  • Multi-Objective Optimization of Organic Rankine Cycle Power Plants Using Pure and Mixed Working Fluids

    • For zeotropic mixtures, the temperature varies during phase change, which is opposed to the isothermal phase change of pure fluids. The use of such mixtures as working fluids in organic Rankine cycle power plants enables a minimization of the mean temperature difference of the heat exchangers, which is beneficial for cycle performance. On the other hand, larger heat transfer surface areas are typically required for evaporation and condensation when zeotropic mixtures are used as working fluids. In order to assess the feasibility of using zeotropic mixtures, it is, therefore, important to consider the additional costs of the heat exchangers. In this study, we aim at evaluating the economic feasibility of zeotropic mixtures compared to pure fluids. We carry out a multi-objective optimization of the net power output and the component costs for organic Rankine cycle power plants using low-temperature heat at 90 ◦C to produce electrical power at around 500 kW. The primary outcomes of the study are Pareto fronts, illustrating the power/cost relations for R32, R134a and R32/R134a (0.65/0.35mole). The results indicate that R32/R134a is the best of these fluids, with 3.4 % higher net power than R32 at the same total cost of 1200 k$.

  • The microINR portable coagulometer: analytical quality and user-friendliness of a PT (INR) point-of-care instrument

    • Regular measurement of prothrombin time as an international normalized ratio PT (INR) is mandatory for optimal and safe use of warfarin. Scandinavian evaluation of laboratory equipment for primary health care (SKUP) evaluated the microINR portable coagulometer (microINR((R))) (iLine Microsystems S.L., Spain) for measurement of PT (INR). Analytical quality and user-friendliness were evaluated under optimal conditions at an accredited hospital laboratory and at two primary health care centres (PHCCs). Patients were recruited at the outpatient clinic of the Laboratory of Medical Biochemistry, St Olavs University Hospital, Trondheim, Norway (n=98) and from two PHCCs (n=88). Venous blood samples were analyzed under optimal conditions on the STA-R((R))Evolution with STA-SPA+reagent (Stago, France) (Owren method), and the results were compared to capillary measurements on the microINR((R)). The imprecision of the microINR((R)) was 6% (90% CI: 5.3-7.0%) and 6.3% (90% CI: 5.1-8.3) in the outpatient clinic and PHCC2, respectively for INR 2.5. The microINR((R)) did not meet the SKUP quality requirement for imprecision 5.0%. For INR amp;lt;2.5 at PHCC2 and at both levels in PHCC1, CV% was 5.0. The accuracy fulfilled the SKUP quality goal in both outpatient clinic and PHCCs. User-friendliness of the operation manual was rated as intermediate, defined by SKUP as neutral ratings assessed as neither good nor bad. Operation facilities was rated unsatisfactory, and time factors satisfactory. In conclusion, quality requirements for imprecision were not met. The SKUP criteria for accuracy was fulfilled both at the hospital and at the PHCCs. The user-friendliness was rated intermediate.

  • ADAMTS9 Regulates Skeletal Muscle Insulin Sensitivity Through Extracellular Matrix Alterations

    • The ADAMTS9 rs4607103 C allele is one of the few gene variants proposed to increase the risk of type 2 diabetes through an impairment of insulin sensitivity. We show that the variant is associated with increased expression of the secreted ADAMTS9 and decreased insulin sensitivity and signaling in human skeletal muscle. In line with this, mice lacking Adamts9 selectively in skeletal muscle have improved insulin sensitivity. The molecular link between ADAMTS9 and insulin signaling was characterized further in a model where ADAMTS9 was overexpressed in skeletal muscle. This selective over expression resulted in decreased insulin signaling presumably mediated through alterations of the integrin 131 signaling pathway and disruption of the intracellular cytoskeletal organization. Furthermore, this led to impaired mitochondria! function in mouse muscle-an observation found to be of translational character because humans carrying the ADAMTS9 risk allele have decreased expression of mitochondrial markers. Finally, we found that the link between ADAMTS9 overexpression and impaired insulin signaling could be due to accumulation of harmful lipid intermediates. Our findings contribute to the understanding of the molecular mechanisms underlying insulin resistance and type 2 diabetes and point to inhibition of ADAMTS9 as a potential novel mode of treating insulin resistance.

    • Equatorial volcanic eruptions are known to impact the atmospheric circulation on seasonal time scales through a strengthening of the stratospheric zonal winds followed by dynamic ocean-atmosphere coupling. This emerges as the positive phase of the North Atlantic Oscillation in the first 5 years after an eruption. In the North Atlantic, other modes of atmospheric circulation contribute to the climate variability but their response to volcanic eruptions has been less studied. We address this by retrieving the stable water isotopic fingerprint of the four major atmospheric circulation modes over the North Atlantic (Atlantic Ridge, Scandinavian Blocking and the negative and positive phases of the North Atlantic Oscillation (NAO − and NAO+)) by using monthly precipitation data from Global Network of Isotopes in Precipitation (GNIP) and 500 mb geo-potential height from the 20th Century Reanalysis. The simulated stable isotopic pattern of each atmospheric circulation mode is further used to assess the retrieved pattern. We test if changes in the atmospheric circulation as well as moisture source conditions as a result of volcanic eruptions can be identified by analyzing the winter climate response after both equatorial and high-latitude North Hemispheric volcanic eruptions in data, reanalysis and simulations. We report of an NAO + mode in the first two years after equatorial eruptions followed by NAO − in year 3 due to a decrease in the meridional temperature gradient as a result of volcanic surface cooling. This emerges in both GNIP data as well as reanalysis. Although the detected response is stronger after equatorial eruptions compared to high latitude eruptions, our results show that the response after high latitude eruptions tend to emerge as NAO − in year 2 followed by NAO + in year 3–4.

  • Selecting analytical tools for characterization of polymersomes in aqueous solution

    • Selecting the appropriate analytical methods for characterizing the assembly and morphology of polymer-based vesicles, or polymersomes are required to reach their full potential in biotechnology. This work presents and compares 17 different techniques for their ability to adequately report size, lamellarity, elastic properties, bilayer surface charge, thickness and polarity of polybutadiene-polyethylene oxide (PB-PEO) based polymersomes. The techniques used in this study are broadly divided into scattering techniques, visualization methods, physical and electromagnetical manipulation and sorting/purification. Of the analytical methods tested, Cryo-transmission electron microscopy and atomic force microscopy (AFM) turned out to be advantageous for polymersomes with smaller diameter than 200 nm, whereas confocal microscopy is ideal for diameters >400 nm. Polymersomes in the intermediate diameter range can be characterized using freeze fracture Cryo-scanning electron microscopy (FF-Cryo-SEM) and nanoparticle tracking analysis (NTA). Small angle X-ray scattering (SAXS) provides reliable data on bilayer thickness and internal structure, Cryo-TEM on multilamellarity. Taken together, these tools are valuable for characterizing polymersomes per se but the comparative overview is also intended to serve as a starting point for selecting methods for characterizing polymersomes with encapsulated compounds or polymersomes with incorporated biomolecules (e.g. membrane proteins).

    • The atomic simulation environment (ASE) is a software package written in the Python programming language with the aim of setting up, steering, and analyzing atomistic simulations. In ASE, tasks are fully scripted in Python. The powerful syntax of Python combined with the NumPy array library make it possible to perform very complex simulation tasks. For example, a sequence of calculations may be performed with the use of a simple 'for-loop' construction. Calculations of energy, forces, stresses and other quantities are performed through interfaces to many external electronic structure codes or force fields using a uniform interface. On top of this calculator interface, ASE provides modules for performing many standard simulation tasks such as structure optimization, molecular dynamics, handling of constraints and performing nudged elastic band calculations.

    • European competition authorities consider environmental and competition policy to be complementary, as each has the objective of improving social welfare. Naturally, one would imagine that environmental considerations have already been accounted for in competition policy practice, but thus far, only in the case of voluntary environmental agreements. This paper analyzes the extent to which the two types of policy are complementary by examining the legal framework of competition policy, voluntary environmental agreements and three competition cases with equal relevance to the environment and then examines the basic theoretical economic observation that less competition implies lower pollution levels relative to more competition. This trade-off between the benefits of reduced emissions on the one side and increased competition on the other implies that disregarding environmental considerations in the implementation of competition policy entails a negative externality, which can be eliminated if environmental considerations are internalized in competition policy. The impact of the externality is found to depend on the emission level, the emission damage, and market size. Further, this implies that if competition authorities were to adopt a broad definition of the relevant market in competition cases, then environmental considerations would play a limited role in competition policy, whereas a narrow definition implies that environmental considerations should have a much more important place than competitive considerations in competition policy.

    • This handbook provides an overall understanding of the concept of the circular economy and of the societal dynamics through which innovations and transitioning processes towards a circular economy are realised. It describes and discusses the specific economic and political context for applying and developing the circular economy in the CIRTOINNO project. The specific contexts are the tourism sectors in the South Baltic partner regions. It investigates and discusses the opportunities for small and medium-sized tourism businesses to adopt circular economy ideas, and to identify possible ‘good practices’ among tourism SMEs in developing and applying circular economy solutions, especially in relation to the fields of foci of the CIRTOINNO project: accommodation, food and spa services.

    • A novel and generally applicable method for determining structures of membrane proteins in solution via small-angle neutron scattering (SANS) is presented. Common detergents for solubilizing membrane proteins were synthesized in isotope-substituted versions for utilizing the intrinsic neutron scattering length difference between hydrogen and deuterium. Individual hydrogen/deuterium levels of the detergent head and tail groups were achieved such that the formed micelles became effectively invisible in heavy water (D2 O) when investigated by neutrons. This way, only the signal from the membrane protein remained in the SANS data. We demonstrate that the method is not only generally applicable on five very different membrane proteins but also reveals subtle structural details about the sarco/endoplasmatic reticulum Ca2+ ATPase (SERCA). In all, the synthesis of isotope-substituted detergents makes solution structure determination of membrane proteins by SANS and subsequent data analysis available to non-specialists.
