    • Cell death is essential for a plethora of physiological processes, and its deregulation characterizes numerous human diseases. Thus, the in-depth investigation of cell death and its mechanisms constitutes a formidable challenge for fundamental and applied biomedical research, and has tremendous implications for the development of novel therapeutic strategies. It is, therefore, of utmost importance to standardize the experimental procedures that identify dying and dead cells in cell cultures and/or in tissues, from model organisms and/or humans, in healthy and/or pathological scenarios. Thus far, dozens of methods have been proposed to quantify cell death-related parameters. However, no guidelines exist regarding their use and interpretation, and nobody has thoroughly annotated the experimental settings for which each of these techniques is most appropriate. Here, we provide a nonexhaustive comparison of methods to detect cell death with apoptotic or nonapoptotic morphologies, their advantages and pitfalls. These guidelines are intended for investigators who study cell death, as well as for reviewers who need to constructively critique scientific reports that deal with cellular demise. Given the difficulties in determining the exact number of cells that have passed the point-of-no-return of the signaling cascades leading to cell death, we emphasize the importance of performing multiple, methodologically unrelated assays to quantify dying and dead cells.

    • A special kind of microstructured optical fiber is proposed and fabricated in which, in addition to the holey region (solid core and silica-air cladding), two large holes exist for electrode insertion. Either Bi-Sn or Au-Sn alloys were selectively inserted into the large holes forming two parallel, continuous and homogeneous internal electrodes. We demonstrate the production of a monolithic device and its use to externally control some of the guidance properties (e. g. polarization) of the fiber.

  • GTPOM: Thermo-economic optimization of whole gas turbine plant

    • Trends towards distributed power generation and the deregulation of energy markets are increasing the requirement for software tools that optimize power generation plant design and operation. In this context, this paper describes the GTPOM (thermo-economic optimization of whole gas turbine plant) European project, funded in part through the European Commissions 5th Framework Programme, focusing on the development and demonstration of an original software tool for the thermo-economic analysis and optimization of conventional and advanced energy systems based on gas turbine plant. PSEconomy, the software tool developed during the GTPOM project, provides a thermo-economic optimization capability for advanced and more-conventional energy systems, enabling the complex trade-offs between system performance and installed costs to be determined for different operational duties and market scenarios. Furthermore, the code is capable of determining the potential benefits of innovative cycles or layout modifications to existing plants compared with current plant configurations. The economic assessment is performed through a complete through-life cycle cost analysis, which includes the total capital cost of the plant, the cost of fuel, O&M costs and the expected revenues from the sale of power and heat. The optimization process, carried out with a GA-based algorithm, is able to pursue different objective functions as specified by the User. These include system efficiency, through-life cost of electricity and through-life internal rate of return. Three case studies demonstrating the capabilities of the new tool are presented in this paper covering a conventional combined cycle system, a biomass plant and a CO2 sequestration gas turbine cycle. The software code is now commercially available and is expected to provide significant advantages in the near and long-term development of energy cycles.

  • Detecting violations of sensory expectancies following cerebellar degeneration

    • Two hypotheses concerning cerebellar function and predictive behavior are the sensory prediction hypothesis and the timing hypothesis. The former postulates that the cerebellum is critical in generating expectancies regarding forthcoming sensory information. The latter postulates that this structure is critical in generating expectancies that are precisely timed; for example, the expected duration of an event or the time between events. As such, the timing hypothesis constitutes a more specific form of prediction. The present experiment contrasted these two hypotheses by examining the mismatch negativity (MMN) response in patients with cerebellar cortical atrophy and matched controls. While watching a silent movie, a stream of task-irrelevant sounds was presented. A standard sound was presented 60% of the time, whereas the remaining sounds deviated from the standard on one of four dimensions: duration, intensity, pitch, or location. The timing between stimuli was either periodic or aperiodic. Based on the sensory prediction hypothesis, the MMN for the patients should be abnormal across all four dimensions. In contrast, the timing hypothesis would predict a selective impairment of the duration MMN. Moreover, the timing hypothesis would also predict that the enhancement of the MMN observed in controls when the stimuli are presented periodically should be attenuated in the patients. Compared to controls, the patients exhibited a delayed latency in the MMN to duration deviants and a similar trend for the intensity deviants, while pitch and location MMNs did not differ between groups. Periodicity had limited and somewhat inconsistent effects. The present results are at odds with a general role for the cerebellum in sensory prediction and provide partial support for the timing hypothesis.

  • Volcanic stratigraphy of large-volume silicic pyroclastic eruptions during Oligocene Afro-Arabian flood volcanism in Yemen

    • A new stratigraphy for bimodal Oligocene flood volcanism that forms the volcanic plateau of northern Yemen is presented based on detailed field observations, petrography and geochemical correlations. The > 1 km thick volcanic pile is divided into three phases of volcanism: a main basaltic stage ( 31 to 29.7 Ma), a main silicic stage ( 29.7 to 29.5 Ma), and a stage of upper bimodal volcanism ( 29.5 to 27.7 Ma). Eight large-volume silicic pyroclastic eruptive units are traceable throughout northern Yemen, and some units can be correlated with silicic eruptive units in the Ethiopian Traps and to tephra layers in the Indian Ocean. The silicic units comprise pyroclastic density current and fall deposits and a caldera-collapse breccia, and they display textures that unequivocally identify them as primary pyroclastic deposits: basal vitrophyres, eutaxitic fabrics, glass shards, vitroclastic ash matrices and accretionary lapilli. Individual pyroclastic eruptions have preserved on-land volumes of up to similar to 850 km(3). The largest units have associated co-ignimbrite plume ash fall deposits with dispersal areas > 1 x 10(7) km(2) and estimated maximum total volumes of up to 5,000 km(3), which provide accurate and precisely dated marker horizons that can be used to link litho-, bio- and magnetostratigraphy studies. There is a marked change in eruption style of silicic units with time, from initial large-volume explosive pyroclastic eruptions producing ignimbrites and near-globally distributed tuffs, to smaller volume (< 50 km(3)) mixed effusive-explosive eruptions emplacing silicic lavas intercalated with tuffs and ignimbrites. Although eruption volumes decrease by an order of magnitude from the first stage to the last, eruption intervals within each phase remain broadly similar. These changes may reflect the initiation of continental rifting and the transition from pre-break-up thick, stable crust supporting large-volume magma chambers, to syn-rift actively thinning crust hosting small-volume magma chambers.
