis: USP grown) and no seed layer (NSL) using two different catalytic gold films of 2 nm and 4 nm, respectively. Consequently, such 1-D nanostructures growth was associated with the vapor-liquid-solid (VLS) and vapor-solid (VS) processes. Scanning electron microscopy (SEM) imaging analysis confirms that heat treatment triggered Au nanoparticles nucleation with varying diameters. The Au nanoparticles size and underneath seed layer texture strongly affect the morphology and aspect ratio of 1-D ZnO nanostructures. The seed layer (1-D USP) sample resulted in the growth of longer nanowires (NWs) with a high aspect ratio. The NSL sample showed the formation of nanorods (NRs) with a low aspect ratio mainly via VS growth process. X-ray diffraction (XRD), X-Ray photoelectron spectroscopy (XPS), and photoluminescence (PL) analysis also revealed the differences in the NWs and NRs properties and confirmed VLS and VS growth mechanisms. CO2 gas sensing performance at different concentrations was demonstrated, and NWs with seed layer showed a relatively higher sensing response. In contrast, NSL samples (NRs) exhibited two times faster response. A detailed gas sensing mechanism with different CO2 adsorption modes based on properties of 1D nanostructures has been discussed. Currently, CO2 sensing and capturing are critical topics in the green transition framework. The present work would be of high significance to the scientific field of NW growth and fulfill the urgent need for CO2 gas sensing.

    • The workability study of the composites enhances the understanding of the degree of plastic deformation that can be employed on it. The current research work highlights the response of the low-cost aluminum composites reinforced with exhausted alkaline battery powders under quasi-static compression. The effect of reinforcements and aspect ratio against the strain hardening exponent and strength coefficients were investigated. The microstructural changes after quasi-static compression were studied and related to the changes in the property of the composites. The composite with 6 wt.% of reinforcement showed the least amount of porosity as 1.2%. In most of the cases, the maximum value of average strain hardening exponent with respect to axial strain was noted in the composites with 6 wt. % of reinforcement. The lowest aspect ratio of 0.5 showed the maximum workability in the composites. The average strength coefficient was found to be maximum (308.58 MPa) in the composite with 2 wt.% reinforcement. The elongated grains and slip bands were observed in the microstructure of the compressed specimens.

  • New kind of echo from quantum black holes

    • We propose that a quantum black hole can produce a new kind of late-time gravitational echoes, facilitated by a near-horizon process analogous to Andreev reflection in condensed matter systems. In comparison to the traditional echo scenarios where the near-horizon region is treated as an ordinary reflector, we argue that, consequent to near-horizon gravitational scattering, this region is better described by an Andreev reflector. Such interactions lead to a novel contribution to gravitational echoes with a characteristic phase difference, an effect which is analogous to how Andreev reflections lead to propagating particlelike and holelike components with a relative phase in certain condensed matter scenarios. Moreover, this novel contribution to the echo signal encodes information about the "near-horizon quantum state," hence offering a possible new window to probe the quantum nature of black holes.
