  • Predicting target profiles with confidence as a service using docking scores

    • Background: Identifying and assessing ligand-target binding is a core component in early drug discovery as one or more unwanted interactions may be associated with safety issues. Contributions: We present an open-source, extendable web service for predicting target profiles with confidence using machine learning for a panel of 7 targets, where models are trained on molecular docking scores from a large virtual library. The method uses conformal prediction to produce valid measures of prediction efficiency for a particular confidence level. The service also offers the possibility to dock chemical structures to the panel of targets with QuickVina on individual compound basis. Results: The docking procedure and resulting models were validated by docking well-known inhibitors for each of the 7 targets using QuickVina. The model predictions showed comparable performance to molecular docking scores against an external validation set. The implementation as publicly available microservices on Kubernetes ensures resilience, scalability, and extensibility.

  • Stormwater Uptake in Sponge-Like Porous Bodies Surrounded by a Pond: A Fluid Mechanics Analysis

    • In this work, a previously published model for the water up take of stormwater in sponge-like porous bodies by the group is further developed. This is done by investigating the highest-performing model and considering the water uptake from the surroundings of a pond and rain-infiltrated soil. This implies that water uptake from impermeable to partially permeable surfaces is examined. Hence, the following cases are considered: (1) impervious bottom surface and no precipitation, (2) impervious bottom surface with precipitation, (3) permeable soil with no precipitation, and (4) permeable soil with precipitation. A mathematical model covering all these cases is presented, where the governing equations are the mass conservation and Darcy’s law together with an assumption of a sharp wetting front being a first-order approximation of the complete Richard’s equation. Results for the water uptake height, pond depth, and wetting front are computed numerically and plotted against time. Analytical solutions are also presented in certain cases, and critical values are obtained. The parametric study includes variations in the ratio of the model- to the surrounding ground surface area, initial pond depth, precipitation, and soil characteristics. To exemplify, the time it takes to absorb the water from the pond after a precipitation period is presented. The results are related to the Swedish rainfall data of 1 h duration with a return period of 10 years. When evaluating efficiency, the focus is on the absorption time. Results vary considerably, demonstrating a general trend that with soil infiltration, the water absorption rate is higher. For most cases, the considered water amount is absorbed completely, although depending on the parameters and conditions. These results serve to optimize the model for each of the cases. The main focus of the research lies in the theoretical aspect.

    • Ethnographic approaches developed gradually during the 1980ies and 1990ies from several sources. The general emergence of a strong turn towards qualitative research had opened this opportunity  and the establishment of ethnography has been described in a text from the early 2000ies as stepwise, ending with a number of pre-conferences to NERA’s yearly congress, starting 1998 (Larsson, 2006). The symposium has been developed looking forward from this history to the future of ethnography of education in the Nordic region. It has been developed through cooperation across two networks: “Gender and Education” and “Justice through Education”.  Its focus is on ethnographic methodologies and their development in education research in the Nordic countries in recent decades, with a particular focus on matters of research for justice through education and education equality.

  • On Developments in Ethnographic Research: The Case of Two Swedish Universities

    • The past 40 years have formed a transitional period in Sweden’s education and political history. The social democratic reforms from the 1940s that aimed to create a centralised, expanded and integrated comprehensive education system came to an end. Decentralisation, neoliberal governance and the introduction of new public management with the creation of private schools and competition have shaped the policy regime since then. Ethnography emerged in Swedish educational research as a significant research methodology during this transitional period. Using a qualitative and quantitative investigation of research dissertations that classified and counted the use of ethnography as either classical (using core references and long-term participation research at one or a limited number of sites), or adapted (used within adaptations to other research methods), the present article explores these developments at two universities. It suggests that Swedish education ethnography has developed along similar kinds of historical trajectories to ethnography in other places, with roots similar to those in other European countries, though also with some variations. For instance, as elsewhere, ethnography needed a breakthrough point in Swedish education research. It got this in the 1980s. However, it quickly became an important part of educational research from the 1990s onwards and a strong quantitative take off early in the new millennium followed. Presently more than half of all PhD dissertations in Education at the two universities have some kind of participant observation, over half of which are also classically ethnographic. This leads us to conclude that education ethnography in Sweden has changed across its period of growth and that though configured in contemporary social science as having originated in anthropology as a methodology that employed long-term embedded participant observation, this does not limit the variations of ethnography’s development or its application.

    • In this symposium we will attempt to paint an updated broad picture of ethnography in education research and its development, covering all Nordic countries, by looking initially at the general past growth of ethnography of education there, along with discussions of present developments and possible future ones too. This will also involve presentations that have been invited to think forward in relation to ethnography of education in the region, whilst also looking back at ethnographic practices in education and their social relations and material histories (Beach, Bagley and Marques da Silva). After this, research addressing respectively and conjointly Gender Equity and Justice in Education will be given particular attention.

  • Evaluation in pig of an intestinal administration device for oral peptide delivery

    • The bioavailability of peptides co-delivered with permeation enhancers following oral administration remains low and highly variable. Two factors that may contribute to this are the dilution of the permeation enhancer in the intestinal fluid, as well as spreading of the released permeation enhancer and peptide in the lumen by intestinal motility. In this work we evaluated an Intestinal Administration Device (IAD) designed to reduce the luminal dilution of drug and permeation enhancer, and to minimize movement of the dosage form in the intestinal lumen. To achieve this, the IAD utilizes an expanding design that holds immediate release mini tablets and places these in contact with the intestinal epithelium, where unidirectional drug release can occur. The expanding conformation limits movement of the IAD in the intestinal tract, thereby enabling drug release at a single focal point in the intestine. A pig model was selected to study the ability of the IAD to promote intestinal absorption of the peptide MEDI7219 formulated together with the permeation enhancer sodium caprate. We compared the IAD to intestinally administered enteric coated capsules and an intestinally administered solution. The IAD restricted movement of the immediate release tablets in the small intestine and histological evaluation of the mucosa indicated that high concentrations of sodium caprate were achieved. Despite significant effect of the permeation enhancer on the integrity of the intestinal epithelium, the bioavailability of MEDI7219 was of the same order of magnitude as that achieved with the solution and enteric coated capsule formulations (2.5–3.8%). The variability in plasma concentrations of MEDI7219 were however lower when delivered using the IAD as compared to the solution and enteric coated capsule formulations. This suggests that dosage forms that can limit intestinal dilution and control the position of drug release can be a way to reduce the absorptive variability of peptides delivered with permeation enhancers but do not offer significant benefits in terms of increasing bioavailability.

    • We carry out a small-scale empirical study of a dialogue strategy (conversational pattern) found in second language learner dialogues where a language-assisting teacher is present, allowing learners to pick up new words and train on them while maintaining a conversation. We also provide a formal model of the observed conversational pattern including several frequently occurring variants, as well as a demonstration implementation which is able to reproduce the most common variant of the pattern.
