  • Safety and immunogenicity of a malaria vaccine, Plasmodium falciparum AMA-1/MSP-1 chimeric protein formulated in montanide ISA 720 in healthy adults

    • BACKGROUND: The P. falciparum chimeric protein 2.9 (PfCP-2.9) consisting of the sequences of MSP1-19 and AMA-1 (III) is a malaria vaccine candidate that was found to induce inhibitory antibodies in rabbits and monkeys. This was a phase I randomized, single-blind, placebo-controlled, dose-escalation study to evaluate the safety and immunogenicity of the PfCP-2.9 formulated with a novel adjuvant Montanide ISA720. Fifty-two subjects were randomly assigned to 4 dose groups of 10 participants, each receiving the test vaccine of 20, 50, 100, or 200 microg respectively, and 1 placebo group of 12 participants receiving the adjuvant only.METHODS AND FINDINGS: The vaccine formulation was shown to be safe and well-tolerated, and none of the participants withdrew. The total incidence of local adverse events (AEs) was 75%, distributed among 58% of the placebo group and 80% of those vaccinated. Among the vaccinated, 65% had events that were mild and 15% experienced moderate AEs. Almost all systemic adverse reactions observed in this study were graded as mild and required no therapy. The participants receiving the test vaccine developed detectable antibody responses which were boosted by the repeated vaccinations. Sixty percent of the vaccinated participants had high ELISA titers (>1:10,000) of antigen-specific antibodies which could also recognize native parasite proteins in an immunofluorescence assay (IFA).CONCLUSION: This study is the first clinical trial for this candidate and builds on previous investigations supporting PfCP-2.9/ISA720 as a promising blood-stage malaria vaccine. Results demonstrate safety, tolerability (particularly at the lower doses tested) and immunogenicity of the formulation. Further clinical development is ongoing to explore optimizing the dose and schedule of the formulation to decrease reactogenicity without compromising immunogenicity.TRIAL REGISTRATION: Chinese State Food and Drug Administration (SFDA) 2002SL0046; Controlled-Trials.com ISRCTN66850051 [66850051].

  • Diffusion layer growth at Zn/Cu interface under uniform and gradient high magnetic fields

    • As a common phenomenon occurring in many material processes, diffusion may induce significant changes in composition and microstructure near the interface. In the present study, liquid/solid (Zn/Cu) interface diffusion experiments in high magnetic fields (up to 12 T) were conducted and the thickness changes of diffusion layer under different magnetic field conditions were examined. It was found that there were no noticeable effects of high magnetic fields on the formation of intermetallic phases at the interface. However, the magnetic flux density exerted a non-linear influence on the diffusion layer thickness. This phenomenon should be attributed to the effect of magnetic fields suppressing natural convection and inducing thermo-electromagnetic convection. In addition, the diffusion of Zn into Cu could be retarded by a magnetic field gradient. These results indicate that both the strength and the gradient of high magnetic fields can be used to control the diffusion behavior.

  • Concentric silicon micro-ring resonators with enhanced transmission notch depth

    • In this work, we have analyzed, fabricated and demonstrated concentric micro-ring resonators in silicon-on-insulator (SOI) structure for enhanced transmission notches. The operation principles of the concentric ring resonators are studied by time-domain coupled-mode theory. Directional coupling between concentric rings offers another freedom in designing deep notch optical filters and ultra-sensitive biosensors. The finite-difference-time-domain (FDTD) simulations have shown the improvement of the notch depth, evenly distributed mode field and the effect of the resonance shift. The device is demonstrated in silicon-on-insulator structure. Transmission notch depth improvement of ~ 15dB is demonstrated for the 21-20.02-μm-radius double-ring structure comparing with the single 21-μm-radius ring.

    • Paper substrate is one of the paramount nominees for Radio Frequency Identification (RFID) tags but at the same time it is extremely prone towards environmental changes. In this paper, antennas for UHF RFID tags on paper based substrate are investigated and analyzed for the first time to evaluate the effect of change in dielectric constant on the antenna parameters and performance. On the basis of analysis a concrete meander line antenna is proposed, designed and evaluated which has tremendous immunity towards variation in dielectric constant.

This paper presents a novel inkjet printed rounded corner bowtie antenna with T-matching stubs on paper substrate which is the cheapest and widest available substrate. The antenna exhibits compact size with outstanding read range.

    • We propose and demonstrate a tunable broadband photonic radio frequency (RF) phase shifter based on a silicon microring resonator. This scheme utilizes the thermal nonlinear effect of the silicon microring to change the electrical phase of the RF signal with a wide tuning range. A prototype of the phase shifter is experimentally demonstrated for a 40-GHz signal with a 0-4.6-rad tuning range.

    • Printed interdigital capacitor DWI on paperboard is a promising solution for low-cost sensors in intelligent packaging applications. The currently available analytical models of multi-layered IDCs are targeted to those fabricated by conventional semiconductor process. For this reason, we have adapted two promising models and assessed their accuracies by comparison with experimental data. We modified these models by treating the paper as non-infinite thick substrate and taking the effect of printed metal thickness into account. The models are studied further to reveal the relationship between the response of capacitance change and various geometric parameters which enables a quick way of obtaining the optimum IDC structure design. The modified Gevorgian model fits our experimental data best, and the sensitivity of IDCs is largely affected by its spatial wavelength and the thickness of sensing material layer, while the finger number, length and metallization ratio have minor impact.

    • This study aimed to determine the effects of advanced glycation end products (AGEs) on endothelial cytoskeleton morphology and permeability, and to detect the underlying signaling mechanisms involved in these responses. Cultured endothelial cells (ECs) were exposed to AGE-modified human serum albumin (AGE-HSA), and EC cytoskeletal changes were evaluated by observing fluorescence of F-actin following ligation with labeled antibodies. Endothelial permeability was detected by measuring the flux of TRITC-albumin across the EC monolayers. To explore the signaling pathways behind AGE-induced EC alteration, ECs were treated with either soluble anti-AGE receptor (RAGE) IgG, or the MAPK inhibitors PD98059 and SB203580 before AGE-HSA administration. To further elucidate possible involvement of the ERK and p38 pathways in AGE-induced EC changes, adenovirus-carried recombinant constitutive dominant-negative forms of upstream ERK and p38 kinases, namely MEK1(A) and MKK6b(A), were pre-infected into ECs 24 h prior to AGE-HSA exposure. AGE-HSA induced actin cytoskeleton rearrangement, as well as EC hyperpermeability, in a dose and time-dependent manner. The effects were attenuated in cells pretreated with anti-RAGE IgG, PD98059 or SB203580, respectively. EC pre-infection with MEK1(A) and MKK6b(A) also alleviated the effect of AGEs. Furthermore, adenovirus-mediated administration of activated forms of either MEK1 or MKK6b alone induced rearrangement of F-actin and hyperpermeability. The results indicate that ERK and p38 MAPK play important roles in the mediation of AGE-induced EC barrier dysfunction associated with morphological changes of the F-actin.
