  • Synthesising of a model composite in the oxide/oxide system : alumina fibre reinforced alumina matrix

    • Ceramic matrix composites have attracted big interest as potential material for high temperature applications. To avoid one of the main problems, namely oxidation at higher temperatures, interest has increased for systems where both reinforcement and matrix material are ceramic oxides. This work presents a possible method of producing long fibre reinforced ceramic matrix composites. The method was applied to single crystal aluminium oxide fibres in a polycrystalline alumina matrix and involves cladding individual fibres with the matrix material in the form of pre-sintered tubes with appropriately small diameter. The composite is then hot isostatically pressed (HIP), by putting several fibre-tube pairs in a container of required diameter. A HIP cycle was found that led to full densification of the composite without visual damage to the fibres. The method produces composites with an ideal uniform fibre spacing; samples of cylindrical shape suitable, for example, for uniaxial testing can be produced

    • The molecular interactions of the Src homology 2 (SH2) domain and the N-terminal proline-rich sequence motifs (pro-1 to pro-5) of the SH2 protein Shb with other components were presently characterised. Using a degenerate phosphopeptide library the preferred binding site for the Shb SH2 domain was determined to pTyr-Thr/Val/Ile-X-Leu at positions +1 to +3 relative the phosphotyrosine residue. Experiments with competing peptides and platelet-derived growth factor (PDGF) beta-receptor mutants with Y to F substitutions in autophosphorylation sites revealed multiple binding sites for the Shb SH2 domain in the receptor. The Shb SH2 domain also binds to in vitro phosphorylated fibroblast growth factor receptor-1 (FGFR-1) mainly through position Y776. The receptor experiments suggest that other residues besides the +1 to +3 positions may also be of significance for Shb binding. The pro-4/pro-5 motif of Shb binds in vitro particularly well to the Src, p85 alpha PI3-kinase and Eps8 SH3 domains expressed as GST fusion proteins. However, the GST-SH3 domain fusion proteins tested bind in vitro to peptides corresponding to the pro-1 to pro-5 motifs of Shb with low affinity and selectivity, suggesting that sequences outside the core proline motif may also be important for Shb-SH3 domain interactions. In vivo association between Shb-SH3 domain proteins v-Src and Eps8 was detected by coimmunoprecipitation. PDGF treatment did not affect the association between Eps8 and Shb. The data suggest that Shb is an adaptor protein linking SH3 domain proteins to tyrosine kinases or other tyrosine phosphorylated proteins.

  • Testing an oral assessment guide during chemotherapy treatment in a Swedish care setting: a pilot study

    • Oral complications are common in patients with haematological malignancies who undergo chemotherapy treatment. A pilot study including 16 haematological patients was carried out to evaluate the oral status using an Oral Assessment Guide (OAG) and to test the reliability of the OAG. The oral assessments were made daily by registered nurses at a Department of Internal Medicine in Sweden. Once a week a dental hygienist made the oral assessments independent of the registered nurses in order to provide data for calculations of inter-rater reliability. All patients had varying degrees of alterations in the oral cavity, especially in the mucous membranes, teeth/dentures and gums. The inter-rater agreement between the nurses and the dental hygienist was good for saliva and swallow, and moderate for voice and gums. Assessments to detect alterations in the oral cavity afford the opportunity for early and individualized interventions and may decrease the risk of oral infections. It is necessary to train the nurses to ensure high levels of reliability in the oral assessments. The OAG seems to be a reliable and clinical useful tool for assessing the oral cavity status and determining changes.

  • A microwave applicator for on line wood drying : Temperature and moisture distribution in wood

    • An especially designed open microwave applicator was analysed using wood as the material to be heated and dried. The idea was to develop an on line microwave construction consisting of several small open applicators, each fed by a small standard magnetron (for example 1.4 kW main power). The process was analysed by measuring the wood temperature during heating using an IR-camera and detecting the moisture distribution during drying by CT-scanning. Pine and birch wood samples were used in the experiments, mainly 40 mm in thickness. The experiments show that the power distribution differs between dry wood and moist wood. The analysis of the temperature fields captured by the IR-camera during the first minutes allows a rather accurate determination of the MW power. Consequently, the drying proceeds unevenly in the wood specimens, especially in the longitudinal direction. The dimensions of the applicator and its relation to the wood dimension are very important. However, the wood was not destroyed, the temperature and moisture gradients did not affect the wood in terms of checks or deformations. The drying rate in different positions of the specimen varied between 0.30 and 0.80 percentage moisture content/min. The uneven energy, meaning temperature and field distribution, is to be compensated in the future by a moving wood load and by alternating the position of each applicator in a larger scale microwave pilot plant.

    • Intracellular application of the sulfonylurea tolbutamide during whole-cell patch-clamp recordings stimulated exocytosis >5-fold when applied at a cytoplasmic Ca2+ concentration of 0.17 microM. This effect was not detectable in the complete absence of cytoplasmic Ca2+ and when exocytosis was elicited by guanosine 5'-O-(3-thiotriphosphate) (GTPgammaS). The stimulatory action could be antagonized by the sulfonamide diazoxide, by the Cl--channel blocker 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS), by intracellular application of the antibody JSB1 [originally raised against a 170-kDa multidrug resistance (mdr) protein], and by tamoxifen (an inhibitor of the mdr- and volume-regulated Cl- channels). Immunocytochemistry and Western blot analyses revealed that JSB1 recognizes a 65-kDa protein in the secretory granules. This protein exhibited no detectable binding of sulfonylureas and is distinct from the 140-kDa sulfonylurea high-affinity sulfonylurea receptors also present in the granules. We conclude that (i) tolbutamide stimulates Ca2+-dependent exocytosis secondary to its binding to a 140-kDa high-affinity sulfonylurea receptor in the secretory granules; and (ii) a granular 65-kDa mdr-like protein mediates the action. The processes thus initiated culminate in the activation of a granular Cl- conductance. We speculate that the activation of granular Cl- fluxes promotes exocytosis (possibly by providing the energy required for membrane fusion) by inducing water uptake and an increased intragranular hydrostatic pressure.

    • The safe application of ionising radiation for diagnosis and therapy requires a high level of knowledge of the underlying processes and of quality assurance. Sophisticated modern equipment can be used effectively for complicated diagnostic and therapeutic techniques only with adequate physics support. In the light of recent analyses and recommendations by national and international societies a joint working group of representatives from ESTRO (European Society for Therapeutic Radiology and Oncology) and from EFOMP (European Federation of Organisations for Medical Physics) was set up to assess the necessary staffing levels for physics support to radiotherapy. The method used to assess the staffing levels, the resulting recommendations and examples of their practical application are described.
