    • Postmortem human brain tissue is widely used in neuroscience research, but use of tissue originating from different brain bank centers is considered inaccurate because of possible heterogeneity in sample quality. There is thus a need for well-characterized markers to assess the quality of postmortem brain tissue. Toward this aim, we determined tryptophan (TRP) concentrations, phosphofructokinase-1 and glutamate decarboxylase activities in 119 brain tissue samples. These neurochemical parameters were tested in samples from autopsied individuals, including control and pathological cases provided by 10 different brain bank centers. Parameters were assessed for correlation with agonal state, postmortem interval, age and gender, brain region, preservation and freezing methods, storage conditions and storage time, RNA integrity, and tissue pH value. TRP concentrations were elevated significantly (p = 0.045) with increased postmortem interval; which might indicate increased protein degradation. Therefore, TRP concentration might be one useful and convenient marker for estimating the quality of human postmortem brain tissue.

  • Generic classification of the Verrucariaceae (Ascomycota) based on molecular and morphological evidence : recent progress and remaining challenges

    • Recent molecular phylogenetic analyses and morphological Studies have shown that it is necessary to revise the present morphology-based generic delineation of the lichen family Verrucariaceae in order to account for evolutionary relatedness between species. Consequently, several genera were recently described or resurrected, and others were re-circumscribed. As an additional step toward this generic revision, three new genera (Hydropunctaria, Parabagliettoa, Wahlenbergiella) and eleven new combinations are proposed here. A summary of the Current taxonomic and morphological circumscription of all genera investigated so far is also presented. Several monophyletic groups are identified for which further taxonomical changes will be required, but for which taxon and gene sampling is presently viewed as insufficient. Clear morphological synapomorphies were found to be rare for newly delimited genera. In some cases (reduced morphology or plesiomorphism), even the combinations of slightly homoplasious phenotypic characters do not allow a clear morphological generic circumscription. Molecular features are envisioned as characters for delimiting these taxa.

  • Changes in Hg fractionation in soil induced by willow

    • This study investigated the effect of willow (Salix viminalis ×  S. schwerinii) on soil characteristics, including changes in Hg fractionation in the soil solid phase, and Hg accumulation and distribution in pot-grown plants cultivated for 32 and 76 days in aged Hg-contaminated soil (30 mg Hg kg−1DW). Changes in soil pH and organic carbon content as well as in Hg fractionation were monitored in both rhizospheric soil and in soil without plants. Mercury fractionation was performed by a 5-step sequential soil extraction procedure. Organic carbon content increased while pH decreased in the rhizospheric soil. Both chemically defined exchangeable Hg (0.1) and Hg bound to humic and fulvic acids (1.1) decreased in the rhizospheric soil, whereas plant accumulation of Hg increased with cultivation time. The sum of the decrease of these two soil Hg fractions after 76 days of cultivation was approximately equal to the amount of Hg accumulated in plants. Furthermore, the major Hg fractions (Hg bound to residual organic matter (53), sulphides (43), and the residual fraction (2.5)) remained stable. Neither whole plant accumulation of Hg from the soil, approximately 0.2 of total Hg in soil after 76 days cultivation, nor the fraction of total plant Hg in the shoots, which accounted for about 3 of the total plant Hg pool regardless of the cultivation time, were high. The overall results suggest that plants might be suitable for phytostabilization of aged Hg-contaminated soil, where root systems trap bioavailable Hg and help to control both leaching of Hg and re-entrainment of Hg-containing particulates from a contaminated site.
