    • Bovine muscle carbonic anhydrase III was purified to homogeneity by the strategy of rapid ion-exchange chromatography. The ionic exchanger used was CM-cellulose, and this is the first application of this technique on a cation exchanger. Nitrogen gas was used to pressurize the chromatographic column to accelerate the elution. The results show that proteins with high isoelectric points can also be purified in this way. The procedure is very time-saving compared with conventional chromatography, reducing the elution time five-to ten-fold. The proteins are in addition protected against oxidation by air.

  • Vertical field MR imaging of upper thorax and spine in small children. Evaluation of a new surface coil

    • To improve image quality in a vertical field MR imaging unit, operating at low field strength (0.3 T), we have designed a half-elliptical coil for use in the upper thoracic region of small children. Our intention was also to shorten the examination time, which until now has been long, because several scans with different coils have been necessary to cover the thoracic region. The experimental coil is designed so that a child's shoulders fit into the central region. The coil consists of 2 serially connected cable-loops, mounted on a foam rubber vest. The coil performance was tested in a phantom and improvements relative to standard coils were demonstrated in in vivo studies. The results indicate that by using the half-elliptical coil, the signal-to-noise (S/N) ratio can be improved by a factor of 2 to 3 in the thoracic region of a child.

  • Aortic pathology revealed by MRI in patients with clinical suspicion of spinal disease

    • In five patients with clinical suspicion of spinal disease, MRI of the spine revealed unexpected aortic pathology explaining the symptoms. No significant intraspinal pathology was found on MRI. However, in one patient with clinical suspicion of spinal stenosis, an aortic occlusion was detected on MR images of the spine. The lower extremity ischaemia, caused by the occlusion, was responsible for the symptoms. In another patient a paravertebral haematoma from a ruptured aortic aneurysm resulted in spinal nerve compression, thought before MRI to be caused by a spinal tumour. In three patients aortic aneurysm or dissection resulted in spinal cord ischaemia with symptoms mimicking those of compressive spinal disease. Thus, if MRI of the spine does not provide an explanation for the patient's symptoms, examination of the aorta is recommended.

  • Bioprocessing of Oats Influence on Phytate Hydrolysis and Mineral Bioavailability

    • The effects of bioprocessing on enzymatic phytate hydrolysis in oats were studied during malting, soaking and breadmaking to elucidate the reasons for poor phytase activity in oats and determine the optimal conditions for phytate hydrolysis. Malting of oats in a pilot plant was studied to investigate whether the malting process developed in the laboratory could be used in large scale preparation of oats with reduced phytate content. The effects of malting on zinc and iron absorption from single meals were examined in humans using a radioisotope technique. The nutritional significance of cereal processing and high dietary phytate intake was also investigated. Optimal conditions for oat phytase was found at pH 4 - 4.5 and 38°C, which differs from those of wheat, rye and barley. Combined malting and soaking reduced the phytate content of oats to very low levels (99% reduction). It was demonstrated that the malting process developed in the laboratory was also applicable on a large scale. In the pilot plant, malting of oats was performed at 15°C for 5 days, which were the conditions found to be optimal for achieving satisfactory phytate reduction. The intracellular location of oat storage proteins is different from that of other cereals, and interactions between phytate and proteins may partly explain the resistance of oat phytate to phytase attack. It is possible that the increased phytate hydrolysis following germination is the result of structural and chemical changes occurring in the oat seed during steeping and germination. Fluorescence microscopy was used to study the microstructure of malted and ungerminated oats. The micrographs indicated differences in phytate location and structure of malted oats when compared with ungerminated oats. The addition of 10% sour dough during rye breadmaking increased the phytate hydrolysis to 97% at pH 4.6 in doughs and bread. The combination of scalding and adding 20 and 30% rye sour dough to breads in which 30% of the wheat flour had been exchanged for oat flour resulted in a phytate reduction of 95-96% at pH values between 4.3 and 4.6 in doughs and bread. The absorption of zinc from oat porridge prepared using malted and soaked oats (phytate content reduced by 77%) doubled the amount of zinc absorbed over the absorption from oat porridge made with untreated oats. The iron absorption was 47% higher from malted oat porridge than from the control porridge. The iron stores in a group of pregnant Pakistani women were significantly lower than in a group of pregnant Norwegian women. The dietary intake of heme and nonheme iron as well as factors known to inhibit iron absorption, except for phytate, did not differ between the groups. The dietary phytate intake was considerably higher in the Pakistani group, owing to the consumption of unleavened Chapati bread, thus suggesting the importance of using efficient food processes to improve the iron bioavailability in cereals.

    • Several lines of evidence indicates that a population of lymphoid cells occurring in most organs of the body with a dendritic morphology is of prime importance in the initial stages of immune responses (1). Immunohistochemical distribution studies using various cell surfaces phenotypic markers has often been used to argue the probable importance of dendritic cells in tissues for the initial immune response (2,3). It has however proven difficult to compare these cells from all tissues functionally in vitro due to low yields and alteration of the phenotype when the cells are enzymatically dislodge from the tissues. Most studies have thus focused on the Langerhans cell of the skin, spleen dendritic cells and negatively selected dendritic cells from human peripheral blood.

  • MRI of extradural spinal tumours at 0.3 T

    • Ninety-one patients with extradural spinal tumours were examined by magnetic resonance imaging. There were 76 metastases (6 from unknown primary tumours). Seven patients had primary spinal tumours and 8 had multiple myeloma. Sixteen had bulging, diseased vertebral bodies compressing the subarachnoid space and 67 had extradural tumour compressing the spinal cord. Sixty patients had paravertebral involvement. Intraspinal involvement did not correlate with the extent of spinal lesions. All patients had vertebral destruction, with hypointense or combined hypo- and isointense signal relative to bone marrow on T1-weighted images. In most of the 22 patients with T2-weighted images the tumours were isointense or slightly hyperintense. It was usually impossible to differentiate the various tumours on the basis of signal intensity and morphology. However, metastases from carcinoma of the prostate were often more hypointense than other tumours on T1- and T2-weighted images. An inhomogeneous pattern in which diffusely low signal is combined with focal lower signal on T1-weighted images may suggest myeloma. In the 22 patients examined with both T1- and T2-weighted images, T1-weighted images gave the best information in 18; in 3 they were equivalent and in 1 inferior to T2-weighted images; they are therefore recommended for routine imaging of epidural spinal tumours.
