Plasma membrane H+-ATPase and 14-3-3 Isoforms of Arabidopsis leaves: Evidence for isoform specificity in the 14-3-3/H+-ATPase interaction

- The plasma membrane H+-ATPase is activated by binding of 14-3-3 protein to the phosphorylated C terminus. Considering the large number of 14-3-3 and H+-ATPase isoforms in Arabidopsis (13 and 11 expressed genes, respectively), specificity in binding may exist between 14-3-3 and H+-ATPase isoforms. We now show that the H'-ATPase is the main target for 14-3-3 binding at the plasma membrane, and that all twelve 14-3-3 istiforms tested bind to the H+-ATPase in vitro. Using specific antibodies for nine of the 14-3-3 isoforms, we show that GF14epsilon, mu, lambda, omega, chi, phi, nu, and upsilon are present in leaves, but that isolated plasma membranes lack GF14chi, phi and upsilon. Northern blots using isoform-specific probes for all 14-3-3 and H+-ATPase isoforms showed that transcripts were present for most of the isoforms. Based on mRNA levels, GF14epsilon, mu, lambda and chi are highly expressed 14-3-3 isoforms, and AHA1, 3, and 11 highly expressed H+-ATPase isoforms in leaves. However, mass peptide fingerprinting identified AHA1 and 2 with the highest score, and their presence could be confirmed by MS/MS. It may be calculated that under 'unstressed' conditions less than one percent of total 14-3-3 is attached to the H+-ATPase. However, during a condition requiring full activation of H+ pumping, as induced here by the presence of the fungal toxin fusicoccin, several percent of total 14-3-3 may be engaged in activation of the H+-ATPase.

Impairment of intra-oral sensation, discrimination ability, and swallowing function following radiotherapy and surgery for oral and pharyngeal cancer

- Oral and pharyngeal cancer is commonly treated with a combination of radiotherapy and surgery. It is a clinical knowledge that patients often experience severe swallowing disorders following treatment. Since surgical sequelae are instantaneous and obvious, little attention has been paid to other concurrent effects of the treatment. To shed light on this subject, the aim of this thesis was twofold (i) to make a retrospective inventory of the sequelae following treatment and (ii) to perform a prospective, inceptive examination at diagnosis, and to follow-up after radiotherapy, six months and 12 months after surgery. The files of ninety-nine patients revealed that following treatment one-third had to use gastric fistulas and more than nine of ten patients had restricted swallowing capacity. Every second patient could only swallow puréed or liquid food. Adequate intra-oral sensation and discrimination ability is essential for bolus preparation and bolus control, for appropriate elicitation of the swallowing reflex and, hence, for the oral phase of swallowing. At the inceptive examination, the prospective part of the study demonstrated intra-oral discrimination ability in patients was equal to that in healthy controls but was impaired six months after treatment and there was no significant improvement after 12 months. It had been expected that the patient’s healthy, non-tumor side would compensate but it did not. An explanation was found when it was revealed that radiotherapy induced a delayed decline in intra-oral sensation. Sensory decline was not demonstrated within a month after radiotherapy but was manifest six months later. Since the radiotherapy field includes the neck, because of the risk for metastasis, it is highly plausible that pharyngeal sensation declines in a manner corresponding to that found intra-orally when the healthy side is irradiated. In accord with this presumption, pharyngeal swallowing function deteriorated in patents with oral tumors. Cineradiographic evaluation of oral and pharyngeal swallowing function disclosed a significant association between the degree of swallowing dysfunction and the degree of sensory decline and with the degree of impairment of shape recognition. Conclusions: Delayed intra-oral sensory decline, found to be induced by radiotherapy, can be expected to appear in the entire radiation field, including the oral cavity and the pharynx, with adverse effect on swallowing. Testing intra-oral sensation close to the last radiotherapy session is not advisable, because sensory decline does not develop immediately after radiotherapy but manifests after six months. Spontaneous sensory rehabilitation cannot be expected after six months. The significant association between degree of swallowing dysfunction and degree of intra-oral sensory decline and impaired discrimination ability must be considered in the quest for functional rehabilitation of patients treated for oral or pharyngeal cancer.

- Results from research on deaf children, who have acquired a sign language late in life, have revealed difficulties in the development of theory of mind, highlighting the role of communication in this development. From this perspective, the development of theory of mind in children with severe speech and physical impairments (SSPI) is most interesting. Findings from our research suggest that the children follow a normal pattern of development but with severe delay compared to children without disability. These results will be discussed in terms of early social and communicative experience as well as in terms of verbal ability and memory capacity.

- Modelling and simulation of the dynamic behaviour of catenary-pantograph interaction is an important part when assessing the capability of a current collection system for railway traffic. The large variation in infrastructure characteristics in different countries and railway companies, different types of traffic, designs of pantographs etc. makes it almost impossible to develop a final simulation model of such a system. Instead, it would be favourable to have a tool that has the ability to set up models of such systems, choose relevant detail of the models, run simulations and finally visualize the results. To make the tool useful for engineers, design experts as well as simulation experts, the functionality of the tool must be worked out. Aspects on computer simulation such as developed models, simulation methods and computer tools are presented. The aim is to develop a scenario that considers different designs, models, solution methods and user levels. The scenario focuses on how to structure the use of simulation of dynamics in catenary-pantograph development. A scenario is sketched built on different user levels, a modular structure and a structure for information management

Luftföroreningar i skogliga provytor - Resultat till och med september 2002

- På uppdrag av luftvårdsförbund, länsstyrelser, kommuner och skogsvårdsstyrelser mäter IVL nedfall av luftföroreningar och markvattnets sammansättning på 109 lokaler i hela Sverige och lufthalter på vissa av dessa. Metoder har varit desamma under hela mätperioden, vilket gör data från slutet av 1980-talet jämförbara med senaste årets data. Syftet med mätningarna är att beskriva nedfallets storlek och markvattnets sammansättning i skogsytorna, men även visa skillnader mellan olika delar av landet och hur förhållandena ändras med tiden. Flertalet provytor ligger i Skogsvårdsorganisationens observationsytor, vilket gör att data avseende luftföroreningar kan jämföras med skogliga uppgifter. Undersökningarna sedan slutet av 1980-talet har visat att nedfallet av svavel och kväve är störst i sydvästra Sverige och avtar åt nordost. Längre norrut finns en gradient med större deposition i Stockholmsområdet och längs Norrlandskusten än inåt i landet. Nedfallet av svavel har minskat, liksom skillnaderna mellan olika regioner i Sverige, och nederbörden har blivit mindre sur. Till stor del förklaras det av minskade utsläpp av svavel i Europa. För kväve är det svårt att se trender. Det är viktigt att internationellt avtalade utsläppsminskningar genomförs för att nå förväntad belastning av försurande och gödande ämnen år 2010. Trots markant minskad försurningsbelastning är det svårt att se någon tydlig minskning av markvattnets surhetsgrad. Främsta orsaken är sannolikt att återhämtningen i skogsmark är långsam och att kritiska belastningsgränser fortfarande överskrids i vissa områden, speciellt i södra Sverige. Störst nedfall av antropogent svavel noterades i fyra granytor i Skåne och Halland, 8-8,7 kg per hektar under hydrologiska året 2001/02. Som genomsnitt för 27 lokaler med varierande trädslag i hela Götaland noterades 3,8 kg svavel per hektar under samma period. Motsvarande från elva lokaler i Svealand var 2,0 kg/ha och 1,1 kg/ha från sju lokaler i de fyra norrlandslänen. När det gäller kväve fanns toppnoteringarna i Halland, 16,9 kg kväve per hektar via nederbörden (varav 14,4 kg oorganiskt och 2,5 kg organiskt kväve). Motsvarande för de två nordliga lokaler var 2 kg per hektar. Speciellt i Hallands län har en risk för utlakning av kväve från områden med växande skog noterats. Lufthalter av svavel och kväve i bakgrundsmiljö visar en tydlig gradient över Sverige. Under 2001/02 noterades tre gånger högre halter av svaveldioxid på lokalerna i södra Sverige (genomsnitt 1 mg/m3) jämfört med i norra Sverige (genomsnitt 0,3 mg/m3). För kvävedioxid var gradienten ännu tydligare med tio gånger högre halter i söder jämfört med i norr; 5 mg/m3 i södra Sverige och 0,5 mg/m3 i norra Sverige. Bakgrundsvärden av svavel- och kvävedioxid är långt under gällande gränsvärden. Däremot indikerar halterna av marknära ozon risk för minskad tillväxt inom både jord- och skogsbruk

- Two Norway spruce (Picea abies (L.) Karst.) genes belonging to class I of the KNOTTED1-like homeobox (KNOX) genes, HBK2 and HBK3, were cloned with PCR-based methods. The expression of these and a previously characterised related gene, HBK1, in different organs and during somatic embryogenesis was studied with RTPCR. Transcripts of all three genes were detected in stems, roots and in cone buds, but not in needles. HBK1 and HBK3 are expressed throughout development in a normal cell line with embryogenic potential and in a cell line unable to form somatic embryos. HBK2 is expressed in the normal cell line, but not in the developmentally arrested cell line. This suggests that the HBK2 gene is involved in the somatic embryo development.

- Föreliggande handbok är framtagen i ett SBUF-projekt med Kjell Wallin, Peab som projektledare. Projektet har genomförts i nära samarbete mellan Peab Sverige AB och Luleå tekniska universitet. Arbetet har inneburit att ett stort antal datorberäkningar har genomförts, och de redovisas i diagramform i denna handbok. Resultaten avses att användas att fylla i en blankett i ett Excel-ark. Syftet är att för typfallet vägg-på-platta ska handboken ge underlag för sprickriskbedömning och i förekommande fall ange vilka typåtgärder som ska sättas in. Avsikten är därvid att man inte ska behöva göra några externa beräkningar, men användaren har full frihet att själv ta fram temperaturen eller tvångssituationen på godtyckligt annat sätt. Handboksmetodiken avser en fortsättning på det som i Bronormen (Bro 94, bilaga 9-5) kallas metod 2, och den redovisade kompletteringen innefattar främst att erforderliga åtgärder (kylning och/eller värmning) för det valda typfallet inkluderas.
